    Der gegenwärtige Wandel der deutschen Bestattungskultur als Widerspiegelung gesellschaftlicher Entwicklungstrends

    "Auf der Grundlage einer quantitativen Erhebung stellt dieser Beitrag das derzeitige Bild der Bestattungskultur in Deutschland dar und erklärt es als Abbild gesellschaftlicher Veränderungen. Befragt wurde eine Auswahl deutscher Bestattungsunternehmen. Aus methodentechnischen Gründen konnten nicht christliche Bestattungen, etwa muslimische, nicht untersucht werden. Es wird begründet, dass Formen und Wandel der Bestattungskultur 'soziale Konstruktionen' und somit ein soziologisches Thema sind. Die Vielfältigkeit der gegenwärtigen Formen und Rituale der Bestattungskultur spiegelt gesellschaftliche Entwicklungstrends wider. Voranschreitende Säkularisierung, Rationalisierung und Ökonomisierung gehen einher mit Prozessen der Prekarisierung, Enttraditionalisierung, Individualisierung, Ästhetisierung der Alltagswelt und der Herausbildung neuer Lebensstile und Milieus. Dies zeigt sich in einer neuen Vielfalt und Polarität der Bestattungskultur: Aufwandsreduzierte, minimalistische Formen, die aus kritisch-konservativer Sicht eine 'Entsorgungsmentalität' darstellen, stehen solchen gegenüber, die soziale Positionen und geschmackliche Dispositionen repräsentieren." Due to methodical reasons, burials other than Christian, such as Muslim burials, are not included in the study. The article argues that forms and changes of funerary culture are social constructions, and as such a sociological topic. Diverse forms and rituals of funerary culture reflect major current trends in social development: secularization, rationalization, economization accompanied by processes of casualization, detraditionalization, individualization, aestheticizing of everyday life, new lifestyles and milieus. These manifest themselves in a new diversity and polarity: minimalistic forms, which are criticized to represent an 'Entsorgungsmentalität', are being opposed by forms that perform social positions and aesthetic preferences."     Comprehensive analysis of beta-catenin target genes in colorectal carcinoma cell lines with deregulated Wnt/beta-catenin signaling

    Background: Deregulation of Wnt/beta-catenin signaling is a hallmark of the majority of sporadic forms of colorectal cancer and results in increased stability of the protein beta-catenin. beta-catenin is then shuttled into the nucleus where it activates the transcription of its target genes, including the proto-oncogenes MYC and CCND1 as well as the genes encoding the basic helix-loop-helix (bHLH) proteins ASCL2 and ITF-2B. To identify genes commonly regulated by beta-catenin in colorectal cancer cell lines, we analyzed beta-catenin target gene expression in two non-isogenic cell lines, DLD1 and SW480, using DNA microarrays and compared these genes to beta-catenin target genes published in the PubMed database and DNA microarray data presented in the Gene Expression Omnibus (GEO) database. Results: Treatment of DLD1 and SW480 cells with beta-catenin siRNA resulted in differential expression of 1501 and 2389 genes, respectively. 335 of these genes were regulated in the same direction in both cell lines. Comparison of these data with published beta-catenin target genes for the colon carcinoma cell line LS174T revealed 193 genes that are regulated similarly in all three cell lines. The overlapping gene set includes confirmed beta-catenin target genes like AXIN2, MYC, and ASCL2. We also identified 11 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways that are regulated similarly in DLD1 and SW480 cells and one pathway - the steroid biosynthesis pathway - was regulated in all three cell lines. Conclusions: Based on the large number of potential beta-catenin target genes found to be similarly regulated in DLD1, SW480 and LS174T cells as well as the large overlap with confirmed beta-catenin target genes, we conclude that DLD1 and SW480 colon carcinoma cell lines are suitable model systems to study Wnt/beta-catenin signaling and associated colorectal carcinogenesis. Furthermore, the confirmed and the newly identified potential beta-catenin target genes are useful starting points for further studies

    Comparative Investigations to Corrosion Fatigue of Al-Cu and Al-Mg-Si Alloys

    One of the serious problems encountered in the use of various materials in technology is the occurrence of fatigue phenomena as an undesirable material damage under cyclic mechanical load. For aluminium alloys this issue is of extremely high importance in case of their utilisation for aircraft purposes, e.g., where a very wide spectrum of frequencies occur. Moreover, the cyclic loading may be joined by the presence of specific electrolyte media. Therefore, the material candidates must be thoroughly examined in view of their sensitivity to fatigue as well as to corrosion fatigue. Usually, the Cu-containing alloy EN-AW 2024 T3 is applied besides 7075 T6 in Airbus aircrafts, but the weldable alloy 6013 T6 is considered to be a potential alternative. Referring to former investigations on the environmental sensitivity (ES) in the fatigue behaviour /1-6/ this paper brings up experimental findings as well as expanded considerations about damaging mechanisms and modelling. The situation with the alloy 6013 T6 is emphasized. The propagation of cracks under cyclic load in different environments, such as vacuum, air or aqueous media, is described by means of fracture mechanics. This enables discrimination in view of the influence of environmental factors and, hence, the participation of corrosion processes

    Ultrahydrophobe chitosanstabilisierte Composite-Schichten auf Aluminiumwerkstoffen

    Selbstreinigende, ultrahydrophobe Oberflächen lassen sich in der Technik vielfältig einsetzen. Das ultrahydrophobe Verhalten beruht einerseits auf einer Rauigkeit im μm-Bereich und andererseits auf der chemischen Zusammensetzung der Oberfläche. Durch den gegebenen Oberflächenaufbau sind derartige Materialien jedoch empfindlich gegen Verschleiß. In diesem Beitrag wird ein Schichtverbund bestehend aus Aluminiumoxid und zwei polymeren Komponenten vorgestellt. Die Aluminiumoxidschicht wird auf dem Wege der anodischen Oxidation erzeugt. Dieses seit langem bekannte Verfahren ermöglicht nicht nur die Oxidation der Aluminiumoberfläche, sondern gestattet es, auch, definierte Oberflächenprofile einzustellen. Durch den gezielten Einbau des hochmolekularen Polymers Chitosan in die mikroprofilierte Aluminiumoxidschicht wurde eine mechanische Stabilisierung der Schicht im Sinne eines anorganisch-organischen Composites erreicht. Außerdem dienten die Amino-Seitengruppen des Chitosans als reaktives Interface für die notwendige chemische Hydrophobierung und als Reaktionszentrum für Vernetzungen, wodurch eine weitere mechanische Stabilisierung bewirkt wird.     Efficacy of a Drug-Eluting Stent Versus Bare Metal Stents for Symptomatic Femoropopliteal Peripheral Artery Disease: Primary Results of the EMINENT Randomized Trial.

    BACKGROUND A clear patency benefit of a drug-eluting stent (DES) over a bare metal stent (BMS) for treating peripheral artery disease of the femoropopliteal segment has not been definitively demonstrated. The EMINENT study (Trial Comparing Eluvia Versus Bare Metal Stent in Treatment of Superficial Femoral and/or Proximal Popliteal Artery) was designed to evaluate the patency of the Eluvia DES (Boston Scientific, Marlborough, MA), a polymer-coated paclitaxel-eluting stent, compared with BMS for the treatment of femoropopliteal artery lesions. METHODS EMINENT is a prospective, randomized, controlled, multicenter European study with blinded participants and outcome assessment. Patients with symptomatic peripheral artery disease (Rutherford category 2, 3, or 4) of the native superficial femoral artery or proximal popliteal artery with stenosis ≥70%, vessel diameter of 4 to 6 mm, and total lesion length of 30 to 210 mm were randomly assigned 2:1 to treatment with DES or BMS. The primary effectiveness outcome was primary patency at 12 months, defined as independent core laboratory-assessed duplex ultrasound peak systolic velocity ratio ≤2.4 in the absence of clinically driven target lesion revascularization or surgical bypass of the target lesion. Primary sustained clinical improvement was a secondary outcome defined as a decrease in Rutherford classification of ≥1 categories compared with baseline without a repeat velocity ratio ≤2.4 in the absence of clinically driven target lesion revascularization. Health-related quality of life and walking function were assessed. RESULTS A total of 775 patients were randomly assigned to treatment with DES (n=508) or commercially available BMS (n=267). Baseline clinical, demographic, and lesion characteristics were similar between the study groups. Mean lesion length was 75.6±50.3 and 72.2±47.0 mm in the DES and BMS groups, respectively. The 12-month incidence of primary patency for DES treatment (83.2% [337 of 405]) was significantly greater than for BMS (74.3% [165 of 222]; P<0.01). Incidence of primary sustained clinical improvement was greater among patients treated with the DES than among those who received a BMS (83.0% versus 76.6%; P=0.045). The health-related quality of life dimensions of mobility and pain/discomfort improved for the majority of patients in both groups (for 66.4% and 53.6% of DES-treated and for 64.2% and 58.1% of BMS-treated patients, respectively) but did not differ significantly. At 12 months, no statistical difference was observed in all-cause mortality between patients treated with the DES or BMS (2.7% [13 of 474] versus 1.1% [3 of 263]; relative risk, 2.4 [95% CI, 0.69-8.36]; P=0.15). CONCLUSIONS By demonstrating superior 1-year primary patency, the results of the EMINENT randomized study support the benefit of using a polymer-based paclitaxel-eluting stent as a first-line stent-based intervention for patients with symptomatic peripheral artery disease attributable to femoropopliteal disease.     Immune responses in healthy adults elicited by a bivalent norovirus vaccine candidate composed of GI.4 and GII.4 VLPs without adjuvant

    The development of an efficacious vaccine against norovirus is of paramount importance given its potential to reduce the global burden of norovirus-associated morbidity and mortality. Here, we report a detailed immunological analysis of a phase I, double-blind, placebo-controlled clinical trial performed on 60 healthy adults, ages 18 to 40. Total serum immunoglobulin and serum IgA against vaccine strains and cross-reactive serum IgG against non-vaccine strains were measured by enzyme immunoassays, whereas cell-mediated immune responses were quantified using intracellular cytokine staining by flow cytometry. A significant increase in humoral and cellular responses, e.g., IgA and CD4+ polypositive T cells, was triggered by the GI.4 Chiba 407 (1987) and GII.4 Aomori 2 (2006) VLP-based norovirus vaccine candidate rNV-2v, which is formulated without adjuvant. No booster effect was observed after the second administration in the pre-exposed adult study population. Furthermore, a cross-reactive immune response was elicited, as shown by IgG titers against GI.3 (2002), GII.2 OC08154 (2008), GII.4 (1999), GII.4 Sydney (2012), GII.4 Washington (2018), GII.6 Maryland (2018), and GII.17 Kawasaki 308 (2015). Due to viral infection via mucosal gut tissue and the high variety of potentially relevant norovirus strains, a focus should be on IgA and cross-protective humoral and cell-mediated responses in the development of a broadly protective, multi-valent norovirus vaccine.Clinical trial registrationhttps://clinicaltrials.gov, identifier NCT05508178.     Superhydrophobic Aluminum Surfaces: Preparation Routes, Properties and Artificial Weathering Impact

    Among the materials that can be treated in order to impart superhydrophobic properties are many originally hydrophilic metals. For this, they must undergo a sequential treatment, including roughening and hydrophobic coating. This contribution presents various preparation routes along with various characterization methods, such as dynamic contact angle (DCA) measurements, scanning electron microscopy (SEM) and spectroscopic techniques (FT–IRRAS, XPS, EIS). Micro-rough surfaces of pure and alloyed aluminum were generated most easily by using a modifie Sulfuric Acid Anodization under Intensifie conditions (SAAi). This produces a micro-mountain-like oxide morphology with peak-to-valley heights of 2 μm and sub-μm roughness components. Additionally, micro-embossed and micro-blasted surfaces were investigated. These micro-roughened initial states were chemically modifie with a solution of a hydrophobic compound, such as the reactive f uoroalkylsilane PFATES, the reactive alkyl group containing polymer POMA, or the polymer Teflo ® AF. Alternatively, the chemical modificatio was made by a Hot Filament Chemical Vapor Deposition (HFCVD) of a PTFE layer. The latter can form a considerably higher thickness than the wet-deposited coatings, without detrimental leveling effects being observed in comparison with the original micro-rough surface. The inherent and controllable morphology of the PTFE layers represents an important feature. The impacts of a standardized artificia weathering (WTH) on the wetting behavior and the surface-chemical properties were studied and discussed in terms of possible damage mechanisms. A very high stability of the superhydrophobicity was observed for the f uorinated wet-deposited PFATES and Teflo ® AF coatings as well as for some of the PTFE layer variants, all on SAAi-pretreated substrates. Very good results were also obtained for specimens produced by appropriate mechanical roughening and PTFE coating

    New Phenotypes of Potato Co-induced by Mismatch Repair Deficiency and Somatic Hybridization

    As plants are sessile they need a very efficient system for repairing damage done by external or internal mutagens to their DNA. Mismatch repair (MMR) is one of the systems that maintain genome integrity and prevent homeologous recombination. In all eukaryotes mismatches are recognized by evolutionary conserved MSH proteins often acting as heterodimers, the constant component of which is MSH2. Changes affecting the function of MSH2 gene may induce a ‘mutator’ phenotype and microsatellite instability (MSI), as is demonstrated in MSH2 knock-out and silenced lines of Arabidopsis thaliana. The goal of this study was to screen for ‘mutator’ phenotypes in somatic hybrids between potato cvs. ‘Delikat’ and ‘Désirée’ and MMR deficient Solanum chacoense transformed using antisense (AS) or dominant negative mutant (DN) AtMSH2 genes. The results demonstrate that first generation fusion hybrids have a range of morphological abnormalities caused by uniparental MMR deficiency; these mutant phenotypes include: dwarf or gigantic plants; bushiness; curled, small, large or abnormal leaves; a deterioration in chloroplast structure; small deep-purple tubers and early dehiscent flowers. Forty percent of the viable somatic hybrids planted in a greenhouse, (10 out of 25 genotypes) had mutant phenotypes accompanied by MSI. The majority of the hybrids with ‘mutator’ phenotypes cultured on media containing kanamycin developed roots so sustaining the presence of selectable marker gene nptII, from the initial constructs. Here for the first time, MMR deficiency combined with somatic hybridization, are used to induce new phenotypes in plants, which supports the role of MMR deficiency in increasing introgressions between two related species

    Quick and Clean Cloning: A Ligation-Independent Cloning Strategy for Selective Cloning of Specific PCR Products from Non-Specific Mixes

    We have developed an efficient strategy for cloning of PCR products that contain an unknown region flanked by a known sequence. As with ligation-independent cloning, the strategy is based on homology between sequences present in both the vector and the insert. However, in contrast to ligation-independent cloning, the cloning vector has homology with only one of the two primers used for amplification of the insert. The other side of the linearized cloning vector has homology with a sequence present in the insert, but nested and non-overlapping with the gene-specific primer used for amplification. Since only specific products contain this sequence, but none of the non-specific products, only specific products can be cloned. Cloning is performed using a one-step reaction that only requires incubation for 10 minutes at room temperature in the presence of T4 DNA polymerase to generate single-stranded extensions at the ends of the vector and insert. The reaction mix is then directly transformed into E. coli where the annealed vector-insert complex is repaired and ligated. We have tested this method, which we call quick and clean cloning (QC cloning), for cloning of the variable regions of immunoglobulins expressed in non-Hodgkin lymphoma tumor samples. This method can also be applied to identify the flanking sequence of DNA elements such as T-DNA or transposon insertions, or be used for cloning of any PCR product with high specificity
