Zanvil Alexander Cohn 1926-1993
Zanvil Alexander Cohn, an editor of this Journal since 1973, died suddenly on June 28, 1993. Cohn is best known as the father of the current era of macrophage biology. Many of his scientific accomplishments are recounted here, beginning with seminal studies on the granules of phagocytes that were performed with his close colleague and former editor of thisJournaI, James Hirsch. Cohn and Hirsch identified the granules as lysosomes that discharged their contents of digestive enzymes into vacuoles containing phagocytosed microbes. These findings were part of the formative era of cell biology and initiated the modern study of endocytosis and cell-mediated resistance to infection. Cohn further explored the endocytic apparatus in pioneering studies of the mouse peritoneal macrophage in culture. He described vesicular inputs from the cell surface and Golgi apparatus and documented the thoroughness of substrate digestion within lysosomal vacuoles that would only permit the egress of monosaccharides and amino acids. These discoveries created a vigorous environment for graduate students, postdoctoral fellows, and junior and visiting faculty. Some of the major findings that emerged from Cohn’s collaborations included the radioiodination of the plasma membrane for studies of composition and turnover; membrane recycling during endocytosis; the origin of the mononuclear phagocyte system in situ; the discovery of the dendritic cell system of antigen-presenting cells; the macrophage as a secretory cell, including the release of proteases and large amounts of prostaglandins and leukotrienes; several defined parameters of macrophage activation, especially the ability of T cell-derived lymphokines to enhance killing of tumor cells and intracellular protozoa; the granule discharge mechanism whereby cytotoxic lymphocytes release the pore-forming protein perforin; the signaling of macrophages via myristoylated substrates of protein kinase C; and a tissue culture model in which monocytes emigrate across tight endothelial junctions. In 1983, Cohn turned to a long-standing goal of exploring host resistance directly in humans. He studied leprosy, focusing on the disease site, the parasitized macrophages of the skin. He injected recombinant lymphokines into the skin and found that these molecules elicited several cell-mediated responses. Seeing this potential to enhance host defense in patients, Cohn was extending his clinical studies to AIDS and tuberculosis. Zanvil Cohn was a consummate physician-scientist who nurtured the relationship between cell biology and infectious disease. He guided with a warm but incisive manner the careers of many individuals. He was deeply committed to several institutions of biomedical research; to medicine in the developing world; to The Rockefeller University, especially its programs for graduate study and patient-oriented research; and to the energy and spirit of this Journal

Development of the Variable Dexterity Test: construction, reliability and validity
Background/Aims: This article introduces a dexterity test designed to assess individual types of
dexterity used to carry out activities of daily living (ADL). The Variable Dexterity Test (VDT) was
developed as part of a wider study, the broader aim being to fully understand dexterity and its effect
on human-product interaction during ADL. This was done with a view to improve occupational therapy
methods when assessing dexterity and general hand function.
Methods: The control group consisted of 24 healthy participants. Estimates of reliability and validity
were evaluated in this pilot study. Inter-rater and test-retest reliability were assessed using a one-way
ANOVA. The validity of the test was estimated by correlating participants’ VDT scores with their
proficiency to complete four ADL task actions and a standardised dexterity test (Purdue Pegboard Test).
Results: The test produced consistent results among the control group with both a single assessor
(test‑retest reliability) and multiple assessors (inter‑rater reliability). High correlations between
participants’ VDT scores and proficiency to perform ADL were found for most of the subtests. There
was also a high correlation between participants’ scores from the Purdue Pegboard Test and the VDT.
Conclusions: The VDT proved to be a flexible, reliable and valid tool that assesses dexterity based on
ability to carry out ADL. Validity and reliability estimates show encouraging values, which recognises
that the VDT can be used as an accurate method to assess more than one type of dexterity.</p

EQ-5D in skin conditions: an assessment of validity and responsiveness
Aims and objectives This systematic literature review aims to assess the reliability, validity and responsiveness of three widely used generic preference-based measures of health-related quality of life (HRQL), i.e., EQ-5D, Health Utility Index 3 (HUI3) and SF-6D in patients with skin conditions. Methods A systematic search was conducted to identify studies reporting health state utility values obtained using EQ-5D, SF-6D, or HUI3 alongside other HRQL measures or clinical indices for patients with skin conditions. Data on test-retest analysis for reliability, known group differences or correlation and regression analyses for validity, and change over time or responsiveness indices analysis were extracted and reviewed. Results A total of 16 papers reporting EQ-5D utilities in people with skin conditions were included in the final review. No papers for SF-6D and HUI3 were found. Evidence of reliability was not found for any of these measures. The majority of studies included in the review (12 out of 16) examined patients with plaque psoriasis or psoriatic arthritis and the remaining four studies examined patients with either acne, hidradenitis suppurativa, hand eczema, or venous leg ulcers. The findings were generally positive in terms of performance of EQ-5D. Six studies showed that EQ-5D was able to reflect differences between severity groups and only one reported differences that were not statistically significant. Four studies found that EQ-5D detected differences between patients and the general population, and differences were statistically different for three of them. Further, moderate-to-strong correlation coefficients were found between EQ-5D and other skin-specific HRQL measures in four studies. Eight studies showed that EQ-5D was able to detect change in HRQL appropriately over time and the changes were statistically significant in seven studies. Conclusions Overall, the validity and responsiveness of the EQ-5D was found to be good in people with skin diseases, especially plaque psoriasis or psoriatic arthritis. No evidence on SF-6D and HUI3 was available to enable any judgments to be made on their performance

Comparison of Disturbed and Undisturbed Soil Core Methods to Estimate Nitrogen-Mineralization Rates in Manured Agricultural Soils
Ion exchange resin/soil cores are a common in situ approach to estimating soil nitrogen (N) mineralization rates. However, no studies compare the two common methods of core preparation (disturbed and undisturbed). The objective of our study was to compare N mineralized and soil temperature in disturbed versus undisturbed cores of manured agricultural soils. Undisturbed cores were prepared by driving aluminum tubes (25 cm long with 10 cm inner diameter) into soil, removing the tubes, and then inserting an ion-exchange resin bag beneath the soil at the bottom of the tube. Disturbed cores were prepared with the same materials, but soil was excavated, mixed, and then filled into tubes fitted with ion-exchange resin bags at the bottom. Soil from six agricultural fields (five of which had more than 10 years of regular dairy manure application) was incubated over four time periods during summer and winter. A total of 13 soil/incubation-period combinations were tested. Disturbed cores tended to have more N mineralized than undisturbed cores (P < 0.10), especially in cores prepared with the lowest clay content soil. However, variability of N mineralized was lower in disturbed cores than undisturbed cores for 11 of the 13 soil/incubation periods. This lower variability was significant in two of the four incubation periods (P < 0.10). There was little difference in mean soil temperatures in disturbed versus undisturbed cores or within cores versus outside but adjacent to cores.
