    Effects of Communication Protocol Stack Offload on Parallel Performance in Clusters

    The primary research objective of this dissertation is to demonstrate that the effects of communication protocol stack offload (CPSO) on application execution time can be attributed to the following two complementary sources. First, the application-specific computation may be executed concurrently with the asynchronous communication performed by the communication protocol stack offload engine. Second, the protocol stack processing can be accelerated or decelerated by the offload engine. These two types of performance effects can be quantified with the use of the degree of overlapping Do and degree of acceleration Daccs. The composite communication speedup metrics S_comm(Do, Daccs) can be used in order to quantify the combined effects of the protocol stack offload. This dissertation thesis is validated empirically. The degree of overlapping Do, the degree of acceleration Daccs, and the communication speedup Scomm characteristic of the system configurations under test are derived in the course of experiments performed for the system configurations of interest. It is shown that the proposed metrics adequately describe the effects of the protocol stack offload on the application execution time. Additionally, a set of analytical models of the networking subsystem of a PC-based cluster node is developed. As a result of the modeling, the metrics Do, Daccs, and Scomm are obtained. The models are evaluated as to their complexity and precision by comparing the modeling results with the measured values of Do, Daccs, and Scomm. The primary contributions of this dissertation research are as follows. First, the metric Daccs and Scomm are introduced in order to complement the Do metric in its use for evaluation of the effects of optimizations in the networking subsystem on parallel performance in clusters. The metrics are shown to adequately describe CPSO performance effects. Second, a method for assessing performance effects of CPSO scenarios on application performance is developed and presented. Third, a set of analytical models of cluster node networking subsystems with CPSO capability is developed and characterised as to their complexity and precision of the prediction of the Do and Daccs metrics

    Efficacy and safety of left atrial appendage closure with WATCHMAN in patients with or without contraindication to oral anticoagulation: 1-Year follow-up outcome data of the EWOLUTION trial

    Background Left atrial appendage (LAA) occlusion with WATCHMAN has emerged as viable alternative to vitamin K antagonists in randomized controlled trials. Objective EWOLUTION was designed to provide data in routine practice from a prospective multicenter registry. Methods A total of 1025 patients scheduled for a WATCHMAN implant were prospectively and sequentially enrolled at 47 centers. Indication for LAA closure was based on European Society of Cardiology guidelines. Follow-up and transesophageal echocardiography (TEE) were performed per local practice. Results The baseline CHA 2 DS 2 -VASc score was 4.5 ± 1.6; the mean age was 73.4 ± 9 years; previous transient ischemic attack/ischemic stroke was present in 312 (30.5%), 155 (15.1%) had previous hemorrhagic stroke, and 320 (31.3%) had a history of major bleeding; and 750 (73%) were deemed unsuitable for oral anticoagulation therapy. WATCHMAN implant succeeded in 1005 (98.5%) of patients, without leaks >5 mm in 1002 (99.7%) with at least 1 TEE follow-up in 875 patients (87%). Antiplatelet therapy was used in 784 (83%), while vitamin K antagonists were used in only 75 (8%). At 1 year, mortality was 98 (9.8%), reflecting the advanced age and comorbidities in this population. Device thrombus was observed in 28 patients at routine TEE (3.7%) and was not correlated with the drug regimen ( P = .14). Ischemic stroke rate was 1.1% (relative risk 84% vs estimated historical data); the major bleeding rate was 2.6% and was predominantly (2.3%) nonprocedure/device related. Conclusion LAA closure with the WATCHMAN device has a high implant and sealing success. This method of stroke risk reduction appears to be safe and effective with an ischemic stroke rate as low as 1.1%, even though 73% of patients had a contraindication to and were not using oral anticoagulation

    Представлен прототип компактного источника электронного пучка для установок электронно-лучевой сварки с расположением электронной пушки и источника высоковольтного напряжения в едином моноблоке. Размещение электронной пушки, источника высоковольтного напряжения, электроники управления пучком и питания накала катода источника электронного пучка для электронно-лучевой сварки в едином корпусе-моноблоке снижает вес и стоимость (за счёт уменьшения количества используемых материалов), объём и занимаемые производственные площади. Это существенно расширяет возможности применения представляемого типа источников электронного пучка в разнообразных областях деятельности человека, в том числе в космических технологиях в открытом пространстве космоса. Цель работы – показать целесообразность концепции компоновки источника электронного пучка в едином корпусе-моноблоке на примере стендовых испытаний прототипа источника-моноблока. Спроектирован и изготовлен прототип источника-моноблока. Проведены его предварительные стендовые испытания с лазерным подогревом катода. Обсуждаются возможные применения. Получен электронный ток источника до 70 мА с энергией 90 кэВ. Данный результат демонстрирует возможность практической реализации нового способа компоновки источника электронного пучкаA prototype of an electron beam compact source for electron-beam welding is presented. The electron gun and a high-voltage source are united in a single monoblock. The placement of the electron gun, the high-voltage source, the beam control electronics and the power supply of the cathode heating of the electron beam source for electron beam welding in a single monoblock housing reduces weight and cost by reducing the amount of materials used, volume and occupied production areas. This significantly expands the possibilities of using the presented type of electron beam sources in various fields of human activity, including space technologies in the open space of space. The purpose of the work is to show the expediency of the concept of arranging the electron beam source in the single monoblock housing as the example of bench tests of the source prototype. The prototype of the monoblock was designed and manufactured. Its preliminary bench tests with laser cathode heating were carried out. Its possible applications are discussed. An electron source current up to 70 mA with an energy of 90 keV was obtained.     The Somatic Genomic Landscape of Glioblastoma

    We describe the landscape of somatic genomic alterations based on multi-dimensional and comprehensive characterization of more than 500 glioblastoma tumors (GBMs). We identify several novel mutated genes as well as complex rearrangements of signature receptors including EGFR and PDGFRA. TERT promoter mutations are shown to correlate with elevated mRNA expression, supporting a role in telomerase reactivation. Correlative analyses confirm that the survival advantage of the proneural subtype is conferred by the G-CIMP phenotype, and MGMT DNA methylation may be a predictive biomarker for treatment response only in classical subtype GBM. Integrative analysis of genomic and proteomic profiles challenges the notion of therapeutic inhibition of a pathway as an alternative to inhibition of the target itself. These data will facilitate the discovery of therapeutic and diagnostic target candidates, the validation of research and clinical observations and the generation of unanticipated hypotheses that can advance our molecular understanding of this lethal cancer

    Multiplatform Analysis of 12 Cancer Types Reveals Molecular Classification within and across Tissues of Origin

    Recent genomic analyses of pathologically-defined tumor types identify “within-a-tissue” disease subtypes. However, the extent to which genomic signatures are shared across tissues is still unclear. We performed an integrative analysis using five genome-wide platforms and one proteomic platform on 3,527 specimens from 12 cancer types, revealing a unified classification into 11 major subtypes. Five subtypes were nearly identical to their tissue-of-origin counterparts, but several distinct cancer types were found to converge into common subtypes. Lung squamous, head & neck, and a subset of bladder cancers coalesced into one subtype typified by TP53 alterations, TP63 amplifications, and high expression of immune and proliferation pathway genes. Of note, bladder cancers split into three pan-cancer subtypes. The multi-platform classification, while correlated with tissue-of-origin, provides independent information for predicting clinical outcomes. All datasets are available for data-mining from a unified resource to support further biological discoveries and insights into novel therapeutic strategies
