    Inhibition of post-translational N-glycosylation by HRD1 that controls the fate of ABCG5/8 transporter

    N-glycosylation of proteins in endoplasmic reticulum is critical for protein quality control. We showed here a post-translational N-glycosylation affected by the HRD1 E3 ubiquitin ligase. Both WT- and E3-defective C329S-HRD1 decreased the level of high mannose form of ABCG8, a protein that heterodimerizes with ABCG5 to control sterol balance. Meanwhile, HRD1 increased the non-glycosylated ABCG8 regardless of its E3 activity, thereby suppressing full maturation of ABCG5/8 transporter. Pulse chase and mutational analysis indicated that HRD1 inhibits STT3B-dependent post-translational N-glycosylation of ABCG8. Whereas, HRD1 had only slight effect on the N-glycosylation status of ABCG5; rather it accelerated ABCG5 degradation in an E3 activity-dependent manner. Finally, RMA1, another E3 ubiquitin ligase, accelerated the degradation of both ABCG5 and ABCG8 via E3 activity-dependent manner. HRD1 and RMA1 may therefore be negative regulators of disease-associated transporter ABCG5/ABCG8. The findings also highlight the unexpected E3 activity-independent role of HRD1 in the regulation of N-glycosylation

    We examined an Adonis species occurring in Okayama Prefecture, Western Honshu, Japan. After studying morphological characters and as-certaining of the chromosome numbers, it was determined as A. multiflora Nishikawa and Koji Ito by having hollow scape, sepals shorter than the petals and the chromosome number of 2n=16. Adonis multiflora is newly reported from Okayama Pefecture, and the Adonis plants hith-erto reported from the limestone areas in Chu-goku District(Atetsu Region)are considered to belong to this species. A karyomorphological study of Adonis multi-flora collected from Okayama and Hiroshima Prefectures was carried out. They proved to be diploid with 2n=16 chromosomes, and eight chro-mosome pairs were recognized by configuration of chromosomes : four pairs with median chro-mosomes and remaining four pairs with subme-dian and/or subterminal chromosomes. Among the submedian and/or subterminal chromosomes, two pairs had satellites on the short arms. This is the first report for the karyotype under the name of A. multiflora, and the karyotype is nearly the same as that of materials with 2n=16 underthenameofA. amurensis Regel and Radde by Suda and Herai(1991)

    Neurogenesis Drives Stimulus Decorrelation in a Model of the Olfactory Bulb

    The reshaping and decorrelation of similar activity patterns by neuronal networks can enhance their discriminability, storage, and retrieval. How can such networks learn to decorrelate new complex patterns, as they arise in the olfactory system? Using a computational network model for the dominant neural populations of the olfactory bulb we show that fundamental aspects of the adult neurogenesis observed in the olfactory bulb -- the persistent addition of new inhibitory granule cells to the network, their activity-dependent survival, and the reciprocal character of their synapses with the principal mitral cells -- are sufficient to restructure the network and to alter its encoding of odor stimuli adaptively so as to reduce the correlations between the bulbar representations of similar stimuli. The decorrelation is quite robust with respect to various types of perturbations of the reciprocity. The model parsimoniously captures the experimentally observed role of neurogenesis in perceptual learning and the enhanced response of young granule cells to novel stimuli. Moreover, it makes specific predictions for the type of odor enrichment that should be effective in enhancing the ability of animals to discriminate similar odor mixtures

    An Autopsy Case of Anaplastic Pancreatic Ductal Carcinoma (Spindle Cell Type) Multiple Onset in the Pancreas

    This is a case of a 75-year-old man who was diagnosed with anaplastic pancreatic ductal carcinoma (spindle cell type). His image findings showed pancreatic head cysts and pancreatic head, body, and tail tumors respectively. EUS-FNA was performed to the pancreatic head and pancreatic body tumors, and the same high atypical type cells suspected of cancer were obtained from either specimen, and finally total pancreatectomy was performed. On the specimen, there were 4 lesions in the pancreas; histology showed that the same anaplastic pancreatic ductal carcinoma (spindle cell type) was obtained from the pancreatic head cyst and the pancreatic tumors
