Investigation of the effects of temperature and ions on the interaction between ECG and BSA by the fluorescence quenching method
The effects of temperature and common ions on binding (-)-epicatechin gallate (ECG) to bovine serum albumin (BSA) are investigated. The binding constants (Ka) between ECG and BSA are 1.20 Ч 106 (17°C), 1.38 Ч 106 (27°C), and 5.69 x 106 L mol-1 (37°C), and the number of binding sites (n) were 1.14, 1.15, and 1.26, respectively. These results showed that the increasing temperature improves the stability of the ECG-BSA system, which results in a higher binding constant and the number of binding sites of the ECG-BSA system. The presence of Co2+ and Zn2+ ions decreased the binding constants (Ka) and the number of binding sites (n) of ECG-BSA complex. However, the presence of Cu2+ and Ni2+ increased the affinity of ECG for BSA largely. The positive ΔH and positive ΔS indicated that hydrophobic forces might play a major role in the binding between ECG and BSA
Breastfeeding in China: a review
This review aims to describe changes in breastfeeding and summarise the breastfeeding rates, duration and reasons of discontinuing 'any breastfeeding' or 'exclusive breastfeeding' in P.R. China. Breastfeeding rates in China fell during the 1970s when the use of breast milk substitutes became widespread, and reached the lowest point in the 1980s. As a result many efforts were introduced to promote breastfeeding. The breastfeeding rate in China started to increase in the 1990s, and since the mid-1990s 'any breastfeeding' rates in the majority of cities and provinces, including minority areas, have been above 80% at four months. But most cities and provinces did not reach the national target of 'exclusive breastfeeding' of 80%. The 'exclusive breastfeeding' rates in minority areas were relatively lower than comparable inland provinces. The mean duration of 'any breastfeeding' in the majority of cities or provinces was between seven and nine months. The common reasons for ceasing breastfeeding, or introducing water or other infant food before four months, were perceived breast milk insufficiency, mother going to work, maternal and child illness and breast problems. Incorrect traditional perceptions have a strong adverse influence on 'exclusive breastfeeding' in less developed areas or rural areas. China is a huge country, geographically and in population size, and there is considerable ethnic diversity. Therefore breastfeeding rates in different parts of China can vary considerably
A decade of change in breastfeeding in China's far north-west
BACKGROUND: There have been considerable changes in breastfeeding practices in China over the past forty years. However China is a very large country, and breastfeeding rates in different parts of China vary considerably. The objective of this paper is to identify and compare breastfeeding types and rates between 1994–1996 and 2003–2004 in Shihezi, Xinjiang Uygur Autonomous Region, PR China. METHODS: In 1994–1996, a study of breastfeeding (n = 2197) was undertaken in Shihezi, Xinjiang, PR China. A decade later in 2003–2004, a longitudinal study (n = 545) of infant feeding practices was undertaken in the same area. RESULTS: The 'any breastfeeding' rates at 1, 4 and 6 months were 94%, 82% and 78% respectively in the early 1990s. A decade later, breastfeeding at 1 month was lower, but rates at 4 and 6 months remained the same. In 2004 the 'full breastfeeding' rate at one month was significantly higher (57%) than a decade earlier (38%), but after 3 months there was a rapid decline. This reflected a shift in the way complementary foods are introduced: the initial introduction was later, but by a higher proportion of mothers. CONCLUSION: The rate of breastfeeding at one month is significantly lower in 2003–2004 when compared to 1994–1996. The 'full breastfeeding' rates were initially higher, but after 3 months were then lower. The Chinese national breastfeeding targets were not reached in either period of the study. These studies show the need to further promote full or exclusive breastfeeding and further longitudinal studies are necessary to provide the detailed knowledge about risk factors required for health promotion programs
The Volume of Hippocampal Subfields in Relation to Decline of Memory Recall Across the Adult Lifespan
Background: The hippocampus is an important limbic structure closely related to memory function. However, few studies have focused on the association between hippocampal subfields and age-related memory decline. We investigated the volume alterations of hippocampal subfields at different ages and assessed the correlations with Immediate and Delayed recall abilities.Materials and Methods: A total of 275 participants aged 20–89 years were classified into 4 groups: Young, 20–35 years; Middle-early, 36–50 years; Middle-late, 51–65 years; Old, 66–89 years. All data were acquired from the Dallas Lifespan Brain Study (DLBS). The volumes of hippocampal subfields were obtained using Freesurfer software. Analysis of covariance (ANCOVA) was performed to analyze alterations of subfield volumes among the 4 groups, and multiple comparisons between groups were performed using the Bonferroni method. Spearman correlation with false discovery rate correction was used to investigate the relationship between memory recall scores and hippocampal subfield volumes.Results: Apart from no significant difference in the left parasubiculum (P = 0.269) and a slight difference in the right parasubiculum (P = 0.022), the volumes of other hippocampal subfields were significantly different across the adult lifespan (P < 0.001). The hippocampal fissure volume was increased in the Old group, while volumes for other subfields decreased. In addition, Immediate recall scores were associated with volumes of the bilateral molecular layer, granule cell layer of the dentate gyrus (GC-DG), cornus ammonis (CA) 1, CA2/3, CA4, left fimbria and hippocampal amygdala transition area (HATA), and right fissure (P < 0.05). Delayed recall scores were associated with the bilateral molecular layer, GC-DG, CA2/3 and CA4; left tail, presubiculum, CA1, subiculum, fimbria and HATA (P < 0.05).Conclusion: The parasubiculum volume was not significantly different across the adult lifespan, while atrophy in dementia patients in some studies. Based on these findings, we speculate that volume changes in this region might be considered as a biomarker for dementia disorders. Additionally, several hippocampal subfield volumes were significantly associated with memory scores, further highlighting the key role of the hippocampus in age-related memory decline. These regions could be used to assess the risk of memory decline across the adult lifespan
Quantitative proteomics identification of phosphoglycerate mutase 1 as a novel therapeutic target in hepatocellular carcinoma
BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most common malignancies worldwide with poor prognosis due to resistance to conventional chemotherapy and limited efficacy of radiotherapy. There is an urgent need to develop novel biomarkers for early diagnosis, as well as to identify new drug targets for therapeutic interventions. PATIENTS AND METHODS: 54 paired HCC samples and 21 normal liver tissues were obtained from West China Hospital of Sichuan University. Informed consent was obtained from all the patients or their relatives prior to analysis, and the project was approved by the Institutional Ethics Committee of Sichuan University. Stable Isotope Labeling with Amino Acids in Cell Culture (SILAC)-based proteomics was employed to profile the differentially expressed proteins between a HepG2 human hepatoma cell line and an immortal hepatic cell line L02. Validation of PGAM1 expression was performed by semi-quantitative RT-PCR, immunoblot and immunohistochemistry using clinical samples. shRNA expressing plasmids specifically targeting PGAM1 were designed and constructed by GenePharma Corporation (Shanghai, China), and were utilized to silence expression of PGAM1 in vitro and in vivo. Cell proliferation was measured by a combination of colony formation assay and Ki67 staining. Apoptosis was examined by flow cytometry and TUNEL assay. RESULTS: A total of 63 dysregulated proteins were identified, including 51 up-regulated proteins, and 12 down-regulated proteins (over 2-fold, p < 0.01). Phosphoglycerate mutase 1 (PGAM1) was found markedly upregulated. Clinico-pathological analysis indicated that overexpression of PGAM1 was associated with 66.7% HCC, and strongly correlated with poor differentiation and decreased survival rates (p < 0.01). shRNAs-mediated repression of PGAM1 expression resulted in significant inhibition in liver cancer cell growth both in vitro and in vivo. CONCLUSION: Our studies suggested that PGAM1 plays an important role in hepatocarcinogenesis, and should be a potential diagnostic biomarker, as well as an attractive therapeutic target for hepatocellular carcinoma
CagA-positive Helicobacter pylori may promote and aggravate scrub typhus
Helicobacter pylori (H. pylori) infection may alter the host’s resistance to tsutsugamushi disease pathogens through the Th1 immune response, leading to potential synergistic pathogenic effects. A total of 117 scrub typhus cases at Beihai People’s Hospital and affiliated hospitals of Youjiang University for Nationalities and Medical Sciences were studied from January to December 2022, alongside 130 healthy individuals forming the control group. All participants underwent serum H. pylori antibody testing. The prevalence of H. pylori infection was significantly higher among scrub typhus patients (89.7%) compared to healthy individuals (54.6%) (p < 0.05). Moreover, type I H. pylori infection was notably more prevalent in scrub typhus cases (67.5%) compared to healthy individuals (30%) (p < 0.05). Multifactorial analysis demonstrated type I H. pylori infection as an independent risk factor for scrub typhus (adjusted odds ratio: 2.407, 95% confidence interval: 1.249–4.64, p = 0.009). Among scrub typhus patients with multiple organ damage, the prevalence of type I H. pylori infection was significantly higher (50.6%) than type II H. pylori infection (15.4%) (χ2 = 4.735, p = 0.030). These results highlight a higher incidence of H. pylori infection in scrub typhus patients compared to the healthy population. Additionally, type I H. pylori strain emerged as an independent risk factor for scrub typhus development. Moreover, individuals infected with type I H. pylori are more susceptible to multiple organ damage. These findings suggest a potential role of H. pylori carrying the CagA gene in promoting and exacerbating scrub typhus
Spatio-Temporal Evolution and Driving Factors of Ecosystem Service Value of Urban Agglomeration in Central Yunnan
Urbanization and human activity have recently resulted in land use/cover change (LUCC), which has had a detrimental effect on the biological environment, on keeping the ecosystem’s sustainable growth and on comprehending the ecosystem’s quality and changes over the past 20 years in the central Yunnan urban agglomeration. The equivalent factor method and hotspot analysis were used to analyze the spatio-temporal changes in land use and ecosystem service value (ESV) in the urban agglomerations of central Yunnan province, and the effects of land use change on ESV were then examined. This study is based on the grid data of land use in 2000, 2005, 2010, 2015, and 2020. Finally, Geodetector was used to investigate the possible causes of ESV. The results showed that: (1) The urban agglomerations in central Yunnan’s land-use structure and pattern clearly changed between 2000 and 2020, with continual declines in grassland, cultivated land, and woodland, and constant increases in construction land. There was significant growth in both speed and area. (2) The average ESV of the land decreased consistently, the hotspot areas shrank, and the cold-spot areas grew as the ecosystem service function declined and the total amount of ESV decreased by 1.517 billion Yuan. These events were mostly explained by an increase in construction land and a decrease in grassland, cultivated land, and woodland. (3) The synergistic effect of numerous factors is what causes the change in ESV in the urban agglomerations of central Yunnan. The key forces behind ESV change in the research area were land-use intensity, normalized difference vegetation index (NDVI), slope, and people density. The results can help decision makers establish policies for ecological conservation and land use
Research on Deep Defect Detection Method of Cable Lead Sealing Based on Improved Pulsed Eddy Current Excitation
In order to reduce power failures caused by lead sealing defects, it is necessary to carry out nondestructive testing of cable lead sealings. However, previous studies have focused on the detection of surface and near-surface defects of lead sealings. Thus, an improved pulsed eddy current detection (IPECD) method is introduced to detect the deep defects of cable lead sealings (with depths ranging from 6 to 12 mm), and the frequency range selection principle and the optimization method of initial phase angles of different frequency components of IPECD, used to maximize the peak value of the excitation signal, are first explained in detail. Then, the detection sensitivities of the deep defects before and after the optimization are compared and analyzed based on a simulation. Finally, using the IPECD method, experiments are conducted to study the effects of the defect depth on features of the lift-off point of intersection and the zero-crossing time, enhancing the foundation for the prediction or rapid detection of the depth of lead sealing defects
A Numerical Strategy for the Approximate Solution of the Nonlinear Time-Fractional Foam Drainage Equation
This study develops a numerical strategy for finding the approximate solution of the nonlinear foam drainage (NFD) equation with a time-fractional derivative. In this paper, we formulate the idea of the Laplace homotopy perturbation transform method (LHPTM) using Laplace transform and the homotopy perturbation method. This approach is free from the heavy calculation of integration and the convolution theorem for the recurrence relation and obtains the solution in the form of a series. Two-dimensional and three-dimensional graphical models are described at various fractional orders. This paper puts forward a practical application to indicate the performance of the proposed method and reveals that all the outputs are in excellent agreement with the exact solutions
