    An examination of academic outcomes for students who attend a school-based afterschool program

    While most research on the benefits of afterschool programs has shown positive behavioral outcomes, the results on academic outcomes have been mixed. This study focused on academic outcomes to further explore whether students who regularly attended a school-based afterschool program showed greater academic gains than students who did not attend. Previous research has shown mixed results in academic outcomes from afterschool programs depending on gender, program location and grade level; therefore, these variables were examined to evaluate where group differences may exist. Afterschool attendance was considered and narrowly defined to provide more understanding about dosage in afterschool outcomes research. Students in this study were in grades 3 through 8 during the 2008-2009 school year, and they attended two charter schools in Western Pennsylvania. The afterschool programs operated within the charter schools. This is a secondary data analysis, using data that were collected for a program evaluation of the afterschool programs. To measure academic gains, a difference score was calculated from students' pretest and posttest scores on the 4Sight reading and mathematics assessments. The design of this study was a quasi-experimental design comparing students who regularly attended the afterschool programs (50% or greater attendance) with a randomly selected comparison group from the same population of charter school students. An overall Analysis of Variance (ANOVA) on the difference scores showed that afterschool participants with regular attendance performed better than nonparticipants in mathematics but not in reading. Further ANOVAs on the mathematics difference scores found no differences in gain scores by gender. Elementary students showed greater mathematics gains than middle school students, and students who attended the afterschool program at School M showed greater mathematics gains than students at School H. The reading gain scores were also further analyzed with ANOVAs, and boys showed greater gains than girls, elementary students showed greater gains than middle school students, and students at School M showed greater gains than students at School H. No correlation was found between the number of days of afterschool attendance and reading or mathematics gain scores. Findings are related to future directions for afterschool research and implications for afterschool providers

    New immunolatex spheres: visual markers of antigens on lymphocytes for scanning electron microscopy

    New immunochemical reagents consisting of antibodies bound to small latex spheres were used as visual markers for the detection and localization of cell surface antigens by scanning electron microscopy. Cross-linked latex spheres of various sizes from 300 to 3,4000 Å in diameter were synthesized by aqueous emulsion copolymerization of methacrylate derivatives containing hydroxyl and carboxyl functional groups. Proteins and other molecules containing primary amino groups were covalently bonded to the acrylic spheres under a variety of mild conditions by the aqueous carbodiimide, cyanogen bromide, and glutaraldehyde methods. For use in the indirect immunochemical-labeling technique, goat antibodies directed against rabbit immunoglobulins were bonded to the spheres. These immunolatex reagents were shown to bind only to cells (red blood and lymphocytes) which had previously been sensitized with rabbit antibodies against cell surface antigens. Mouse spleen lymphocytes with exposed immunoglobulins on their surface (B cells) were labeled with these spheres and distinguished from unlabeled or T lymphocytes by scanning electron microscopy. The distribution of Ig receptors on lymphocytes was also studied using the spheres as visual markers. When lymphocytes were fixed with glutaraldehyde and subsequently labeled with the immunolatex reagents, a random distribution was observed by scanning electron microscopy; a patchy distribution was observed when unfixed lymphocytes were used. These results are consistent with studies using ferritin-labeled antibodies (S. De Petris and M. Raff. 1973. Nature [Lond.]. 241:257.) and support the view that Ig receptors on lymphocytes undergo translational diffusion. In addition to serving as visual markers for scanning electron microscopy, these latex spheres tagged with fluorescent or radioactive molecules have applications as highly sensitive markers for fluorescent microscopy and as reagents for quantitative studies of cell surface antigens and other receptors

    The effect of stellar limb darkening values on the accuracy of the planet radii derived from photometric transit observations

    We study how the precision of the exoplanet radius determination is affected by our present knowledge of limb darkening in two cases: when we fix the limb darkening coefficients and when we adjust them. We also investigate the effects of spots in one-colour photometry. We study the effect of limb darkening on the planetary radius determination both via analytical expressions and by numerical experiments. We also compare some of the existing limb darkening tables. When stellar spots affect the fit, we replace the limb darkening coefficients, calculated for the unspotted cases, with effective limb darkening coefficients to describe the effect of the spots. There are two important cases. (1) When one fixes the limb darkening values according to some theoretical predictions, the inconsistencies of the tables do not allow us to reach accuracy in the planetary radius of better than 1-10% (depending on the impact parameter) if the host star's surface effective temperature is higher than 5000 K. Below 5000 K the radius ratio determination may contain even 20% error. (2) When one allows adjustment of the limb darkening coefficients, the a/Rs ratio, the planet-to-stellar radius ratio, and the impact parameter can be determined with sufficient accuracy (<1%), if the signal-to-noise ratio is high enough. However, the presence of stellar spots and faculae can destroy the agreement between the limb darkening tables and the fitted limb darkening coefficients, but this does not affect the precision of the planet radius determination. We also find that it is necessary to fit the contamination factor, too. We conclude that the present inconsistencies of theoretical stellar limb darkening tables suggests one should not fix the limb darkening coefficients. When one allows them to be adjusted, then the planet radius, impact parameter, and the a/Rs can be obtained with the required precision.Comment: Astronomy & Astrophysics Vol. 549, A9 (2013) - 11 page

    A gas-liquid solid phase peptide and protein sequenator

    A new miniaturized protein and peptide sequenator has been constructed which uses gas phase reagents at the coupling and cleavage steps of the Edman degradation. The sample is embedded in a matrix of Polybrene dried onto a porous glass fiber disc located in a small cartridge-style reaction cell. The protein or peptide, though not covalently attached to the support, is essentially immobile throughout the degradative cycle, since only relatively apolar, liquid phase solvents pass through the cell. This instrument can give useful sequence data on as little as 5 pmol or protein, can perform extended sequence runs (greater than 30 residues) on subnanomole quantities of proteins purified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and can sequence hydrophobic peptides to completion. The sequenator is characterized by a high repetitive yield during the degradation, low reagent consumption, low maintenance requirements, and a degradative cycle time of only 50 min using a complete double cleavage program
