Effectively Integrating Technology to Engage Students and Meet Learning Objectives in Language Arts Classrooms
Technology is an ever-changing resource that is more advanced now than it has ever been. This action research project explored how teachers intentionally plan the use of technology in their classroom based on the objectives that need to be taught. The main study that this action research explores is how teacher align objectives to integrate intentional technology that engages students in the middle school ELA classroom? By collecting data via questioning, interviewing, observing and collaboration among participants, the findings show that the use of technology in the classroom is most efficient when implementing a backwards design approach to lesson planning

Media literacy's role in the mitigation of disinformation effects on substance misuse
Objective: Misinformation and substance use both increased substantially during the COVID-19 pandemic. This study examined potential links between misinformation beliefs and substance use among adults, along with the potential for media literacy to mitigate misinformation's influences on problematic use of widely available substances of misuse.Method: Structural equation modeling (SEM) was used to test a theoretical model of media literacy's effects on substance use, fully mediated by disinformation beliefs, with a nationally representative sample of U.S. adults recruited through a Qualtrics panel of adults using census-based quotas for geographic region, population density, ethnic diversity and gender (N = 1264). The sample was 51.5% male (N = 651); 46.7% female (N = 591); 1.1% nonbinary (N = 13); and 0.7% (N = 9) not reporting.Results: Media literacy for source of news positively associated with media literacy for content of news (b = 0.814, p < 0.001). Media literacy for content of news then positively associated with science media literacy (b = 0.192, p < 0.001). Science media literacy then negatively associated with disinformation beliefs (b = -0.586, p < 0.001), and COVID-19 disinformation beliefs associated with an increase in substance use (b = 0.466, p < 0.001). Disinformation beliefs also associated with alcohol and sleep medication co-use (odds = 1.956, p < 0.05).Conclusions: Results demonstrate media literacy's value for substance misuse prevention and effective public health messaging
Personalized rTMS for Depression: A Review
Personalized treatments are gaining momentum across all fields of medicine.
Precision medicine can be applied to neuromodulatory techniques, where focused
brain stimulation treatments such as repetitive transcranial magnetic
stimulation (rTMS) are used to modulate brain circuits and alleviate clinical
symptoms. rTMS is well-tolerated and clinically effective for
treatment-resistant depression (TRD) and other neuropsychiatric disorders.
However, despite its wide stimulation parameter space (location, angle,
pattern, frequency, and intensity can be adjusted), rTMS is currently applied
in a one-size-fits-all manner, potentially contributing to its suboptimal
clinical response (~50%). In this review, we examine components of rTMS that
can be optimized to account for inter-individual variability in neural function
and anatomy. We discuss current treatment options for TRD, the neural
mechanisms thought to underlie treatment, differences in FDA-cleared devices,
targeting strategies, stimulation parameter selection, and adaptive closed-loop
rTMS to improve treatment outcomes. We suggest that better understanding of the
wide and modifiable parameter space of rTMS will greatly improve clinical
outcome
Parabacteroides distasonis:intriguing aerotolerant gut anaerobe with emerging antimicrobial resistance and pathogenic and probiotic roles in human health
Parabacteroides distasonis is the type strain for the genus Parabacteroides, a group of gram-negative anaerobic bacteria that commonly colonize the gastrointestinal tract of numerous species. First isolated in the 1930s from a clinical specimen as Bacteroides distasonis, the strain was re-classified to form the new genus Parabacteroides in 2006. Currently, the genus consists of 15 species, 10 of which are listed as 'validly named' (P. acidifaciens, P. chartae, P. chinchillae, P. chongii, P. distasonis, P. faecis, P. goldsteinii, P. gordonii, P. johnsonii, and P. merdae) and 5 'not validly named' (P. bouchesdurhonensis, P. massiliensis, P. pacaensis, P. provencensis, and P. timonensis) by the List of Prokaryotic names with Standing in Nomenclature. The Parabacteroides genus has been associated with reports of both beneficial and pathogenic effects in human health. Herein, we review the literature on the history, ecology, diseases, antimicrobial resistance, and genetics of this bacterium, illustrating the effects of P. distasonis on human and animal health
AIDS virus–specific CD8+ T lymphocytes against an immunodominant cryptic epitope select for viral escape
Cryptic major histocompatibility complex class I epitopes have been detected in several pathogens, but their importance in the immune response to AIDS viruses remains unknown. Here, we show that Mamu-B*17+ simian immunodeficiency virus (SIV)mac239-infected rhesus macaques that spontaneously controlled viral replication consistently made strong CD8+ T lymphocyte (CD8-TL) responses against a cryptic epitope, RHLAFKCLW (cRW9). Importantly, cRW9-specific CD8-TL selected for viral variation in vivo and effectively suppressed SIV replication in vitro, suggesting that they might play a key role in the SIV-specific response. The discovery of an immunodominant CD8-TL response in elite controller macaques against a cryptic epitope suggests that the AIDS virus–specific cellular immune response is likely far more complex than is generally assumed
An Investigation of Abstract Construal on Impression Formation: A Multi-Lab Replication of McCarthy and Skowronski (2011)
Perceivers often view individuals described as “warm” to be generally positive and individuals described as “cold” to be generally negative. Consistent with the tenets of Construal Level Theory, McCarthy and Skowronski (2011) demonstrated this difference was larger among perceivers who were instructed the information was psychologically distant rather than psychologically near; however, those results have never been subjected to replication attempts. To test the replicability of those results, we closely replicated the methods of McCarthy and Skowronski (2011) Study 1b at eight separate data collection sites and pooled the results into a random-effects meta-analysis. Within the replication attempts, the overall effect was not significantly different from zero (d = 0.10, 95% CI [–0.01, 0.22]) and an equivalence test confirmed this effect was smaller than our smallest effect size of interest. However, when the original study was incorporated into the meta-analysis, the overall effect was significantly different from zero in the theoretically-consistent direction (d = 0.13, 95% CI [0.02, 0.24]). The weight of the overall evidence suggests the traits “warm” and “cold” are more influential among participants who were presented with information that was psychologically distant; however, this effect is small. Future research should try to identify more potent moderators, which would make the effect more affordable to detect

Buffy coat specimens remain viable as a DNA source for highly multiplexed genome-wide genetic tests after long term storage
<p>Abstract</p> <p>Background</p> <p>Blood specimen collection at an early study visit is often included in observational studies or clinical trials for analysis of secondary outcome biomarkers. A common protocol is to store buffy coat specimens for future DNA isolation and these may remain in frozen storage for many years. It is uncertain if the DNA remains suitable for modern genome wide association (GWA) genotyping.</p> <p>Methods</p> <p>We isolated DNA from 120 Action to Control Cardiovascular Risk in Diabetes (ACCORD) clinical trial buffy coats sampling a range of storage times up to 9 years and other factors that could influence DNA yield. We performed TaqMan SNP and GWA genotyping to test whether the DNA retained integrity for high quality genetic analysis.</p> <p>Results</p> <p>We tested two QIAGEN automated protocols for DNA isolation, preferring the Compromised Blood Protocol despite similar yields. We isolated DNA from all 120 specimens (yield range 1.1-312 ug per 8.5 ml ACD tube of whole blood) with only 3/120 samples yielding < 10 ug DNA. Age of participant at blood draw was negatively associated with yield (mean change -2.1 ug/year). DNA quality was very good based on gel electrophoresis QC, TaqMan genotyping of 6 SNPs (genotyping no-call rate 1.1% in 702 genotypes), and excellent quality GWA genotyping data (maximum per sample genotype missing rate 0.64%).</p> <p>Conclusions</p> <p>When collected as a long term clinical trial or biobank specimen for DNA, buffy coats can be stored for up to 9 years in a -80degC frozen state and still produce high yields of DNA suitable for GWA analysis and other genetic testing.</p> <p>Trial Registration</p> <p>The Action to Control Cardiovascular Risk in Diabetes (ACCORD) trial is registered with ClinicalTrials.gov, number <a href="http://www.clinicaltrials.gov/ct2/show/NCT00000620">NCT00000620</a>.</p
Synergistic roles of climate warming and human occupation in Patagonian megafaunal extinctions during the Last Deglaciation
The causes of Late Pleistocene megafaunal extinctions (60,000 to 11,650 years ago, hereafter 60 to 11.65 ka) remain contentious, with major phases coinciding with both human arrival and climate change around the world. The Americas provide a unique opportunity to disentangle these factors as human colonization took place over a narrow time frame (~15 to 14.6 ka) but during contrasting temperature trends across each continent. Unfortunately, limited data sets in South America have so far precluded detailed comparison. We analyze genetic and radiocarbon data from 89 and 71 Patagonian megafaunal bones, respectively, more than doubling the high-quality Pleistocene megafaunal radiocarbon data sets from the region.We identify a narrowmegafaunal extinction phase 12,280 ± 110 years ago, some 1 to 3 thousand years after initial human presence in the area. Although humans arrived immediately prior to a cold phase, the Antarctic Cold Reversal stadial, megafaunal extinctions did not occur until the stadial finished and the subsequent warming phase commenced some 1 to 3 thousand years later. The increased resolution provided by the Patagonian material reveals that the sequence of climate and extinction events in North and South America were temporally inverted, but in both cases, megafaunal extinctions did not occur until human presence and climate warming coincided. Overall, metapopulation processes involving subpopulation connectivity on a continental scale appear to have been critical for megafaunal species survival of both climate change and human impacts.Fil: Metcalf, Jessica L..
