Oriented antibody immobilization to polystyrene macrocarriers for immunoassay modified with hydrazide derivatives of poly(meth)acrylic acid
BACKGROUND: Hydrophobic polystyrene is the most common material for solid phase immunoassay. Proteins are immobilized on polystyrene by passive adsorption, which often causes considerable denaturation. Biological macromolecules were found to better retain their functional activity when immobilized on hydrophilic materials. Polyacrylamide is a common material for solid-phase carriers of biological macromolecules, including immunoreagents used in affinity chromatography. New macroformats for immunoassay modified with activated polyacrylamide derivatives seem to be promising. RESULTS: New polymeric matrices for immunoassay in the form of 0.63-cm balls which contain hydrazide functional groups on hydrophilic polymer spacer arms at their surface shell are synthesized by modification of aldehyde-containing polystyrene balls with hydrazide derivatives of poly(meth)acrylic acid. The beads contain up to 0.31 μmol/cm(2) active hydrazide groups accessible for covalent reaction with periodate-oxidized antibodies. The matrices obtained allow carrying out the oriented antibody immobilization, which increases the functional activity of immunosorbents. CONCLUSIONS: An efficient site-directed antibody immobilization on a macrosupport is realized. The polymer hydrophilic spacer arms are the most convenient and effective tools for oriented antibody coupling with molded materials. The suggested scheme can be used for the modification of any other solid supports containing electrophilic groups reacting with hydrazides
Fourier transform spectroscopy and coupled-channel deperturbation treatment of the A1Sigma+ ~ b3Pi complex of KCs molecule
The laser induced fluorescence (LIF) spectra A1Sigma ~ b3Pi --> X1Sigma+ of
KCs dimer were recorded in near infrared region by Fourier Transform
Spectrometer with a resolution of 0.03 cm-1. Overall more than 200 LIF spectra
were rotationally assigned to 39K133Cs and 41K133Cs isotopomers yielding with
the uncertainty of 0.003-0.01 cm-1 more than 3400 rovibronic term values of the
strongly mixed singlet A1Sigma+ and triplet b3Pi states. Experimental data
massive starts from the lowest vibrational level v_A=0 of the singlet and
nonuniformly cover the energy range from 10040 to 13250 cm-1 with rotational
quantum numbers J from 7 to 225. Besides of the dominating regular A1Sigma+ ~
b3P Omega=0 interactions the weak and local heterogenous A1S+ ~ b3P Omega=1
perturbations have been discovered and analyzed. Coupled-channel deperturbation
analysis of the experimental 39K133Cs e-parity termvalues of the A1S+ ~ b3P
complex was accomplished in the framework of the phenomenological 4 x 4
Hamiltonian accounting implicitly for regular interactions with the remote
states manifold. The resulting diabatic potential energy curves of the
interacting states and relevant spin-orbit coupling matrix elements defined
analytically by Expanded Morse Oscillators model reproduce 95% of experimental
data field of the 39K133Cs isotopomer with a standard deviation of 0.004 cm-1
which is consistent with the uncertainty of the experiment. Reliability of the
derived parameters was additionally confirmed by a good agreement between the
predicted and experimental termvalues of 41K133Cs isotopomer. Calculated
intensity distributions in the A ~ b --> X LIF progressions are also consistent
with their experimental counterparts.Comment: 17 pages, 14 figure
Lexical-Semantic Explication of Concept DEATH in Yakut language
Еhe results of the lexical-semantic explication of a universal concept of DEATH in the Yakut language picture of the world are presented. The relevance of the work is determined by the fact that the study of concepts, occupying a significant place in the Yakut linguistic culture, is not currently completed. It is also noted that linguists have always shown interest in the content of the concept of DEATH and peculiarities of its lexical representation by means of different languages, because it belongs to the universal concepts. Euphemization in of death designation is emphasized. The material of the research were sustainable verbal complexes of the Yakut language, signifying death. Prepared by the authors the literal translations of collocations are presented. For example, the expressions are given which literally can be translated as follows: “to disappear from white sun world,” “to go into the pit,” “to go out on the hillside,” “soul became cold” etc. The analysis involve phraseology with other meanings: ‘to deprive of life, kill,’ ‘to go on a deadly risk’ etc. They confirm and clarify the metaphors identified in the primary material. Vocabulary is analysed with respect to ethnographic background information about the traditions of the Yakut graves, about the attitude to suicide and others. Following models of ideas about death are revealed on the Yakut material: “death is a transmigration to another world,” “death is the end,” “death is darkness,” “death is sleep” etc
