    Quality of hydro-alcoholic products used in Senegal: pilot study

    Antisepsis and disinfection have always played an important role in the fight against infectious diseases. The use of these products has been effective in breaking the chain of transmission of microorganisms. Today, with the advent of COVID-19, the main recommendations are, among other things, physical distance and the use of antiseptic products, including hydro-alcoholic products. In Senegal, with poor regulations on the acquisition and distribution of antiseptics and disinfectants, this situation has led to a proliferation of antiseptics and disinfectants on the national market. this work aimed to study the quality of hydroalcoholic products found in the trade. We evaluated ten samples of hydro-alcohol products collected during the month of March 2020. Physical and microbiological controls were performed at the National Drug Control Laboratory. The alcohol content of the samples ranged from 63 to 85% and were consistent with WHO recommendations except for samples E3 and E5. The pH values varied between 4.02 and 6.64 and the densities of the hydro-alcoholic gel samples ranged from 0.84 to 0.92 g/cm3; E2, E4, E5, E6 samples had densities greater than 0.89 g/cm3. The samples of hydro-alcoholic products all conformed to the sterility test and no microbiological contamination was observed. Antimicrobial activities of the hydro-alcoholic samples tested ranged from 58.3 to 100% with two samples showing no activity (E2 and E5). In view of the results of this study, it would be relevant to expand and deepen investigations by a significant increase in the number of samples and by carrying out as complete an analysis as possible

Nationally, the share of the population in poverty was reduced from 67.9% in 1994/95 to 57.1% in 2001/02, and finally 50.8% in 2005/06.     Étude in vitro de l’effet antifalcémiant des globules rouges et de l’activité antioxydante d’extraits de la poudre de racines de Maytenus senegalensis Lam (Celestraceae)

    La drépanocytose est une hémoglobinopathie largement répandue chez les populations africaines noires. Actuellement, très peu de traitements conventionnels existent sur le marché, d’où le recours à la médecine traditionnelle. Maytenus senegalensis est une plante prescrite par les tradipraticiens dans la prise en charge de la drépanocytose. L’activité antifalcémiante des extraits de la poudre de racines de Maytenus senegalensis a été étudiée dans ce travail. Après obtention de l’extrait hexanique par extraction à l’aide d’un Soxhlet, le marc est macéré dans de l’acétate d’éthyle puis dans du méthanol pour donner les extraits correspondants. L’activité des fractions a été évaluée sur des drépanocytes de type SS. Les activités antifalcémiantes les plus importantes ont été observées pour les fractions polaires du méthanol et de l’acétate d’éthyle avec des taux d’inversion de la falciformation respectivement de 77% et de 65% à 10 mg/mL. Les activités antioxydantes de M. senegalensis (CI50 = 0,195 ± 0,004 mg/mL) et de l’acide ascorbique (CI50 = 0,078 ± 0,002 mg/mL) ont été déterminées. Les pouvoirs antioxydants (PA) montrent que l’activité antiradicalaire de l’acide ascorbique (PA = 12,85) est 2,5 fois meilleure que celle de M. senegalensis (PA = 5,14). Les résultats de l’étude justifient l’utilisation traditionnelle des racines de Maytenus senegalensis dans la prise en charge de la drépanocytose.Mots clés : Hémoglobine, drépanocytose, Maytenus senegalensis, activité antifalcémiante, stress oxydant, radicaux libres, activité antioxydante

    Validation of an UV-Visible spectrophotometry assay method for the determination of chlorpheniramine maleate tablets without prior extraction

    Methods proposed by pharmacopeias to check the quality of chlorpheniramine maleate tablets are multi steps methods which involve extraction and present issues with repeatability. An alternative method is proposed with a sensitive, reliable, simple and rapid UV-VISIBLE spectrophotometry method developed for the determination without extraction of chlorpheniramine maleate in tablets. The method was validated using the accuracy profile approach with an accuracy ranging from 99.70 to 100.46%. Analysis was done using 0.25 mol/L sulfuric acid, distilled water, and raw material in a room at 23 °C for 25 to 30 minutes. Chlorpheniramine maleate concentration varied from 0.018 to 0.03 mg/mL. The method was found to be specific with the appearance of the corresponding maxima at 265 nm and a correlation coefficient (R2 ) of 0.9993. Limits of detection and quantification were respectively 1.39 × 10-4and 2.26 × 10-3 mg/mL. The tolerance limits for the different concentration levels (75, 100 and 125%) were respectively: 88.37; 92.74 and91.62% for lower limits and 111.03; 108.17 and 108.02% for upper limits. It was observed that the tolerance limits were within the limits of acceptability set at 20%. Consequently, the method was declared valid and reliable for routine analysis of chlorpheniramine maleate in tablets containing chlorphenamine as active principle.Keywords : Chlorpheniramine maleate, tablets, accuracy profile, assay method without extraction, validatio

    Etude physico-chimique et minéralogique comparative des attapulgites de Mbodiène (Sénégal) et de Mormoiron (France)

    La valorisation des ressources locales constitue un impératif afin d’atteindre des objectifs de développement durable. Cette étude a consisté à évaluer les caractéristiques physiques, chimiques et minéralogiques d’une argile provenant de Mbodiène (Sénégal), l’attapulgite, comparativement avec un médicament antidiarrhéique, constitué d’attapulgite, dénommé Actapulgite®. L’échantillon d’attapulgite brute a été purifié avec une solution de HNO3 1M. Une étude morphologique effectuée par visualisation à la Microscopie Electronique à Balayage a montré pour les deux argiles étudiées, une structure fibreuse caractéristique de la palygorskite, représentant le minéral dominant. L’analyse chimique par la spectrométrie à fluorescence X a donné pour l’attapulgite purifiée : 65,69% Si, 7,89% Al, 5,92% Fe, 17,90% Mg, 0,43% Ca et 0,93% K contre respectivement 63,98 % Si, 12,92% Al, 10,32% Fe, 6,20% Mg, 2,48% Ca et 2,25% K pour l’Actapulgite®. La diffraction aux rayons X a montré une présence de palygorskite, quartz et carbonates dans les échantillons étudiés. L’étude des isothermes d’adsorption a montré une surface spécifique nettement plus élevée pour l’attapulgite purifiée, 138,96 m2.g-1 contre 98,72 m2.g-1 pour l’Actapulgite®. Ces résultats ont montré que l’attapulgite de Mbodiène pourrait être utilisée comme antidiarrhéique orale après lavage à l’eau et séchage.© 2016 International Formulae Group. A sample of attapulgite from Mbodiène was purified with a 1M HNO3 solution to remove impurities including carbonates. A morphological study using Scanning Electron Microscopy showed for both types of clay studied, a fibrous structure characteristic of palygorskite which represent major mineral. Chemical analysis by XRF spectrometry gave for purified attapulgite: 65.69% Si, 7.89% Al, 5.92% Fe, 17.90% Mg, 0.43% Ca and 0.93% K against respectively 63.98% Si, 12.92% Al, 10.32% Fe, 6.20% Mg, 2.48% Ca and 2.25% K for Actapulgite®. X-ray diffraction showed the presence of palygorskite, quartz and carbonates for both samples studied. The study of adsorption isotherms showed a significantly higher surface area for purified attapulgite, 138.96 m2.g-1 versus 98.72 m2.g-1 for Actapulgite®.     Antibiotic susceptibility profile of Streptococcus pneumoniae isolated from acute respiratory infection in Dakar: a cross sectional study

    Streptococcus pneumoniae is a pathogen causing pneumonia, meningitis, otitis and bacteraemia. Nowadays, S. pneumoniae is developing antibacterial resistance, particularly for those with reduced susceptibility to penicillin. The objective of this study was to assess the susceptibility profile of S. pneumoniae strains isolated from acute respiratory infections (ARIs) in children younger than 5 years of age in Dakar, Senegal. S. pneumoniae strains were isolated from broncho-alveolar lavages (BALs), nasopharyngeal swabs, and middle ear secretion from children in the Paediatric Department of Abass Ndao University Teaching Hospital and Paediatric Department of Roi Baudouin Hospital in Dakar, Senegal. The strains were cultivated on Columbia agar supplemented with 5% of horse blood and gentamicin (6 mg/L). Antibiotic susceptibility testing was performed using E-test method. A total of 34 strains of S. pneumoniae were isolated and identified in this study, among them 7 strains (20.58%) showed penicillin-resistance. Antibiotics such as amoxicillin/clavulanic acid (MIC90=0.036 μg/mL), cefuroxim (MIC90=0.38 μg/mL), cefixim (MIC90=1.5 μg/mL), as well as macrolides (azithromycin MIC90=1.5 μg/mL, clarithromycin MIC90=0.125 μg/mL) and fluoroquinolone (levofloxacin MIC90=1 μg/mL, ofloxacin MIC90=2 μg/mL) were mostly active. However, all S. pneumoniae strains were resistant to sulfamethoxazole/trimethoprim (MIC90: 32 μg/mL). Except of S. pneumoniae strains penicillin-resistance or reduced susceptibility, most strains were susceptible to β-lactams antibiotics commonly used in ARI treatment. Continuous surveillance of antimicrobial resistance patterns of pneumococcus strains is still crucial for effective control of ARIs in children

    Les études chimiques et biologiques de deux Annonaceae récoltées au Sénégal et largement utilisées en médecine traditionnelle, Uvaria chamae (racines et graines) et Annona senegalensis (racines ont été effectuées. Vingt-sept acétogénines ont été isolées dont cinq de structure originale ainsi que deux alcaloïdes isoquinoleiques et un diterpène de type kaurane. Différentes techniques chromatographiques ont été mises en œuvre pour la purification et l'isolement des molécules : chromatographie sur colonne sur diffe rents supports. L'identification des molécules a été réalisée à l'aide de différentes méthodes spectroscopiques : résonnance magnétique nucléaire du proton et du carbone treize, spectrométrie de mase, infrarouge et ultraviolette.
