[์˜๋ฌธ]In mice, dental mesenchymal cells comprise the dental papilla and dental follicle in the developing mammalian tooth bud. It is thought that dental follicle cells have the ability to differentiate into fibroblasts, cementoblasts, and osteoblasts. However, cellular differentiation and the effects of environmental factors are not known exactly. In this study, the expression of BSP and OPN, using immunohistochemical markers for hard tissue, was not detected at E14 dental follicle, and positive BSP and OPN reactions were observed in the alveolar bone area at PN8. Despite the lack of hard tissue formation in the E14 dental follicles, dental follicle cells showed the potential to form hard tissue in the following experiment. In order to characterize this potential of dental follicle cells in vitro, E14 dental follicle cells were separated into single cells, then cultured. We assessed the expression of both Runx2 and Bsp in the aggregated cells, and determined to the ability of the cells to differentiate into osteoblasts and their potential to form bone. Furthermore, after the addition of bone inducers into the cultured dental follicle cells, including developing calvaria and BMP4, Runx2 and Bsp expressed in the aggregated cells showed the effects of environmental factors on differentiation into osteoblasts. The decrease in the number of school-age students leds to a decrease in the number of classes and faculties eventually creating unused spaces in schools. Small-scale consolidation of schools took place in areas with a sharp decline in the school-age population. The statistics of Korea's future population predicts the number of school-age students will fall by more than 30 percent in 2047 compared to 2017. Under these circumstances, various projects have been carried out according to the interest of the school's unused space. The Ministry of Culture, Sports and Tourism conducted the "Create Happy Schools with Culture" project for 51 schools across the country from 2008 to 2013 under the project to improve unused space in schools. In addition, the Ministry of Educations project, innovating spaces of school 2019 is on going for 300 schools. There are various prior studies on how to utilize unused space in schools as a way to reverse the unused or urban decline of the educational infrastructure. However, we believe that the unused educational infrastructure has worked together with external environment factors that cannot be determined by internal factors alone, and in this study we look at the phenomena from an urban perspective beyond short-term solutions. In this study, the target sites with different physical environments were selected, focusing on schools, to identify the pattern of unused educational infrastructure according to the characteristics of the surrounding areas. Therefore, Seongnam City, which can see changes in the new town and the old town at a glance due to the creation of the first and second new towns, was designated as the primary target area, and the number of students dropped sharply for elementary schools within the range of living areas, or as of 2019, was divided into the new and old towns, and three target areas were selected according to the slightly different surrounding characteristics of the school and the surrounding infrastructure was analyzed. According to the analysis, there was a difference between the ratio of the school-age population and the elderly population, but around the school, the distribution of commercial facilities and households was slightly different from one destination to another. The causal relationship could not be clarified, but the educational infrastructure and the surrounding environment could be seen as being influenced by each other. In addition, it was confirmed that undersized schools were also created in new city schools, where the population was steadily flowing in, which was a break in the formula of overcrowding in the new town. The decrease in the school-age population is a common problem faced by schools, but the analysis of the study showed that unused educational infrastructure had different peripheral characteristics for different locations. Unused infrastructure does not end as a viscous component, but rather as a chain of occurrences in the surrounding area. Unused educational infrastructure can also be attributed to the damage of workers or parents, including faculty members of the school concerned, and the decline of the school can also affect the surrounding facilities and areas.     Rodents have a toothless diastema between the incisor and the first molar, which may contain rudimentary tooth germs. In the lower diastema region of mice at E13, the rudimentary tooth germs, which developed into the bud stage before its removal by apoptosis, was found. The immunoreactivity to tenascin was observed in the condensed mesenchyme around the normal tooth bud and was detected in only the basement membrane in the diastema bud. This result shows that the relationship between mesenchymal condensation and tooth development. The similar patterns of Msx-1 and Msx-2 expression between the tooth bud and the diastema bud show that the diastema bud may have some other genetic mechanism in the developmental arrest of the rudimentary tooth germs rather than the Msx-1 and Msx-2 expression. Strikingly, the induction of the tooth formation was possible using tissue recombination between the oral epithelium of the diastema bud and the dental mesenchyme of the molar tooth bud, which indicates the potential capability of the diastema in the tooth formation.     Dental follicle cells in the periodontium are known to have the ability to differentiate into fibroblasts, cementoblasts, and osteoblasts during mouse periodontal development. From embryonic day 14 (E14) to postnatal day 11 (PN11), histological observations showed dramatic alterations in the relative width of the periodontal ligament (PDL)-forming region between the alveolar bone-forming and tooth root-forming area. At PN2, the width of the PDL-forming region showed a minimum, but with a higher expression of NOGGIN and proliferation cell nuclear antigen than the other regions. At PN11, the relative width of the PDL-forming region had expanded. Transplantation of individual regions of the developing tooth germ under the kidney renal capsule showed that dental follicle cells at E14 possessed the potential to develop into mineralized tissue after 3 weeks. These results suggested that the recovery of PDL width at PN11 may have resulted from cell proliferation and molecular interactions between osteogenic factors and their antagonists, such as interactions between bone morphogenetic protein 4 (BMP4) and NOGGIN, simlilar to those observed in suture, limb, and somite formation. To confirm the molecular interaction between BMP4 and NOGGIN, NOGGIN-protein bead implantation onto cultures was employed in vitro. This study thus indicates that harmonious interactions between NOGGIN and BMP in PDL-forming cells, which show higher cell proliferation than neighboring cells, might be important for proper periodontium development.ope
