    Everyday Social Functioning in People with Autism Spectrum Disorder

    Positive social relationships are important for both our physical and psychological well-being, and depend on the ability to engage appropriately with other people across different situational contexts. Although qualitative and clinical reports indicate that people with autism spectrum disorder (ASD) struggle with their everyday interactions, there is a paucity of experimental work examining the nature and severity of their difficulties, especially those of higher-functioning adults. In the present thesis, university students with ASD and matched control participants completed a range of novel or pre-existing scenario-based tasks recreating some of the features and demands of common social situations. Overall, the findings showed that, compared to neurotypical people, individuals with ASD tended to generate less socially skilled strategies in their interactions. Although they often showed relatively intact awareness of many social and normative expectations underpinning everyday situations, their understanding of these appeared to be more limited. The results are interpreted in light of the main social and non-social theoretical models of ASD, and their implications are discussed. In particular, it is hoped that the findings will contribute to the advancement of the current body of evidence, by helping to bridge the gap between theoretical accounts of ASD and performance in the real world, and to guide the development of tailored intervention programmes for higher-functioning adults

    Going 'Above and Beyond': Are Those High in Autistic Traits Less Pro-social?

    Few studies have explored how the cognitive differences associated with autistic spectrum disorder translate into everyday social behaviour. This study investigated pro-social behaviour in students scoring high and low on the autism-spectrum quotient (AQ), using a novel scenario task: 'Above and Beyond'. Each scenario involved an opportunity to behave pro-socially, and thus required balancing the needs of a character against participants' own interests. High AQ participants both generated responses and selected courses of action that were less pro-social than those of the low AQ group. For actions of low pro-social value they gave higher self-satisfaction ratings; conversely, they gave lower self-satisfaction ratings for high pro-social actions. The implications for everyday functioning are considered for those with high autistic traits

    Using and reasoning about social strategies in autism spectrum disorder in everyday situations

    Although a substantial amount of previous work has been dedicated to the study of the possible theoretical mechanisms underpinning autism spectrum disorder (ASD), little research has examined the types of difficulties experienced by individuals in their everyday social functioning. University students with ASD and matched control participants performed the Social Strategy task. In this, they read a range of descriptions of social interactions, all ending with an awkward question asked by the story main character. The types of strategies they used to answer the awkward questions were examined. Compared to control participants, those with ASD used a more negative emotional tone in responding, generated more simple strategies including acquiescence or refusal, and fewer sophisticated strategies that considered all parties' perspectives. In a novel task condition, participants rank-ordered simple and sophisticated strategies, and gave justifications for their use. Rank-ordering did not differentiate the groups. Justifications given by those with ASD for simple strategies were more practical and less often character-based than those given by control participants; the groups did not differ in the justifications given for sophisticated strategies. The possible explanations of these findings and implications for informing current intervention programmes are considered

    Contribution of sleep to the repair of neuronal DNA double-strand breaks:evidence from flies and mice

    Exploration of a novel environment leads to neuronal DNA double-strand breaks (DSBs). These DSBs are generated by type 2 topoisomerase to relieve topological constrains that limit transcription of plasticity-related immediate early genes. If not promptly repaired, however, DSBs may lead to cell death. Since the induction of plasticity-related genes is higher in wake than in sleep, we asked whether it is specifically wake associated with synaptic plasticity that leads to DSBs, and whether sleep provides any selective advantage over wake in their repair. In flies and mice, we find that enriched wake, more than simply time spent awake, induces DSBs, and their repair in mice is delayed or prevented by subsequent wake. In both species the repair of irradiation-induced neuronal DSBs is also quicker during sleep, and mouse genes mediating the response to DNA damage are upregulated in sleep. Thus, sleep facilitates the repair of neuronal DSBs

    Decline in macrolide resistance rates among Streptococcus pyogenes causing pharyngitis in children isolated in Italy

    Macrolides are often used to treat group A streptococcus (GAS) infections, but their resistance rates reached high proportions worldwide. The aim of the present study was to give an update on the characteristics and contemporary prevalence of macrolide-resistant pharyngeal GAS in Central Italy. A total of 592 isolates causing pharyngitis in children were collected in the period 2012-2013. Clonality was assessed by emm typing and pulsed-field gel electrophoresis (PFGE) for all macrolide-resistant strains and for selected susceptible isolates. Genetic determinants of resistance were screened by polymerase chain reaction (PCR). Forty-four GAS were erythromycin-resistant (7.4 %). Among them, 52.3 % and 50 % were clindamycin- and tetracycline-resistant, respectively. erm(B)-positive isolates (52.3 %) expressed the constitutive cMLSB phenotype. mef(A) and its associated M phenotype were recorded in 40.9 % of the cases. The remaining erm(A)-positive isolates expressed the iMLSB phenotype. Seventeen tetracycline-resistant isolates carried tet(M) and five isolates carried tet(O). Twenty-five emm types were found among all strains, with the predominance of emm types 12, 89, 1, and 4. Eleven emm types and 12 PFGE clusters characterized macrolide-resistant strains, with almost two-thirds belonging to emm12, emm4, and emm11. Macrolide-susceptible and -resistant emm types 12, 89, 11, and 4 shared related PFGE profiles. There was a dramatic decline in macrolide resistance in Central Italy among pharyngeal GAS isolates in 2012-2013 when compared to previous studies from the same region (p < 0.05), although macrolide consumption remained stable over the past 15 years.     Great Expectations: The Role of Rules in Guiding Pro-social Behaviour in Groups with High Versus Low Autistic Traits

    Measuring autistic traits in the general population has proven sensitive for examining cognition. The present study extended this to pro-social behaviour, investigating the influence of expectations to help others. A novel task describing characters in need of help was administered to students scoring high versus low on the Autism-Spectrum Quotient. Scenarios had two variants, describing either a 'clear-cut' or 'ambiguous' social rule. Participants with high versus low autistic traits were less pro-social and sympathetic overall towards the characters. The groups' ratings of characters' expectations were comparable, but those with high autistic traits provided more rule-based rationales in the clear-cut condition. This pattern of relatively intact knowledge in the context of reduced pro-social behaviour has implications for social skill training programmes
