    The Structure of Tumours Derived from Mouse Cells after “Spontaneous” Transformation in vitro

    The histological structure of 58 primary tumours derived from spontaneously transformed tissue culture cell lines from embryonic (14-16 days), young (3-20 days) and old (28-34 months) C3H and C57 mice is described. Although the cell lines were derived from a number of different organs the tumours were similar in morphology. The pattern was mixed, with “fibrosarcomatous”, “myxoid”, “epithelioid” and giant cell areas. The tumours resemble some types of haemangiopericytoma

    An Ultrastructural Analysis of the Physical Organization of Collagenous (Type I) Matrices: One Determinant of Urothelium Maintenance In Vitro

    Collagenous matrices, used as cell culture substrata, can be prepared from different collagen types in a variety of forms using a range of polymerization procedures. Type I collagen has been most frequently used either as dried collagen films or hydrated collagen gels. Sheets of isolated bladder urothelium, when plated onto such matrices prepared from type I collagen by different polymerization methods (eg. air-drying; NaOH; NaCl; NH3; or NH3 followed by glutaraldehyde crosslinking) demonstrate the capability of urothelial cells to attach to a variety of differently prepared matrices irrespective of polymerization procedure. In contrast, both cell proliferation and maintenance of the urothelium are markedly influenced by the polymerized form of the collagen matrix. Comparative ultrastructural (scanning and transmission electron microscopy) analysis of these matrices demonstrates dissimilarities in their physical organization. The level of filamentous, fibrillar or fibrous reaggregation of solubilized collagen molecules varies in relation to the polymerization procedure used viz, a) air dried matrices form a dense meshwork of many forms of collagen fibrils and associated filaments with an irregular surface array of coarser collagen fibres; b) matrices prepared by NaOH, NaCl and NH3 polymerization present no major differences and form a felt of interlocking collagen fibres with discrete filamentous networks associated with these fibres; and c) matrices polymerized by NH3 and crosslinked with glutaraldehyde form a dense meshwork of filaments with a more occasional distribution of fibrils associated with filaments or dense amorphous aggregates. The level of supramolecular reassemblage of solubilized collagen may be, therefore, a significant factor in determining urothelial cell growth and differentiation on collagen matrices

    The effect of carbohydrate mouth rinsing on fencing performance and cognitive function following fatigue-inducing fencing

    This study investigated the impact that mouth rinsing carbohydrate solution has on skill-specific performance and reaction time following a fatigue inducing bout of fencing in epee fencers. Nine healthy, national level epee fencers visited a laboratory on 2 occasions, separated by a minimum of 5 days, to complete a 1-minute lunge test and Stroop test pre and post fatigue. Heart rate and ratings of perceived exertion (RPE) were recorded during completion of the fatiguing protocol. Between fights the participants mouth rinsed for 10 seconds, either 25ml of 6.7% maltodextrin solution (MALT) or water (PLAC). Blood lactate and glucose were recorded at baseline, pre- and post-testing. Results showed an increase in heart rate and overall RPE over time in both conditions. There were no differences in blood glucose (F(1,8)=.63, P=.4, ηp=.07) or blood lactate levels (F(1,8)=.12, P=.70, ηp=.01) between conditions as a function of time. There was a significant improvement in lunge test accuracy during the MALT trial (F(1,8)=5.21, P=.05, ηp=.40) with an increase from pre (81.2 ±8.3%) to post (87.6 ±9.4%), whereas there was no significant change during the placebo (pre 82.1 ±8.8%, post 78.8 ±6.4%). There were no recorded differences between conditions in response time to congruent (F(1,8)=.33, P=.58, ηp=.04) or incongruent stimuli (F(1,8)=.19, P=.68, ηp=.02). The study indicates that when fatigued mouth rinsing MALT significantly improves accuracy of skill-specific fencing performance but no corresponding influence on reaction time was observed

    An adaptive moving mesh method for forced curve shortening flow

    We propose a novel adaptive moving mesh method for the numerical solution of a forced curve shortening geometric evolution equation. Control of the mesh quality is obtained using a tangential mesh velocity derived from a mesh equidistribution principle, where a positive adaptivity measure or monitor function is approximately equidistributed along the evolving curve. Central finite differences are used to discretise in space the governing evolution equation for the position vector and a second-order implicit scheme is used for the temporal integration. Simulations are presented indicating the generation of meshes which resolve areas of high curvature and are of second-order accuracy. Furthermore, the new method delivers improved solution accuracy compared to the use of uniform arc-length meshes
