Influence of parameters of drying on laser induced diffuse reflectance of banana discs
Banana discs of 1 cm thickness were immersed
into different antioxidant solutions to slow down
potentially disturbing discoloration during drying. Samples
were randomly split into 8 groups according to the 2^p
experimental design. Two antioxidant solutions with
1.66% and 4.59% ascorbic acid, two levels of drying
temperature with 50°C and 80°C, two levels of drying time
with 6h and 8h were used or adjusted. Laser diodes of
seven wavelengths (532, 635, 650, 780, 808, 850 and 1064
nm) were selected to illuminate the surface and light
penetration pattern was evaluated on the basis of radial
profiles. Profiles acquired at three wavelengths (532, 635
and 650 nm) were found to respond sensitively to adjusted
parameters. As a result of drying, intensity decay was
observed to move closer to incident point. Significant
effect (p<0.01) of temperature, drying time and their
interaction was found on extracted descriptive attributes of
intensity profiles: full width at half maximum (FWHM),
distance of inflection point (DIP) and slope of logarithmic
decay (SLD). Beyond their presence, antioxidant
concentration was neutral factor without significant
contribution to the model. Results were in agreement with
reference spectroscopic measurements, especially with
NDVI index. Promising results suggest that evaluated
method might be suitable for monitoring purposes during
drying of fruits
Molecular cytogenetic characterisation of Salix viminalis L. using repetitive DNA sequences
Abstract Salix viminalis L. (2n=38) is a diploid dicot
species belonging to the Salix genus of the Salicaceae
family. This short-rotation woody crop is one of the most
important renewable bioenergy resources worldwide. In
breeding for high biomass productivity, limited knowledge
is available on the molecular cytogenetics of willow, which
could be combined with genetic linkage mapping. The present
paper describes the adaptation of a fluorescence in situ
hybridisation (FISH) protocol as a new approach to analyse
the genomic constitution of Salix viminalis using the heterologous
DNA clones pSc119.2, pTa71, pTa794, pAs1, Afafamily,
pAl1, HT100.3, ZCF1 and the GAA microsatellite
marker. Three of the nine probes showed unambiguous
signals on the metaphase chromosomes. FISH analysis with
the pTa71 probe detected one major 18S-5.8S-26S rDNA
locus on the short arm of one chromosome pair; however,
the pTa794 rDNA site was not visible. One chromosome
pair showed a distinct signal around the centromeric
region after FISH with the telomere-specific
DNA clone HT100.3. Two chromosome pairs were
found to have pAs1 FISH signals, which represent a
D-genome-specific insert from Aegilops tauschii. Based
on the FISH study, a set of chromosomes with characteristic
patterns is presented, which could be used to
establish the karyotype of willow species
GABA, glutamát és nitrogén monoxid tartalmú myentericus neuronok térbeli mintázatának ontogenetikus és pathológiás változása = Ontogenetical and pathological changes in the spatial pattern of GABA, glutamate and nitrogen oxide-containing myenteric neurons
Vizsgálataink elsőként bizonyították, hogy a krónikus alkoholkezelés bélszakasz-függő módon befolyásolja a nitrogén monoxid szintáz (NOS) enzim aktivitását, a NOS fehérjetartalmat és a nitrerg myentericus neuronok számát patkány bélidegrendszerében (Alcohol Clin Exp Res. 2006 30(6):967-73). A streptozotocin-indukálta diabeteszes patkányok bélcsatornájának különböző szakaszaiban megvizsgáltuk a diabetes által okozott motilitási problémák és a nitrerg myentericus neuronszám változás összefüggéseit. Bizonyítottuk azt is, hogy az inzulinkezelés megakadályozta a rendellenes bélperisztaltika és a nitrerg neuropathia kialalkulását. Ereményeink mutatták az első bizonyítékát annak, hogy a bélcsatorna különböző szakaszaiban a nitrerg neuronok eltérő mértékben érzékenyek a diabeteses állapotra és az inzulinkezelésre (Diabetes Res Clin Pract. 2008 Jan 31; [Epub ahead of print]). Munkánk során immunhisztokémia módszerekkel vizsgáltuk a VGLUT1-3-immunreaktivitás jelenlétét 14 és 23 hetes humán magzatok myentericus plexusában. Mindhárom vezikuláris glutamát transzporter egyedi térbeli és időbeli megoszlást mutatott. A VGLUT1-immunreaktivitás volt a legkifejezettebb az egész vizsgált periódus alatt (Histol Histopathol 2008 in press). | Our study has demonstrated for the first time that chronic ethanol consumption has a differential effect on nitric oxide synthase (NOS) activity, NOS protein content, and the number of nitrergic neurons in different intestinal segments, suggesting that chronic ethanol administration affects the nitric oxide pathways in the enteric nervous system (Alcohol Clin Exp Res. 2006 30:967-73). The streptozotocin-induced diabetic rat model was used to investigate the relation between the deranged gut motility and the segment-specific quantitative changes in the nitrergic myenteric neurons. Additionally, we studied the effectiveness of early insulin replacement to prevent the diabetes-induced changes. These findings comprise the first evidence that the nitrergic neurons located in different intestinal segments exhibit different susceptibilities to a diabetic state and to insulin treatment (Diabetes Res Clin Pract. 2008 Jan 31; [Epub ahead of print]). Immunocytochemistry was applied to examine the distribution of the immunoreactivity of all three VGLUTs during prenatal development of the myenteric plexus in the human small intestine. We also investigated changes in their localization in the different segments of the small intestine and in the different compartments of the developing myenteric ganglia. Each transporter displayed a characteristic spatiotemporal expression pattern, depending on the fetal age, the gut segment or the ganglionic compartment (Histol Histopathol 2008 in press)

Az óragének és a környezeti fényviszonyok szerepe az N-acetiltranszferáz génexpressziójának szabályozásában csirke retinában = The role of clock-genes and environmental lighting conditions in the regulation of N-acetyltransferase gene expression in the chicken retina
Csirke retina és tobozmirigy sejtekben a melatonin (MT) szintézis kulcsenzimének, az arilalkilamin N-acetiltranszferáznak génje (cAanat) co-expresszálódik az óragénekkel. Emellett a dopamin (DA) a fő katekolamin a retinában, mely neuroregulatorként hat. Tanulmányoztuk: 1. a cBmal1 és cAanat mRNS expressziók dinamikáját retinális/pineális sejtekben in vivo és in vitro RT-PCR-ral. A cBmal1 mRNS csúcsa a fényes fázisban volt Zeitgeber idő (ZT) 8 és 10 között, megelőzve az a cAanat expresszió éjszakai emelkedését ZT 16-17-nál. A retinális/pineális cBmal1 és cAanat mRNS ritmusainak fázisai arra utalnak, hogy kapcsolat van az expressziók között; 2. locked nucleinsav tartalmú cBmal1 antisense oligonukleotidok (AON) hatását a cAanat transzkriptumokra és az MT termelésre pinealocita kultúrában, melyet szuperfúziós rendszerben transzfektáltunk. Az AON, mely vagy az RNáz H-t aktiválta, vagy a transzlációs masinériához való kötődést blokkolta szignifikánsan csökkentette a cAanat transzkripcióját és az MT szekrécióját jelezvén a cBmal1 kulcsszerepét az indol metabolizmus szabályozásában; 3. két fő DA-receptor család (D1 és D2) szerepét a circadián retinális MT ritmus szabályozásában eye-cup modellen. A D2 agonista Quinpirol szignifikánsan csökkentette az MT szekrécióját sötét fázisban, míg a D1 agonista SKF hatástalan volt, amely arra utal, hogy a DA gátló hatást fejt ki a retinális MT szekrécióra (reciprok negativ feed-back), melyet elsősorban a D2 receptorok mediálnak. | In chicken retinal and pineal cells gene of arylalkylamine N-acetyltransferase (cAanat), the key enzyme of melatonin (MT) synthesis is co-expressed with clock genes. In addition, dopamine (DA) is the major catecholamine in the retina serving as a neuroregulator. We studied: 1. the temporal profile of cBmal1 and cAanat mRNA expressions in retinal/pineal cells in vivo and in vitro using RT-PCR. cBmal1 mRNA had a peak in the light phase between Zeitgeber time (ZT) 8 and 10, preceding the nocturnal increase in cAanat expression at ZT 16 to 17. The phase of the rhythms of retinal/pineal cBmal1 and cAanat mRNAs suggests a link between their expressions; 2. the effect of cBmal1 antisense oligonucleotides (AONs) containing locked nucleic acid on cAanat transcripts and MT production in cultured pinealocytes transfected in superfusion system. These AONs synthesized for activating RNase H or blocking the binding of the translation machinery were able to reduce significantly cAanat transcription and MT secretion indicating the key role of cBmal1 in the regulation of indole metabolism; 3. the role of the two major DA-receptor families (D1 and D2) in the regulation of circadian retinal MT rhythm on an eye-cup model. Quinpirole (D2 agonist) reduced significantly MT secretion in dark phase, whereas SKF (D1 agonist) was ineffective suggesting that DA exerts an inhibitory effect on retinal MT secretion (reciprocal negative feed-back), which is mediated primarily via D2 receptors

PACAP and PAC1 Receptor Expression in Human Insulinomas
Pituitary adenylate cyclase activating polypeptide (PACAP) is a neuropeptide with widespread occurrence and diverse functions. PACAP binds to specific PAC1 and non-specific VPAC1/2 receptors. PACAP is considered as a growth factor, as it plays important roles during development and participates in reparative processes. Highest concentrations are found in the nervous system and endocrine glands, where several functions are known, including actions in tissue growth, differentiation and tumour development. Therefore, we have investigated expression of PACAP and its receptors in different tumours, including those of endocrine glands. We showed earlier that PACAP and PAC1 receptor staining intensity decreased in pancreatic ductal adenocarcinoma. In the present study we aimed to investigate alterations of PACAP and PAC1 receptor in human insulinoma and compared the immunostaining pattern with samples from chronic pancreatitis patients. We collected perioperative and histological data of patients who underwent operation because of insulinoma or chronic pancreatitis over a five-year-long period. Histology showed chronic pancreatitis with severe scar formation in pancreatitis patients, while tumour samples evidenced Grade 1 or 2 insulinoma. PACAP and PAC1 receptor expression was studied using immunohistochemistry. Staining intensity was very strong in the Langerhans islets of normal tissue and discernible staining was also observed in the exocrine pancreas. Immunostaining intensity for both PACAP and PAC1 receptor was markedly weaker in insulinoma samples, and disappeared from chronic pancreatitis samples except for intact islets. These findings show that PAC1 receptor/PACAP signalling is altered in insulinoma and this suggests a possible involvement of this system in tumour growth or differentiation
Nyelőcső-perforatio miatt végzett oesophagusexstirpatio rekonstrukciója hypopharyngogastrostoma képzésével
A sebészi és az intenzív terápia jelentős fejlődése ellenére a nyelőcső-perforatio napjainkban is súlyos, életet veszélyeztető állapot. A háttérben álló kiváltó okok, a kísérő betegségek, a lokalizáció és a kialakuló gyulladás eltérő mértéke miatt néha váratlan szituációval találkozik a sebész. Az 58 éves nőbetegnél a gyomor mellűri inkarcerált herniája miatt kialakult oesophagusperforatio és a következményes nekrotizáló mediastinitis miatt végeztünk oesophagusexstirpatiót, cervicalis oesophagostomiát alakítottunk ki. A tápcsatorna rekonstrukciója során a nyelőcsőcsonk ismeretlen eredetű, teljes hosszát érintő „zsugorodását” találtuk. A csőgyomrot a hypopharynxra anasztomizáltuk, a kialakult insufficientia konzervatív terápiára szanálódott. A beteg komplex nyelésterápia után visszanyerte részleges nyelési képességét. A nem tumoros alapbetegség miatt képzett hypopharyngogastrostoma irodalmi ritkaság. Kényszerhelyzetben, mint amilyen az esetünk is volt, választható műtéti megoldás. A beavatkozást azonban teammunkán alapuló rehabilitációnak kell követnie, amelynek során kiemelt szerepe van a nyelési terápiának
