    The stress-responsive kinase DYRK2 activates heat shock factor 1 promoting resistance to proteotoxic stress

    To survive proteotoxic stress, cancer cells activate the proteotoxic-stress response pathway, which is controlled by the transcription factor heat shock factor 1 (HSF1). This pathway supports cancer initiation, cancer progression and chemoresistance and thus is an attractive therapeutic target. As developing inhibitors against transcriptional regulators, such as HSF1 is challenging, the identification and targeting of upstream regulators of HSF1 present a tractable alternative strategy. Here we demonstrate that in triple-negative breast cancer (TNBC) cells, the dual specificity tyrosine-regulated kinase 2 (DYRK2) phosphorylates HSF1, promoting its nuclear stability and transcriptional activity. DYRK2 depletion reduces HSF1 activity and sensitises TNBC cells to proteotoxic stress. Importantly, in tumours from TNBC patients, DYRK2 levels positively correlate with active HSF1 and associates with poor prognosis, suggesting that DYRK2 could be promoting TNBC. These findings identify DYRK2 as a key modulator of the HSF1 transcriptional programme and a potential therapeutic target

    Most breast cancer patients die due to metastases, and the early onset of this multistep process is usually missed by current tumor staging modalities. Therefore, ultrasensitive techniques have been developed to enable the enrichment, detection, isolation and characterization of disseminated tumor cells in bone marrow and circulating tumor cells in the peripheral blood of cancer patients. There is increasing evidence that the presence of these cells is associated with an unfavorable prognosis related to metastatic progression in the bone and other organs.     Qual è il ruolo che i sistemi territoriali a vocazione rurale possono svolgere nel perseguimento della sostenibilità del benessere? Partendo dalle riflessioni sul concetto di sostenibilità e di benessere, nate nell’ambito delle attività di studio condotte nella Fondazione Simone Cesaretti, in questo volume gli autori cercano di rispondere a tale domanda, offrendo una costante rilettura della prospettiva territoriale e delle determinanti della sua sostenibilità. Affinché i sistemi territoriali a vocazione rurale possano fornire il loro importante contributo nel perseguimento della sostenibilità territoriale ed, attraverso quest’ultima, al complessivo raggiungimento della sostenibilità dello sviluppo, è necessario impostare un nuovo importante percorso innovativo, stavolta non solo investendo sulle tradizionali leve di sviluppo settoriale ma puntando sempre più a vettori di sviluppo economico, sociale, ambientale e culturale che sintetizzino un forte orientamento alla sostenibilità. Raccogliendo i contributi provenienti da diverse realtà territoriali ed evidenziando le relative esperienze su specifici strumenti e sull’adozione di determinate politiche, il volume propone una visione del territorio come punto di partenza fondamentale per l’impostazione di piani per la sostenibilità, facendo emergere la caratterizzazione di ciascun contesto e la necessità di adeguare gli strumenti non solo agli obiettivi ma soprattutto alle connotazioni identitarie dei relativi territori

    The circular economy refers to a development strategy that allows economic growth by optimising the use of natural resources, minimising environmental pressures, transforming supply chains and consumption patterns and redesigning production systems so that they are restorative or regenerative by intention and design. A transition to a more circular economy requires a systemic approach that considers the interconnections that exist within and between sectors, institutions and local actors.     The abuse of alcoholic beverages (beer, spirits and wine), also included in Agenda 2030 as one of the limits to the sustainability of well-being, results in considerable social and economic repercussions due not only to the increase in health spending but above all because of Of the loss of productivity arising in recent years especially from the youth component of human capital. Although the health benefit of a moderate consumption of wine is widely studied, the damage caused by its abuse is not so well known, and because the communication strategies implemented by the companies tend to make full use of only the positive aspect. From this point of view, starting from the health effects of wine consumption (both in case of abuse and in moderation) the paper provides the design of a web communication strategy to promote for Young people a culture of drinking in a sustainable way
