    CEREBROSIDE GALACTOSIDASE: A METHOD FOR DETERMINATION AND A COMPARISON WITH OTHER LYSOSOMAL ENZYMES IN DEVELOPING RAT BRAIN 1

    (1) A method is described for assaying brain for cerebroside galactosidase activity. The enzyme was liberated by sonication and addition of sodium taurocholate, then by digestion with pancreatic enzymes. It was further purified by precipitation at pH 3. The enzyme was then incubated with an emulsion of galactose-labelled cerebroside in taurocholate and oleate at pH 4·5, and the liberated galactose was determined by scintillation counting. (2) The content of cerebroside galactosidase in rat brain at various ages has been determined. The enzyme was present before cerebroside appears in noticeable amounts (4 days) and the amount rose considerably during the period of active cerebroside deposition and myelination. The amount then remained at a high concentration even in the adult. (3) Comparison with other lysosomal brain enzymes was made in the age study. Nitrophenyl galactoside hydrolase also increased during myelination but levelled off earlier; its activity paralleled the amount of ganglioside. Nitrophenyl glucoside hydrolase started at a lower level and decreased with age. Sulphatase activity rose during myelination, then decreased somewhat after 15 days. Ceramidase followed a pattern similar to that of nitrophenyl galactoside hydrolase; it is suggested that both of these enzymes reflect ganglioside metabolism. (4) The relative amounts of brain enzymes in different states were determined as a function of age in the case of cerebrosidase, nitrophenyl galactoside hydrolase and sulphatase. The proportion found in the high speed supernatant fraction was low but increased after myelination. The proportion that could be ‘solubilized’ by sonication decreased after myelination but the values differed greatly for the three enzymes.     The household economic burden of eating disorders and adherence to treatment in Australia

    © 2014 Gatt et al.; licensee BioMed Central Ltd. Background: This study investigated the household economic burden of eating disorders and cost-related non-adherence to treatment in Australia. Methods: Multi-centre prospective observational study using a structured questionnaire. Ninety participants were recruited from two clinic settings in New South Wales, Australia and from the community using social media. The primary outcome measures were household economic burden of illness measured in terms of out-of-pocket expenditure, household economic hardship and cost-related non-adherence. Results: The pattern of out-of-pocket expenditure varied by diagnosis, with Bulimia Nervosa associated with the highest total mean expenditure (per three months). Economic hardship was reported in 96.7% of participants and 17.8% reported cost-related non-adherence. Those most likely to report cost-related non-adherence had a longer time since diagnosis. Cost-related non-adherence and higher out-of-pocket expenditure were associated with poorer quality of life, a more threatening perception of the impact of the illness and poor self-reported health. Conclusions: This study is the first to empirically and quantitatively examine the household economic burden of eating disorders from the patient perspective. Results indicate that households experience a substantial burden associated with the treatment and management of an eating disorder. This burden may contribute to maintaining the illness for those who experience cost-related non-adherence and by negatively influencing health outcomes. Current initiatives to implement sustainable and integrated models of care for eating disorders should strive to minimise the economic impact of treatment on families

    Rethinking symbolic and visual context in referring expression generation

    Situational context is crucial for linguistic reference to visible objects, since the same description can refer unambiguously to an object in one context but be ambiguous or misleading in others. This also applies to Referring Expression Generation (REG), where the production of identifying descriptions is always dependent on a given context. Research in REG has long represented visual domains through symbolic information about objects and their properties, to determine identifying sets of target features during content determination. In recent years, research in visual REG has turned to neural modeling and recasted the REG task as an inherently multimodal problem, looking at more natural settings such as generating descriptions for objects in photographs. Characterizing the precise ways in which context influences generation is challenging in both paradigms, as context is notoriously lacking precise definitions and categorization. In multimodal settings, however, these problems are further exacerbated by the increased complexity and low-level representation of perceptual inputs. The main goal of this article is to provide a systematic review of the types and functions of visual context across various approaches to REG so far and to argue for integrating and extending different perspectives on visual context that currently co-exist in research on REG. By analyzing the ways in which symbolic REG integrates context in rule-based approaches, we derive a set of categories of contextual integration, including the distinction between positive and negative semantic forces exerted by context during reference generation. Using this as a framework, we show that so far existing work in visual REG has considered only some of the ways in which visual context can facilitate end-to-end reference generation. Connecting with preceding research in related areas, as possible directions for future research, we highlight some additional ways in which contextual integration can be incorporated into REG and other multimodal generation tasks

    Coherent vs incoherent pairing in 2D systems near magnetic instability

    We study the superconductivity in 2D fermionic systems near antiferromagnetic instability, assuming that the pairing is mediated by spin fluctuations. This pairing involves fully incoherent fermions and diffusive spin excitations. We show that the competition between fermionic incoherence and strong pairing interaction yields the pairing instability temperature TinsT_{ins} which increases and saturates as the magnetic correlation length ξ→∞\xi \to \infty. We argue that in this quantum-critical regime the pairing problem is qualitatively different from the BCS one.Comment: 7 pages, 2 figure

    Drosophila Klp67A is required for proper chromosome congression and segregation during meiosis I

    Drosophila Klp67A belongs to the Kip3 subfamily of Kinesin-type microtubule catastrophe factors. In primary spermatocytes, loss of klp67A leads to defects in karyokinesis and cytokinesis. We show that these cells formed disorganised, bipolar spindles that contained increased numbers of microtubules. The kinetochore fibres were wavy and bent, whereas astral microtubules appeared abnormally robust and formed cortical bundles. Time-lapse studies revealed that during biorientation, the chromosomes in klp67A mutant cells continued to reorient for about twice as long as those in control cells. Metaphase plates were poorly defined in the mutants and often formed at non-equatorial positions. Consistent with the above abnormalities in chromosome congression, we found that in wild-type cells Klp67A associated with prometaphase/metaphase kinetochores before redistributing to the central spindle at anaphase onset. Although the timing of this redistribution of kinetochores argues against a role in anaphase chromosome segregation, dyads in the mutants disjoined but exhibited greatly diminished poleward velocities. They travelled on average at approximately 34% of the velocity of their wild-type counterparts and often decondensed at non-polar locations.
