    Addressing road-river infrastructure gaps using a model-based approach

    The world's rivers are covered over and fragmented by road infrastructure. Road-river infrastructure result in many socio-environmental questions and documenting where different types occur is challenged by their sheer numbers. Equally, the United Nations has committed the next decade to ecosystem restoration, and decision makers across government, non-government, and private sectors require information about where different types of road-river infrastructure occur to guide management decisions that promote both transport and river system resilience. Field-based efforts alone cannot address data and information needs at relevant scales, such as across river basins, nations, or regions to guide road-river infrastructure remediation. As a first step toward overcoming these data needs in Great Britain, we constructed a georeferenced database of road-river infrastructure, validated a subset of locations, and used a Boosted Regression Tree model-based approach with environmental data to predict which infrastructure are bridges and culverts. We mapped 110,406 possible road-river infrastructure locations and were able to either validate or predict which of 110,194 locations were bridges (n=60,385) or culverts (n=49,809). Upstream drainage area had the greatest contribution to determining infrastructure type: when <10 km2 our model correctly predicted culverts 73% of the time but only 60% of the time for bridges. Road type and stream gradient also influenced model results. Our model-based approach is readily applied to other locations and contexts and can be used to inform decisions about management of smaller infrastructure that are frequently overlooked worldwide

    Convergent Evolution of Chromosomal Sex-Determining Regions in the Animal and Fungal Kingdoms

    Sexual identity is governed by sex chromosomes in plants and animals, and by mating type (MAT) loci in fungi. Comparative analysis of the MAT locus from a species cluster of the human fungal pathogen Cryptococcus revealed sequential evolutionary events that fashioned this large, highly unusual region. We hypothesize that MAT evolved via four main steps, beginning with acquisition of genes into two unlinked sex-determining regions, forming independent gene clusters that then fused via chromosomal translocation. A transitional tripolar intermediate state then converted to a bipolar system via gene conversion or recombination between the linked and unlinked sex-determining regions. MAT was subsequently subjected to intra- and interallelic gene conversion and inversions that suppress recombination. These events resemble those that shaped mammalian sex chromosomes, illustrating convergent evolution in sex-determining structures in the animal and fungal kingdoms

    Early Acid/Base and Electrolyte Changes in Permanent Middle Cerebral Artery Occlusion: Aged Male and Female Rats

    BACKGROUND: Early changes in acid/base and electrolyte concentrations could provide insights into the development of neuropathology at the onset of stroke. We evaluated associations between acid/base and electrolyte concentrations, and outcomes in permanent middle cerebral artery occlusion (pMCAO) model. METHODS: 18-month-old male and female Sprague-Dawley rats underwent pMCAO. Pre-, post- (7 min after occlusion), and at 72 hr of pMCAO venous blood samples provided pH, carbon dioxide, oxygen, glucose, hematocrit, hemoglobin, and electrolyte values of ionized calcium, potassium, and sodium. Multiple linear regression determined predictors of infarct and edema volumes from these values, Kaplan-Meier curve analyzed morality between males and females at 72 hr, and a Cox regression model was used to determine predictors for mortality. RESULTS: Analysis indicated significant differences in acid/base balance and electrolyte levels in aged rats not dependent on sex between the three time points in the pMCAO model. Changes in pH (from pre- to post and post- to 72 hr) and changes in sodium and ionized calcium (from post- to 72 hr) were predictors of infarct volume and edema volume, respectively. Cox Regression revealed there is a 3.25 times increased risk for mortality based on changes in bicarbonate (pre- to post-MCAO). CONCLUSIONS: These early venous blood changes in acid/base balance and electrolytes can be used to predict stroke outcomes in our rat model of stroke. This study provides potential biomarkers to be examined in the human condition that could provide profound prognostic tools for stroke patients

    Translational Evaluation of Acid/Base and Electrolyte Alterations in Rodent Model of Focal Ischemia

    BACKGROUND AND PURPOSE: Acid/base and electrolytes could provide clinically valuable information about cerebral infarct core and penumbra. We evaluated associations between acid/base and electrolyte changes and outcomes in 2 rat models of stroke, permanent, and transient middle cerebral artery occlusion. METHODS: Three-month old Sprague-Dawley rats underwent permanent or transient middle cerebral artery occlusion. Pre- and post-middle cerebral artery occlusion venous samples for permanent and transient models provided pH, carbon dioxide, oxygen, glucose, and electrolyte values of ionized calcium, potassium, and sodium. Multiple regression determined predictors of infarct volume from these values, and Kaplan-Meier curve analyzed morality between permanent and transient middle cerebral artery occlusion models. RESULTS: Analysis indicated significant differences in the blood gas and electrolytes between pre- to post-middle cerebral artery occlusion. A decrease in pH and sodium with increases in carbon dioxide, potassium, ionized calcium, and glucose changes were found in both middle cerebral artery occlusion models; while hematocrit and hemoglobin were significant in the transient model. pH and ionized calcium were predictors of infarct volume in the permanent model, as changes in pH and ionized calcium decreased, infarct volume increased. CONCLUSIONS: There are acute changes in acid/base balance and electrolytes during stroke in transient and permanent rodent models. Additionally, we found pH and ionized calcium changes predicted stroke volume in the permanent middle cerebral artery occlusion model. These preliminary findings are novel, and warrant further exploration in human conditions

The cost and ease of use of each method was also evaluated. S. aureus was recovered using all methods on both porous and non-porous surfaces.     A Strategy for Finding Near Earth Objects with the SDSS Telescope

    We present a detailed observational strategy for finding Near Earth Objects (NEOs) with the Sloan Digital Sky Survey (SDSS) telescope. We investigate strategies in normal, unbinned mode as well as binning the CCDs 2x2 or 3x3, which affects the sky coverage rate and the limiting apparent magnitude. We present results from 1 month, 3 year and 10 year simulations of such surveys. For each cadence and binning mode, we evaluate the possibility of achieving the Spaceguard goal of detecting 90% of 1 km NEOs (absolute magnitude H <= 18 for an albedo of 0.1). We find that an unbinned survey is most effective at detecting H <= 20 NEOs in our sample. However, a 3x3 binned survey reaches the Spaceguard Goal after only seven years of operation. As the proposed large survey telescopes (PanStarss; LSST) are at least 5-10 years from operation, an SDSS NEO survey could make a significant contribution to the detection and photometric characterization of the NEO population.Comment: Accepted by AJ -- 12 pages, 11 figure

    Antarctic penguin response to habitat change as Earth's troposphere reaches 2°C above preindustrial levels

The definitive version was published in Ecological Monographs 80 (2010): 49–66, doi:10.1890/08-2289.1.We assess the response of pack ice penguins, Emperor (Aptenodytes forsteri) and Adélie (Pygoscelis adeliae), to habitat variability and, then, by modeling habitat alterations, the qualitative changes to their populations, size and distribution, as Earth's average tropospheric temperature reaches 2°C above preindustrial levels (ca. 1860), the benchmark set by the European Union in efforts to reduce greenhouse gases. First, we assessed models used in the Intergovernmental Panel on Climate Change Fourth Assessment Report (AR4) on penguin performance duplicating existing conditions in the Southern Ocean. We chose four models appropriate for gauging changes to penguin habitat: GFDL-CM2.1, GFDL-CM2.0, MIROC3.2(hi-res), and MRI-CGCM2.3.2a. Second, we analyzed the composited model ENSEMBLE to estimate the point of 2°C warming (2025–2052) and the projected changes to sea ice coverage (extent, persistence, and concentration), sea ice thickness, wind speeds, precipitation, and air temperatures. Third, we considered studies of ancient colonies and sediment cores and some recent modeling, which indicate the (space/time) large/centennial-scale penguin response to habitat limits of all ice or no ice. Then we considered results of statistical modeling at the temporal interannual-decadal scale in regard to penguin response over a continuum of rather complex, meso- to large-scale habitat conditions, some of which have opposing and others interacting effects. The ENSEMBLE meso/decadal-scale output projects a marked narrowing of penguins' zoogeographic range at the 2°C point. Colonies north of 70° S are projected to decrease or disappear: 50% of Emperor colonies (40% of breeding population) and 75% of Adélie colonies (70% of breeding population), but limited growth might occur south of 73° S. Net change would result largely from positive responses to increase in polynya persistence at high latitudes, overcome by decreases in pack ice cover at lower latitudes and, particularly for Emperors, ice thickness. Adélie Penguins might colonize new breeding habitat where concentrated pack ice diverges and/or disintegrating ice shelves expose coastline. Limiting increase will be decreased persistence of pack ice north of the Antarctic Circle, as this species requires daylight in its wintering areas. Adélies would be affected negatively by increasing snowfall, predicted to increase in certain areas owing to intrusions of warm, moist marine air due to changes in the Polar Jet Stream.This project was funded by the World Wildlife Fund and the National Science Foundation, NSF grant OPP-0440643 (D.
