    Glaucomatous Visual Field Defect Severity and the Prevalence of Motor Vehicle Collisions in Japanese: A Hospital/Clinic-Based Cross-Sectional Study

    Purpose. This study examined the association between the severity of visual field defects and the prevalence of motor vehicle collisions (MVCs) in subjects with primary open-angle glaucoma (POAG). Methods. This is a cross-sectional study. Japanese patients who have had driver’s licence between 40 and 85 years of age were screened for eligibility. Participants answered a questionnaire about MVCs experienced during the previous 5 years. Subjects with POAG were classified as having mild, moderate, or severe visual field defect. We evaluated associations between the severity of POAG and the prevalence of MVCs by logistic regression models. Results. The prevalence of MVCs was significantly associated with the severity of POAG categorized by worse eye MD (control: 30/187 = 16.0%; mild POAG: 17/92 = 18.5%; moderate POAG: 14/60 = 23.3%; severe POAG: 14/47 = 29.8%; P=0.025, Cochran-Armitage trend test). Compared to the control group, the adjusted OR for MVC prevalence in subjects with mild, moderate, or severe POAG in the worse eye was 1.07 (95% CI: 0.55 to 2.10), 1.44 (95% CI: 0.68 to 3.08), and 2.28 (95% CI: 1.07 to 4.88). Conclusions. There is a significant association between the severity of glaucoma in the worse eye MD and the prevalence of MVCs

    An NMR strategy for fragment-based ligand screening utilizing a paramagnetic lanthanide probe

    A nuclear magnetic resonance-based ligand screening strategy utilizing a paramagnetic lanthanide probe is presented. By fixing a paramagnetic lanthanide ion to a target protein, a pseudo-contact shift (PCS) and a paramagnetic relaxation enhancement (PRE) can be observed for both the target protein and its bound ligand. Based on PRE and PCS information, the bound ligand is then screened from the compound library and the structure of the ligand–protein complex is determined. PRE is an isotropic paramagnetic effect observed within 30 Å from the lanthanide ion, and is utilized for the ligand screening in the present study. PCS is an anisotropic paramagnetic effect providing long-range (~40 Å) distance and angular information on the observed nuclei relative to the paramagnetic lanthanide ion, and utilized for the structure determination of the ligand–protein complex. Since a two-point anchored lanthanide-binding peptide tag is utilized for fixing the lanthanide ion to the target protein, this screening method can be generally applied to non-metal-binding proteins. The usefulness of this strategy was demonstrated in the case of the growth factor receptor-bound protein 2 (Grb2) Src homology 2 (SH2) domain and its low- and high-affinity ligands

    本研究は、北海道産にしんみその調理科学的特性を明らかにし、にしんみそを北海道食材や大学生対象の調理実習ならびに二次加工品への応用を検討することを目的とした。1 色調は、にしんみそに調味料を添加することによって、明るさを示す明度が高くなった。2 にしんみその粘度は、25℃において1220~1550mPa.sを示していた。3 官能検査の結果、評点はにしんみそペーストでは味、にしんみそディップでは総合評価が高くなった。官能検査結果より、いずれの項目においてもプラスの評点を示し、にしんみそを用いた展開が可能であることが示唆された。4大学生が「使いたい」と思うにしんみその応用例では、焼きおにぎり、石狩鍋、野菜スティックのみそなどが挙げられていた。5 大学生が考案したにしんみそを用いたレシピでは、にしんジャージャー麺、にしん梅そぼろそば、にしんマフィンや蒸しパンなどが挙げられていた。In this study,we clarified the scientific cooking characteristics of herring miso from Hokkaido, with the aim of applying herring miso to cooking training for university students and as a commercial product of Hokkaido. The addition of herring miso as a seasoning leads to an increase in the luminosity, indicating brightness, of the food. Herring miso shows a viscosity of 1220~1550mPa.s at 25℃. The results of sensory testing showed that herring miso paste rated highly for taste, while herring miso dip rated highly overall. Both items were positively rated, indicating the possible application of herring miso. The applications that university students reported as "wanting to try", included fried rice balls, Ishikari-nabe and vegetable sticks with miso. The recipes devised by students utilizing herring miso, included herring noodles, herring plum soboro (rousong) soba, and herring muffins and herring mushi pan (steamed bread). Based on the above, it appears there is a definite possibility of using herring miso in dishes using Hokkaido produce as well as in the development of cooking training for university students, and we hereafter plan to examine future applications

    Post-intervention Status in Patients With Refractory Myasthenia Gravis Treated With Eculizumab During REGAIN and Its Open-Label Extension

    OBJECTIVE: To evaluate whether eculizumab helps patients with anti-acetylcholine receptor-positive (AChR+) refractory generalized myasthenia gravis (gMG) achieve the Myasthenia Gravis Foundation of America (MGFA) post-intervention status of minimal manifestations (MM), we assessed patients' status throughout REGAIN (Safety and Efficacy of Eculizumab in AChR+ Refractory Generalized Myasthenia Gravis) and its open-label extension. METHODS: Patients who completed the REGAIN randomized controlled trial and continued into the open-label extension were included in this tertiary endpoint analysis. Patients were assessed for the MGFA post-intervention status of improved, unchanged, worse, MM, and pharmacologic remission at defined time points during REGAIN and through week 130 of the open-label study. RESULTS: A total of 117 patients completed REGAIN and continued into the open-label study (eculizumab/eculizumab: 56; placebo/eculizumab: 61). At week 26 of REGAIN, more eculizumab-treated patients than placebo-treated patients achieved a status of improved (60.7% vs 41.7%) or MM (25.0% vs 13.3%; common OR: 2.3; 95% CI: 1.1-4.5). After 130 weeks of eculizumab treatment, 88.0% of patients achieved improved status and 57.3% of patients achieved MM status. The safety profile of eculizumab was consistent with its known profile and no new safety signals were detected. CONCLUSION: Eculizumab led to rapid and sustained achievement of MM in patients with AChR+ refractory gMG. These findings support the use of eculizumab in this previously difficult-to-treat patient population. CLINICALTRIALSGOV IDENTIFIER: REGAIN, NCT01997229; REGAIN open-label extension, NCT02301624.
