Hierarchical QoS routing in next generation optical networks
In this paper, we study the problem of inter-domain routing with two additive QoS constraints in hierarchical optical networks. We develop an inter-domain routing protocol that (1) identifies the QoS supported by the paths, (2) selects an inter-domain path that satisfies the QoS requirement of a connection request, and (3) reserves the wavelength on each link along the path in such a way that the number of wavelength converters needed is minimized. Both formal analyses and extensive simulation experiments show that our inter-domain routing protocol outperforms the existing protocols. © 2006 IEEE.published_or_final_versio
Quality-of-service routing with two concave constraints
Routing is a process of finding a network path from a source node to a destination node. A good routing protocol should find the "best path" from a source to a destination. When there are independent constraints to be considered, the "best path" is not well-defined. In our previous work, we developed a line segment representation for Quality-of-Service routing with bandwidth and delay requirements. In this paper, we propose how to adopt the line segment when a request has two concave constraints. We have developed a series of operations for constructing routing tables under the distance-vector protocol. Hop-by-hop routing in wireless mesh networks with bandwidth guarantees
Wireless Mesh Network (WMN) has become an important edge network to provide Internet access to remote areas and wireless connections in a metropolitan scale. In this paper, we study the problem of identifying the maximum available bandwidth path, a fundamental issue in supporting quality-of-service in WMNs. Due to interference among links, bandwidth, a well-known bottleneck metric in wired networks, is neither concave nor additive in wireless networks. We propose a new path weight which captures the available path bandwidth information. We formally prove that our hop-by-hop routing protocol based on the new path weight satisfies the consistency and loop-freeness requirements. The consistency property guarantees that each node makes a proper packet forwarding decision, so that a data packet does traverse over the intended path. Our extensive simulation experiments also show that our proposed path weight outperforms existing path metrics in identifying high-throughput paths. © 2012 IEEE.published_or_final_versio

Annual rings in a native Hawaiian tree, Sophora chrysophylla, on Maunakea, Hawaiʻi
Annual rings are not commonly produced in tropical trees because they grow in a relatively aseasonal environment. However, in the subalpine zones of Hawaiʻi's highest volcanoes, there is often strong seasonal variability in temperature and rainfall. Using classical dendrochronological methods, annual growth rings were shown to occur in Sophora chrysophylla, a native tree species on Maunakea, Hawaiʻi. Chronologies were established from nearby non-native, live conifer trees and these were used to verify the dates from a total of 52 series from 22 S. chrysophylla trees, establishing an 86-y chronology (1926–2011). Ring-width patterns were significantly correlated with monthly rainfall from August of the previous year. A systematic review of economic evaluations of whole genome sequencing for the surveillance of bacterial pathogens
Whole-genome sequencing (WGS) has unparalleled ability to distinguish between bacteria, with many public health applications. The generation and analysis of WGS data require significant financial investment. We describe a systematic review summarizing economic analyses of genomic surveillance of bacterial pathogens, reviewing the evidence for economic viability. The protocol was registered on PROSPERO (CRD42021289030). Six databases were searched on 8 November 2021 using terms related to ‘WGS’, ‘population surveillance’ and ‘economic analysis’. Quality was assessed with the Drummond–Jefferson checklist. Following data extraction, a narrative synthesis approach was taken. Six hundred and eighty-one articles were identified, of which 49 proceeded to full-text screening, with 9 selected for inclusion. All had been published since 2019. Heterogeneity was high. Five studies assessed WGS for hospital surveillance and four analysed foodborne pathogens. Four were cost–benefit analyses, one was a cost–utility analysis, one was a cost-effectiveness analysis, one was a combined cost-effectiveness and cost–utility analysis, one combined cost-effectiveness and cost–benefit analyses and one was a partial analysis. All studies supported the use of WGS as a surveillance tool on economic grounds. The available evidence supports the use of WGS for pathogen surveillance but is limited by marked heterogeneity. Further work should include analysis relevant to low- and middle-income countries and should use real-world effectiveness data
Engineering synucleinopathy‐resistant human dopaminergic neurons by CRISPR‐mediated deletion of the SNCA gene
An emerging treatment for Parkinson's disease (PD) is cell replacement therapy. Authentic midbrain dopaminergic (mDA) neuronal precursors can be differentiated from human embryonic stem cells (hESCs) and human induced pluripotent stem cells (iPSCs). These laboratory‐generated mDA cells have been demonstrated to mature into functional dopaminergic neurons upon transplantation into preclinical models of PD. However, clinical trials with human fetal mesenchephalic cells have shown that cell replacement grafts in PD are susceptible to Lewy body formation suggesting host‐to‐graft transfer of α‐synuclein pathology. Here, we have used CRISPR/Cas9n technology to delete the endogenous SNCA gene, encoding for α‐synuclein, in a clinical‐grade hESC line to generate SNCA+/− and SNCA−/− cell lines. These hESC lines were first differentiated into mDA neurons, and then challenged with recombinant α‐synuclein preformed fibrils (PFFs) to seed the formation for Lewy‐like pathology as measured by phosphorylation of serine‐129 of α‐synuclein (pS129‐αSyn). Wild‐type neurons were fully susceptible to the formation of protein aggregates positive for pS129‐αSyn, while SNCA+/− and SNCA−/− neurons exhibited significant resistance to the formation of this pathological mark. This work demonstrates that reducing or completely removing SNCA alleles by CRISPR/Cas9n‐mediated gene editing confers a measure of resistance to Lewy pathology

A tecnologia IPTV explora ao máximo as características da Internet,
com a utilização de mecanismos de Qualidade de Serviço. Surge ainda como uma revolução dentro do panorama televisivo, abrindo portas a novos investimentos por parte das empresas de telecomunicações. A Internet também permite fazer chamadas telefónicas sobre a rede IP. Este serviço é denominado VoIP (Voz sobre IP) e encontra-se em funcionamento já há algum tempo.
Desta forma surge a oportunidade de poder oferecer ao consumidor final, um serviço que
inclua os serviços de Internet, de VoIP e de IPTV denominado serviço Triple Play. O serviço Triple Play veio obrigar a revisão de toda a rede de transporte de forma a preparar a mesma para suportar este serviço de uma forma eficiente (QoS), resiliente (recuperação de falhas) e optimizado
(Engenharia de tráfego).
Em redes de telecomunicações, tanto a quebra de uma ligação como a congestão nas redes
pode interferir nos serviços oferecidos aos consumidores finais. Mecanismos de sobrevivência são aplicados de forma a garantir a continuidade do serviço mesmo na ocorrência de uma falha.
O objectivo desta dissertação é propor uma solução de uma arquitectura de rede capaz de
suportar o serviço Triple Play de uma forma eficiente, resiliente e optimizada através de um encaminhamento óptimo ou quase óptimo. No âmbito deste trabalho, é realizada a análise do impacto das estratégias de encaminhamento que garantem a eficiência, sobrevivência e optimização das redes IP existentes, bem como é determinado o número limite de clientes permitido numa situação de pico de uma dada rede.
Neste trabalho foram abordados os conceitos de Serviços Triple Play, Redes de Acesso,
Redes Núcleo, Qualidade de Serviço, MPLS (Multi-Protocolo Label Switching), Engenharia de Tráfego e Recuperação de falhas. As conclusões obtidas das simulações efectuadas através do simulador de rede NS-2.33 (Network Simulator versão 2.33) serviram para propor a solução da arquitectura de uma rede capaz de suportar o serviço Triple Play de uma forma eficiente, resiliente e optimizada.Orientador: Paulo Nazareno Maia Sampai
Plasticity of the human visual system after retinal gene therapy in patients with Leber's congenital amaurosis.
Much of our knowledge of the mechanisms underlying plasticity in the visual cortex in response to visual impairment, vision restoration, and environmental interactions comes from animal studies. We evaluated human brain plasticity in a group of patients with Leber's congenital amaurosis (LCA), who regained vision through gene therapy. Using non-invasive multimodal neuroimaging methods, we demonstrated that reversing blindness with gene therapy promoted long-term structural plasticity in the visual pathways emanating from the treated retina of LCA patients. The data revealed improvements and normalization along the visual fibers corresponding to the site of retinal injection of the gene therapy vector carrying the therapeutic gene in the treated eye compared to the visual pathway for the untreated eye of LCA patients. After gene therapy, the primary visual pathways (for example, geniculostriate fibers) in the treated retina were similar to those of sighted control subjects, whereas the primary visual pathways of the untreated retina continued to deteriorate. Our results suggest that visual experience, enhanced by gene therapy, may be responsible for the reorganization and maturation of synaptic connectivity in the visual pathways of the treated eye in LCA patients. The interactions between the eye and the brain enabled improved and sustained long-term visual function in patients with LCA after gene therapy

pressure terms are analysed in terms of the constraints parting from energy
density, solar-relativistic effects and galactic dynamics along with the dark
