    Malignancies in Korean Patients with Inflammatory Myopathy

    The aim of this study was to assess the prevalence and the common type of malignancies in Korean patients with polymyositis (PM) and dermatomyositis (DM) and to evaluate the differences of clinical and laboratory findings between patients with malignancy and those without malignancy. Forty-one Korean patients, who were diagnosed as PM or DM, were enrolled in this study. They fulfilled the Bohan and Peter's criteria for a definite diagnosis of PM and DM. Patients with PM were 25 and those with DM were 16. Eleven out of 41 patients (26.8%) had malignancies. The malignancy was diagnosed simultaneously or later in 81.8% of patients with inflammatory myopathy (IM). The breast cancer was the most common malignancy. In this study, forty three years old as a screening age for malignancy had 88.9% sensitivity and 50.2% specificity. The serum levels of creatine kinase (CK) were significantly lower in patients with malignancy than those without malignancy

    A Study on Self-Diagnosis Method to Prevent the Spread of COVID-19 Based on SVM

    In this paper, a study was conducted to find a self-diagnosis method to prevent the spread of COVID-19 based on machine learning. COVID-19 is an infectious disease caused by a newly discovered coronavirus. According to WHO(World Health Organization)’s situation report published on May 18th, 2020, COVID-19 has already affected 4,600,000 cases and 310,000 deaths globally and still increasing. The most severe problem of COVID-19 virus is that it spreads primarily through droplets of saliva or discharge from the nose when an infected person coughs or sneezes, which occurs in everyday life. And also, at this time, there are no specific vaccines or treatments for COVID-19.Because of the secure diffusion method and the absence of a vaccine, it is essential to self-diagnose or do a self-diagnosis questionnaire whenever possible. But self-diagnosing has too many questions, and ambiguous standards also take time. Therefore, in this study, using SVM(Support Vector Machine), Decision Tree and correlation analysis found two vital factors to predict the infection of the COVID-19 virus with an accuracy of 80%. Applying the result proposed in this paper, people can self-diagnose quickly to prevent COVID-19 and further prevent the spread of COVID-19

    Toward a Standardized Strategy of Clinical Metabolomics for the Advancement of Precision Medicine

    Despite the tremendous success, pitfalls have been observed in every step of a clinical metabolomics workflow, which impedes the internal validity of the study. Furthermore, the demand for logistics, instrumentations, and computational resources for metabolic phenotyping studies has far exceeded our expectations. In this conceptual review, we will cover inclusive barriers of a metabolomics-based clinical study and suggest potential solutions in the hope of enhancing study robustness, usability, and transferability. The importance of quality assurance and quality control procedures is discussed, followed by a practical rule containing five phases, including two additional "pre-pre-" and "post-post-" analytical steps. Besides, we will elucidate the potential involvement of machine learning and demonstrate that the need for automated data mining algorithms to improve the quality of future research is undeniable. Consequently, we propose a comprehensive metabolomics framework, along with an appropriate checklist refined from current guidelines and our previously published assessment, in the attempt to accurately translate achievements in metabolomics into clinical and epidemiological research. Furthermore, the integration of multifaceted multi-omics approaches with metabolomics as the pillar member is in urgent need. When combining with other social or nutritional factors, we can gather complete omics profiles for a particular disease.     Role of G{alpha}12 and G{alpha}13 as Novel Switches for the Activity of Nrf2, a Key Antioxidative Transcription Factor

    G{alpha}12 and G{alpha}13 function as molecular regulators responding to extracellular stimuli. NF-E2-related factor 2 (Nrf2) is involved in a protective adaptive response to oxidative stress. This study investigated the regulation of Nrf2 by G{alpha}12 and G{alpha}13. A deficiency of G{alpha}12, but not of G{alpha}13, enhanced Nrf2 activity and target gene transactivation in embryo fibroblasts. In mice, G{alpha}12 knockout activated Nrf2 and thereby facilitated heme catabolism to bilirubin and its glucuronosyl conjugations. An oligonucleotide microarray demonstrated the transactivation of Nrf2 target genes by G{alpha}12 gene knockout. G{alpha}12 deficiency reduced Jun N-terminal protein kinase (JNK)-dependent Nrf2 ubiquitination required for proteasomal degradation, and so did G{alpha}13 deficiency. The absence of G{alpha}12, but not of G{alpha}13, increased protein kinase C {delta} (PKC {delta}) activation and the PKC {delta}-mediated serine phosphorylation of Nrf2. G{alpha}13 gene knockout or knockdown abrogated the Nrf2 phosphorylation induced by G{alpha}12 deficiency, suggesting that relief from G{alpha}12 repression leads to the G{alpha}13-mediated activation of Nrf2. Constitutive activation of G{alpha}13 promoted Nrf2 activity and target gene induction via Rho-mediated PKC {delta} activation, corroborating positive regulation by G{alpha}13. In summary, G{alpha}12 and G{alpha}13 transmit a JNK-dependent signal for Nrf2 ubiquitination, whereas G{alpha}13 regulates Rho-PKC {delta}-mediated Nrf2 phosphorylation, which is negatively balanced by G{alpha}12

    ALTERATIONS IN JOINT KINEMATICS AND KINETICS DURING DOWNHILL RUNNING

    The purpose of this investigation was to find how joint kinematics and kinetics during downhill running change compared to level running. Fifteen recreational runners ran on a force plate imbedded treadmill with three different slopes (0 ΒΊ, -6ΒΊ, and -9ΒΊ) at a controlled speed of 3.2 m/s. Ten steps on each slope were selected for analysis. Increased knee flexion with decreased ankle plantar-flexion and hip flexion was found during downhill running compared to level running. Decreased peak propulsive ground reaction force and posterior impulse were found during downhill running compared to level running. Additionally, increased extension moment with increased negative joint power at the knee and decreased plantar-flexion moment with decreased negative joint power at the ankle were found during downhill running compared to level running
