The migratory behavior of the osteoblast-like cells with oligopeptides derived from fibronectin according to the application method

학위논문 (석사)-- 서울대학교 대학원 : 치의학과, 2012. 2. 구영.목적: 최근까지 진행되고 있는 합성 올리고펩타이드에 대한 연구의 대부분은 합성 올리고펩타이드를 적용할 때, 용액(solution)으로 적용하는 방법보다는 운반체(carrier)에 부착하여 적용하는 방법을 사용하고 있다. 이 논문의 목적은 FN(Fibronectin, 파이브로넥틴) 유래 올리고펩타이드를 합성 후, 이들을 운반체에 부착하여 적용하는 방법과 용액으로 적용하는 방법에 따른 조골양세포의 이동 양상을 비교하는 것이다. 방법: FN 유래 합성 올리고펩타이드를 제작한 후, 이 펩타이드를 운반체에 부착하여 적용하는 방법과 용액으로 적용하는 방법으로 조골양세포에 각각 적용하였다. 0, 6, 12, 18시간 후 각 방법에 따른 세포를 고정, 염색하여 이동한 세포 수와 이동 면적을 통해 조골양세포의 이동 양상을 관찰하였다. 결과: FN 유래 합성 올리고펩타이드를 운반체에 부착하여 적용하는 방법과 용액으로 적용하는 방법을 비교 관찰했을 때, 조골양세포의 이동 수는 12시간에서 이동 면적은 12, 18시간에서 통계적으로 유의한 차이가 존재하였다(p<0.05). 이러한 결과로 볼 때, 운반체에 부착하여 적용하는 방법에 비해 용액으로 적용하는 방법이 초기 상처 치유 시 세포의 이동을 보다 촉진시킬 것으로 생각된다.Objective Almost all the researches on synthetic oligopeptides had been carried out by the carrier-related method (pre-coating to the well before cell seeding) rather than the liquid-related method (releasing after cell seeding) until now. The purpose of this study was to evaluate the migratory behaviors of the osteoblast-like cells in each applied method. Materials and method Synthetic oligopeptides derived from fibronectin were applied to the cell before and after seeding cells. The cells were fixed and stained each for 0, 6, 12 and 18 hours. After that, we investigated the cells migratory behaviors by counting the number of the cells showing migratory behaviors, and calculating the space which these cells occupy. Results As with the liquid-related method in comparison with the carrier-related method, the number of the cells was highly enhanced at 12 hours and the space which these cells occupy was highly enhanced at 12, 18 hours. Within the limit of this study, the fibrin-related oligopeptides in the liquid-related method may play a significant role in the wound healing process during the early period by promoting the cells migratory behaviors.Maste
