These compounds have been designed by combining a kynurenamine framework with an amidine moiety in order to improve selectivity for the inducible isoform. In general, the in vitro inhibitory assays exhibited better inhibition values on the iNOS isoform, being the N-(3-(2-amino-5-methoxyphenyl)-3-hydroxypropyl)-4-(trifluoromethyl)benzimidamide 4i the most active inhibitor with the highest iNOS selectivity, without inhibiting eNOS. Docking studies on the two most active compounds suggest a different binding mode on both isozymes, supporting the experimentally observed selectivity towards the inducible isoform. Bioactive imidamide-based compounds targeted against nitric oxide synthase
The selective inhibition of inducible nitric oxide synthase (iNOS) has become an interesting goal for the treatment of diseases where the immune and inflammatory response of the organism is involved. Septic shock is one prominent example of this type of affections. In this paper, the design and synthesis of twelve substituted pyridinyl- imidamide derivatives is described, together with their biological evaluation as NOS inhibitors. The most potent and selective compound was N-(3-hydroxy-3-(pyridin-3-yl)propyl)acetimidamide 9a (IC50 = 4.6 µM, against iNOS). Pharmacological assays in aortic rat tissue, have confirmed its inhibitory activity on iNOS and the absence of undesired cardicovascular effects. In silico analysis of the most promising compounds (9a, 9b, 9e and 9g) have predicted good drug-likeness properties. Furthermore, they have shown an adequate cell viability. Docking studies carried out on 9a suggest a particular binding mode that involves the essential residue Glu377, and might explain its iNOS selectivity. 4,5-DIHYDRO-1H-PYRAZOLE as nNOS selective inhibitors: synthesis and structure-activity relationship
INTRODUCCIÓN
La oxido nítrico sintasa (NOS) es la enzima que cataliza la biosíntesis de óxido nítrico (NO) a partir de
L-arginina.1 Hasta el momento, se han descubierto cuatro isoformas:2 nNOS, iNOS, eNOS y mtNOS.
El NO es un biomensajero relacionado con importantes funciones fisiológicas.3Sin embargo, se ha
demostrado que una sobreproducción de NO por la nNOS está implicada en procesos
neurodegenerativos.4 Este hecho justifica la necesidad terapéutica de encontrar inhibidores selectivos
de la nNOS que permitan luchar contra enfermedades tales como Alzheimer, Parkinson, esclerosis
lateral amiotrófica y corea de Huntington.
Nuestro grupo de investigación ha sintetizado y evaluado una serie de derivados kinurenínicos5 1 y
kinurenamínicos6 2 como agentes neuroprotectores que resultaron desprovistos de actividad
inhibitoria frente a la enzima kinurenina-3-hidroxilasa (KYN3OH), lo que demuestra que su actividad
neuroprotectora se debe a la inhibición de la nNOS.OBJETIVO
Basándonos en estos antecedentes, hemos sintetizado y realizado la evaluación biológica in vitro
frente a las isoformas nNOS e iNOS de una serie de derivados de 4,5-dihidro-1H-pirazol con
estructura general 3, con objeto de encontrar inhibidores selectivos de alguna de estas isoformas.
METODOLOGÍA
Tomando como referencia los derivados kinurenínicos y kinurenamínicos, hemos sintetizado los
análogos rígidos con un resto de 4,5-dihidro-1H-pirazol. Además de la restricción conformacional, se
han llevado a cabo otras modificaciones, como la introducción de distintos sustituyentes en el anillo
aromático y la modificación del grupo acilo en el anillo de pirazolina.
CONCLUSIÓN /DISCUSIÓN
Todos los compuestos ensayados inhiben nNOS. La inhibición de iNOS es ínfima en la mayoría de los
casos, por lo que se pueden considerar selectivos, y no hay inhibición de KYN3OH. Por consiguiente,
el potencial neuroprotector de estos derivados se debe únicamente a la inhibición de nNOS.INTRODUCTION
Nitric Oxide Synthase (NOS) is the enzyme which catalyses the biosynthesis of Nitric Oxide (NO)
from L-arginine1. Four NOS isoforms have been described:2 nNOS, iNOS, eNOS and mtNOS. NO is
a biological messenger involved in several physiologic processes.3 However, an overproduction of
NO by nNOS produces neurotoxicity which has been associated with various neurological disorders4.
Therefore, it is necessary to found nNOS inhibitors to fight pathologies such as Alzheimer’s disease,
Parkinson, amyotrophic lateral sclerosis and Huntington’s disease.
In previous papers, our research group have described the synthesis of a series of kynurenine5 1 and
kynurenamine6 2 derivatives as neuroprotective agents which are not active versus kynurenine-3-
hydroxylase (KYN3OH). This fact demonstrates that their neuroprotective activity is only due to the
nNOS inhibition.OBJECTIVE
Basing on these precedents, we have developed and evaluated in vivo, versus nNOS and iNOS, a
series of 4,5-dihydro-1H-pyrazole derivatives with general structure 3 in order to find new selective
compounds.
METHODOLOGY
Taking kynurenine and kynurenamine derivatives as reference, we have synthesized rigid analogous
with a ring of 4,5-dihydro-1H-pyrazole . Besides the conformacional restriction, other modifications
have been carried out, as the introduction of different substituents in the aromatic ring and the
modification of the acyl group in the pyrazoline ring .
CONCLUSION /DISCUSION
All compounds inhibit nNOS. In most of cases, the inhibition of iNOS is negligible. Thus, they can be
considered selective. On the other hand, there is no KYN3OH inhibition. Consequently, the
neuroprotective potential of these derivatives is due only to the inhibition of nNOS

Thiadiazoline- and Pyrazoline-Based Carboxamides and Carbothioamides: Synthesis and Inhibition against Nitric Oxide Synthase
Two new families of pyrazoline and thiadiazoline heterocycles have been developed. Their inhibitory activities against two different isoforms of nitric oxide synthase (inducible and neuronal NOS) are reported. The novel derivatives were synthesized combining the arylthiadiazoline or arylpyrazoline skeleton and a carboxamide or carbothioamide moiety, used as starting material ethyl 2-nitrobenzoates or substituted nitrobenzaldehydes, respectively. The structure-activity relationships of final molecules are discussed in terms of the R1 radical effects in the aromatic ring, the Y atom in the heterocyclic system, the X heteroatom in the main chain, and the R2 substituent in the carboxamide or carbothioamide rest. In general, thiadiazolines (5a–e) inhibit preferentially the neuronal isoform; among them, 5a is the best nNOS inhibitor (74.11% at 1 mM, IC50 = 420 μM). In contrast, pyrazolines (6a–r) behave better as iNOS than nNOS inhibitors, 6m being the best molecule of this series (76.86% at 1 mM of iNOS inhibition, IC50 = 130 μM) and the most potent of all tested compounds.This work was partially supported by the Instituto de Salud

Utilización y aplicación de la herramienta "google docs" en la docencia universitaria dentro del marco del EEES
Como consecuencia de la realización del curso 'Aplicaciones de la Web 2.0 en la Investigación y la Docencia' organizado por La Fundación General Universidad de Granada-Empresa en el curso académico 2009-2010, los autores del presente trabajo presentamos el Proyecto de Innovación Docente concedido por el Vicerrectorado para la Garantía de la calidad de la Universidad de Granada,titulado 'Utilización de la herramienta “Google Docs” en la Docencia Universitaria dentro del marco del EEES. Creemos que el proyecto tiene un diseño aplicable a cualquier asignatura de la actual Licenciatura y/o Grado de Farmacia. En este trabajo se expone la experiencia piloto llevada a cabo en la asignatura de Química Farmacéutica, incluyendo el desarrollo, objetivos, metodología, resultados y las conclusiones que se están obteniendo en el trascurso del mism

Este proyecto ha sido puesto en marcha por un equipo de profesores con la intención de acercar a los niños y niñas al mundo de la lectura. Este objetivo es algo que se ha estado persiguiendo durante varios años y, observando que el alumnado estaba cada vez más entusiasmado, se decidió convertir esas intenciones en un proyecto innovador. El descubrimiento de la música puede resultar muy divertido no sólo para los niños y niñas sino también para los padres y maestros, por lo que se decidió que la música y, concretamente, Mozart (aprovechando su 250 aniversario) sería la motivación para que todos se acercasen a la biblioteca a ver qué encontraban. No sólo podemos escuchar música sino también leerla, conocer quién la escribe y para quién y, lo que es más importante, todos podemos escribir música para que los demás la lean y la escuchen tal y como han hecho muchos músicos a lo largo de la historia. Las nuevas tecnologías también son un elemento importante no sólo de consulta (Internet) sino también de agente motivador para el objetivo que se propuso.Junta de Andalucía.
