    Functional Differences between GDNF-Dependent and FGF2-Dependent Mouse Spermatogonial Stem Cell Self-Renewal

Earlier studies showed that glial cell line-derived neurotrophic factor (GDNF) was indispensable for SSC self-renewal by binding to the GFRA1/RET receptor. Mice with mutations in these molecules showed impaired spermatogenesis, which was attributed to SSC depletion. Here we show that SSCs undergo GDNF-independent self-renewal. A small number of spermatogonia formed colonies when testis fragments from a Ret mutant mouse strain were transplanted into heterologous recipients. Moreover, fibroblast growth factor 2 (FGF2) supplementation enabled in vitro SSC expansion without GDNF. Although GDNF-mediated self-renewal signaling required both AKT and MAP2K1/2, the latter was dispensable in FGF2-mediated self-renewal. FGF2-depleted testes exhibited increased levels of GDNF and were enriched for SSCs, suggesting that the balance between FGF2 and GDNF levels influences SSC self-renewal in vivo. Our results show that SSCs exhibit at least two modes of self-renewal and suggest complexity of SSC regulation in vivo

    Invited Paper: Effective Retrieval of Educational Resources by Using Learning Object Metadata for K-12 Schools in Japan

    We have customized learning object metadata (LOM) for educational resources on the basis of the LOM investigated by IEEE and IMS, in order to make it consistent with Japanese K-12 school education. Since many specifications for the metadata are included in the customized LOM, we extracted necessary metadata especially for taking into account the retrieval of educational resources in schools. The extracted LOM has been installed with more than 13,000 educational resources individually. These resources have been registered in the search engine we developed using the LOM and are now available on the Internet as a first trial for applying the LOM. Using the LOM in the search engine makes it easy for teachers and students to find appropriate resources for their learning needs

    Emission characteristics of diethylhexyl phthalate (DEHP) from building materials determined using a passive flux sampler and micro-chamber.

    Emission rates of diethylhexyl phthalate (DEHP) from building materials, such as vinyl floorings and wall paper, determined using a passive flux sampler (PFS) were constant over the week-long measurement period. Emission rates for vinyl floorings and wallpaper were linearly correlated to the inverse of diffusion distance, which corresponds to the internal depth of the PFS. Surface-air DEHP concentrations (y0) were estimated as 1.3-2.3 μg/m3 for materials having a boundary layer molecular diffusion rate-limiting step. The partition coefficient (Kmaterial-air) was estimated as 3.3-7.5 × 1010 for these materials. Additionally, emission rates of DEHP from same building materials determined using a micro-chamber were 4.5-6.1 μg/m2/h. Mass transfer coefficients in the micro-chamber (hm) were estimated by comparing the results using the PFS and micro-chamber, and these were 1.1-1.2 × 10-3 and 8.1 × 10-4 m/s for vinyl floorings (smooth surface) and wallpaper (rough surface), respectively. The thickness of boundary layer on the surface of building materials in the micro-chamber were estimated to be 2.5-2.6 and 3.7 mm for vinyl floorings and wallpaper, respectively

    Three-year evaluation of different adhesion strategies in non-carious cervical lesion restorations: a randomized clinical trial

    Objective: To evaluate non-carious cervical lesions (NCCLs) restored with different adhesion strategies. Methodology: This is a prospective, randomized, double-blind, split-mouth study. An adhesive restorative system (Single Bond Universal/Filtek Z350XT – SBU) was evaluated both without and with selective enamel conditioning (E-SBU), resin-modified glass-ionomer cements (Vitremer; RMGIC), and ethylenediaminetetraacetic acid pretreatment (EDTA; E-RMGIC). In total, 200 restorations, placed in 50 patients, were evaluated at baseline and at a 3-year follow-up using the modified United States Public Health Service (USPHS) criteria. Data were analyzed using the two-proportion equality test, multinomial logistic regression, Wilcoxon test, and Kaplan-Meier survival curves. Results: In total, 42 (84%) patients returned for the 3-year follow-up. SBU showed restoration losses statistically different from RMGIC. Retention was also statistically different in SBU between baseline and the 3-year follow-up. Marginal defects and surface texture were statistically significant for all groups in the period studied, except for the surface texture of SBU and the marginal integrity in E-RMGIC. We observed no statistically significant difference in wear, secondary caries, anatomical form, surface staining, and color over time. Recession degree was the only factor to influence retention rates. Cumulative survival (%) was 89, 98, 98, and 95.3, for SBU, SE-SBU, RMGIC, and E-RMGIC, respectively, without significant differences among them. There was a statistically significant difference between survival curves; however, multiple comparison procedures found no statistical differences. Conclusion: Selective enamel etching affected the retention of non-carious cervical restorations. Adhesion using EDTA and resin-modified glass-ionomer cements delayed marginal defects over time. The degree of gingival recession influenced retention rates. Resin composite restorations showed initial marginal defects, and ionomer restorations, reduced surface luster. EDTA pre-treatment followed by resin-modified glass-ionomer cements may be a promising adhesion strategy for NCCL restorations
