Evidence for a Massive Dark Object in NGC 4350
In this work we build a detailed dynamic model for a S0 galaxy possibly
hosting a central massive dark object (MDO). We show that the photometric
profiles and the kinematics along the major and minor axes, including the h3
and h4 profiles, imply the presence of a central MDO of mass M = 1.5 - 9.7 10^8
solar masses, i.e. 0.3-2.8% of the mass derived for the stellar spheroidal
component. Models without MDO are unable to reproduce the kinematic properties
of the inner stars and of the rapidly rotating nuclear gas.
The stellar population comprise of an exponential disc (27% of the light) and
a diffuse spheroidal component (73% of the light) that cannot be represented by
a simple de Vaucouleurs profile at any radius. The M/L ratios we found for the
stellar components (respectively 3.3 and 6.6) are typical of those of disc and

Spontaneous creation of discrete breathers in Josephson arrays
We report on the experimental generation of discrete breather states
(intrinsic localized modes) in frustrated Josephson arrays. Our experiments
indicate the formation of discrete breathers during the transition from the
static to the dynamic (whirling) system state, induced by a uniform external
current. Moreover, spatially extended resonant states, driven by a uniform
current, are observed to evolve into localized breather states. Experiments
were performed on single Josephson plaquettes as well as open-ended Josephson
ladders with 10 and 20 cells. We interpret the breather formation as the result
of the penetration of vortices into the system.Comment: 5 pages, 5 figure
The WINGS Survey: a progress report
A two-band (B and V) wide-field imaging survey of a complete, all-sky X-ray
selected sample of 78 clusters in the redshift range z=0.04-0.07 is presented.
The aim of this survey is to provide the astronomical community with a complete
set of homogeneous, CCD-based surface photometry and morphological data of
nearby cluster galaxies located within 1.5 Mpc from the cluster center. The
data collection has been completed in seven observing runs at the INT and
ESO-2.2m telescopes. For each cluster, photometric data of about 2500 galaxies
(down to V~23) and detailed morphological information of about 600 galaxies
(down to V~21) are obtained by using specially designed automatic tools.
As a natural follow up of the photometric survey, we also illustrate a long
term spectroscopic program we are carrying out with the WHT-WYFFOS and AAT-2dF
multifiber spectrographs. Star formation rates and histories, as well as
metallicity estimates will be derived for about 350 galaxies per cluster from
the line indices and equivalent widths measurements, allowing us to explore the
link between the spectral properties and the morphological evolution in high-
to low-density environments, and across a wide range in cluster X-ray

gp91phox-dependent expression of platelet CD40 ligand
Background-CD40 ligand (CD40L) expression on platelets is mediated by agonists, but the underlying mechanism is still unclear. Methods and Results-CD40L expression was measured in platelets from healthy subjects both with and without the addition of antioxidants or a phospholipase A2 (PLA2) inhibitor and in platelets from 2 patients with an inherited deficiency of gp91phox. Immunoprecipitation analysis was also performed to determine whether normal platelets showed gp91phox expression. Unlike catalase and mannitol, superoxide dismutase inhibited agonist-induced platelet CD40L expression in healthy subjects. Immunoprecipitation analysis also showed that platelets from healthy subjects expressed gp91phox. In 2 male patients with inherited gp91phox deficiency, collagen-, thrombin-, and arachidonic acid-stimulated platelets showed an almost complete absence of superoxide anion (O2-) and CD40L expression. Incubation of platelets from healthy subjects with a PLA2 inhibitor almost completely prevented agonist-induced O2- and CD40L expression. Conclusions-These data provide the first evidence that platelet CD40L expression occurs via arachidonic acid-mediated gp91phox activation
3D electron diffraction in nano-geology: present and perspectives
When working on advanced research topics in geosciences, one must often deal with small yields and cryptocrystalline polyphasic samples. Conventional optical and X-ray crystallographic tools may not be sufficient for the proper characterization of these samples. The development of efficient probes able to investigate the nanoworld becomes
therefore crucial for pushing forward our understanding about the geochemical and mineralogical processes that regulate Earth and extraterrestrial environments.
In the last ten years, electron diffraction (ED) evolved from a qualitative method restricted to few dedicated TEM users, to a robust protocol for phase identification and abinitio structure determination [1]. Such change has been mostly propelled by the development of routines for 3D data collection. This methodology is in principle equivalent to
single-crystal X-ray diffraction, but allows sampling crystals of few tens on nanometers. We will show here some examples of recent applications of ED in geosciences, namely how to achieve an easy and relatively fast characterization of minor and cryptocrystalline phases in natural and experimental samples. We were able identify and characterize modulated phases able to carry hydrogen at upper-mantle conditions, to follow aragonite growth from the first nucleation seeds [2] and to identify mineralogical phases and polytypes in non-equilibrated extraterrestrial samples and in impact rocks [3]
Vitamin C inhibits platelet expression of CD40 ligand
Upon stimulation with agonists, platelets express CD40 ligand (CD40L), a transmembrane protein implicated in the initiation and progression of atherosclerotic disease. We have recently discovered that oxidative stress plays a major role in platelet CD40L expression. In this study, we sought to determine whether vitamin C, a known antioxidant, is able to influence platelet CD40L expression. In vitro experiments were done by stimulating platelets with collagen in the presence or absence of vitamin C (50-100 mu M) or vehicle as control. An in vivo study was done in 10 healthy subjects who were randomized to intravenous infusion of placebo or 1 g vitamin C for 45 min in a crossover design. At the end of infusion platelet CD40L and O2- were measured. The in vitro study demonstrated that vitamin C dose dependently inhibited platelet CD40L expression without affecting agonist-induced platelet aggregation. In subjects treated with placebo no changes of platelet CD40L and O2- were observed; conversely, vitamin C infusion caused a significant and parallel decrease of platelet O2- (-70%, P < 0.001) and CD40L (-68%, P < 0.001). Platelet aggregation was not modified by either treatment. This study suggests that water-soluble antioxidants, which scavenge superoxide radicals, may reduce platelet CD40L expression. Insecticide resistance in Anopheles arabiensis in Sudan: temporal trends and underlying mechanisms
Background: Malaria vector control in Sudan relies mainly on indoor residual spraying (IRS) and the use of long lasting insecticide treated bed nets (LLINs). Monitoring insecticide resistance in the main Sudanese malaria vector, Anopheles arabiensis, is essential for planning and implementing an effective vector control program in this country.
Methods: WHO susceptibility tests were used to monitor resistance to insecticides from all four WHO-approved classes of insecticide at four sentinel sites in Gezira state over a three year period. Insecticide resistance mechanisms were studied using PCR and microarray analyses.
Results: WHO susceptibility tests showed that Anopheles arabiensis from all sites were fully susceptible to bendiocarb and fenitrothion for the duration of the study (2008–2011). However, resistance to DDT and pyrethroids was detected at three sites, with strong seasonal variations evident at all sites. The 1014 F kdr allele was significantly associated with resistance to pyrethroids and DDT (P 7 in allelic tests). The 1014S allele was not detected in any of the populations tested. Microarray analysis of the permethrinresistant population of An. arabiensis from Wad Medani identified a number of metabolic genes that were significantly over-transcribed in the field-collected resistant samples when compared to the susceptible Sudanese An. arabiensis Dongola strain. These included CYP6M2 and CYP6P3, two genes previously implicated in pyrethroid resistance in Anopheles gambiae s.s, and the epsilon-class glutathione-S-transferase, GSTe4.
Conclusions: These data suggest that both target-site mechanisms and metabolic mechanisms play an important role in conferring pyrethroid resistance in An. arabiensis from Sudan. Identification in An. arabiensis of candidate loci that have been implicated in the resistance phenotype in An. gambiae requires further investigation to confirm the role of these genes
Oxidative stress-mediated platelet CD40 ligand upregulation in patients with hypercholesterolemia: effect of atorvastatin
Objectives: We speculated that in patients with hypercholesterolemia CD40L overexpression could depend on low-density lipoprotein (LDL)-induced enhanced intraplatelet formation of O-2(.-) and statin could reduce platelet CD40L via interference with platelet O-2(.-) production. Background: CD40L is a protein with inflammatory and thrombotic properties. CD40L is upregulated in platelets from hypercholesterolemic (HC) patients but the underlying mechanism is unclear. Methods: Collagen-induced platelet CD40L and platelet O-2(.-) expression were investigated in 40 HC patients and 40 healthy subjects. HC patients were then randomized to either a diet (n = 20) (group A) or atorvastatin 10 mg day (n = 20) (group B); the above variables were measured at baseline and after 3 and 30 days of treatment. O-2(.-) and CD40L were also measured in vitro in LDL-treated platelets with or without nicotinamide adenine dinucleotide phosphate (NADPH) oxidase inhibitor or atorvastatin added. Results: Compared with controls, HC patients showed higher values of platelet CD40L (P < 0.001) and O-2(.-) (P < 0.001). Platelet CD40L was significantly correlated with O-2(.-) (P < 0.001). The interventional trial showed no changes in group A and a significant and parallel decrease in platelet CD40L (P < 0.001) and O-2(.-) (P < 0.001) in group B. In vitro studies demonstrated that LDL-induced platelet CD40L and GP IIb/IIIa (PAC1 binding) activation via the NADPH oxidase pathway. CD40L upregulation was counteracted by atorvastatin in a dose-dependent fashion. Conclusions: This study suggests that in patients with hypercholesterolemia platelet CD40L is upregulated via NADPH oxidase-dependent O-2(.-) generation. Atorvastatin downregulated CD40L with an oxidative stress-mediated mechanism likely involving platelet NADPH oxidase, an effect that seemed to be independent of its cholesterol-lowering action
