    Autophagy Inhibition by Chloroquine Sensitizes Cervical Cancer SiHa Cells to CPT Treatment

    自噬诱导是肿瘤细胞对化疗药物抵抗性的原因之一,该研究探讨溶酶体抑制剂氯喹对喜树碱(camptothecin,CPT)诱导的宫颈癌细胞Si Ha死亡的增敏效果。CPT和/或氯喹处理宫颈癌Si Ha细胞,MTT法检测细胞增殖,DAPI和TUNEL染色观察细胞凋亡,Western blot和免疫荧光检测自噬及凋亡相关蛋白。结果发现,CPT处理后,Si Ha细胞MAP1LC3B荧光点和LC3II(microtubuleassociated protein light chain 3II)蛋白水平增加,p62荧光点和蛋白质水平则减少;而采用氯喹特异抑制自噬后,可明显提高CPT诱导的细胞凋亡、caspase-9的激活和PARP(poly ADP-ribose polymerase)的切割,而全长caspase-2水平显著下降。以上结果提示,氯喹可通过抑制细胞自噬而增强宫颈癌细胞株Si Ha对CPT诱导细胞凋亡的敏感性。The autophagy induction is one of the reasons for the resistance of tumor cells to chemotherapy drugs. In this study, the enhanced sensitivity of cervical cancer Si Ha cells to camptothecin(CPT)-induced cell death by chloroquine(a lysosome inhibitor) was investigated. The cell viability was detected by MTT assay, meanwhile, apoptosis was observed by DAPI and TUNEL, autophagy related proteins and apoptosis proteins were analyzed by immunofluorescence(IF) staining and Western blot in Si Ha cells after CPT treatment alone or combined with chloroquine. The results found that in Si Ha cells with CPT treatment the autophagy related protein LC3 foci and microtubule-associated protein light chain 3II(LC3II) protein level was increased, but p62 foci and protein level was decreased. When autophagy was inhibited by chloroquine, the CPT-induced apoptosis was obviously enhanced, and caspase-9 was activated and PARP was cleaved, but full length caspase-2 was decreased.     Intratumoral Delivery of a PD-1-blocking scFv encoded in Oncolytic HSV-1 Promotes Antitumor Immunity and Synergizes with TIGIT Blockade

    恶性肿瘤已严重威胁人类健康和生命,现有的治疗手段远远未能满足临床需求。厦门大学研究团队联合浙江养生堂生物科技有限公司、养生堂有限公司进行协同攻关,研制出新一代肿瘤免疫治疗药物——“注射用重组人PD-1抗体单纯疱疹病毒”。研究发现,重组表达PD-1单链抗体的溶瘤病毒具有“双药合一”抗肿瘤的突出优势,提高肿瘤治愈率。相关成果于2020年3月3日以研究论文形式在线发表于Cancer Immunology Research(《癌症免疫学研究》)期刊,为指导新型溶瘤病毒的升级改造和突破肿瘤免疫耐受提供了新的思路,为重组表达PD-1单链抗体的溶瘤病毒药物运用于肿瘤临床治疗奠定了坚实的理论基础。 厦门大学公共卫生学院夏宁邵教授和黄承浩助理教授为该论文的共同通讯作者,博士生林超龙和任文峰为该论文的共同第一作者。【Abstract】Oncolytic virotherapy can lead to systemic antitumor immunity, but the therapeutic potential of oncolytic viruses (OVs) in humans is limited due to their insufficient ability to overcome the immunosuppressive tumor microenvironment (TME). Here, we showed that locoregional oncolytic virotherapy upregulated the expression of PD-L1 in the TME, which was mediated by virus-induced type I and type II interferons (IFNs). To explore PD-1/PD-L1 signaling as a direct target in tumor tissue, we developed a novel immunotherapeutic herpes simplex virus (HSV), OVH-aMPD-1, that expressed a single-chain variable fragment (scFv) against PD-1 (aMPD-1 scFv). The virus was designed to locally deliver aMPD-1 scFv in the TME to achieve enhanced antitumor effects. This virus effectively modified the TME by releasing damage associated molecular patterns (DAMPs), promoting antigen cross-presentation by dendritic cells, and enhancing the infiltration of activated T cells; these alterations resulted antitumor T cell activity which led to reduced tumor burdens in a liver cancer model. Compared with OVH, OVH-aMPD-1 promoted the infiltration of myeloid-derived suppressor cells (MDSCs),resulting in significantly higher percentages of CD155+ G-MDSCs and M-MDSCs in tumors. In combination with TIGIT blockade, this virus enhanced tumor-specific immune responses in mice with implanted subcutaneous tumors or invasive tumors. These findings highlighted that intratumoral immunomodulation with an OV expressing aMPD-1 scFv could be an effective standalone strategy to treat cancers or drive maximal efficacy of a combination therapy with other immune checkpoint inhibitors.This work was supported by grant 2018ZX10301404-001-002 from the National Science and Technology Major Project and grant 81571990 from the National Natural Science Foundation of China.该研究获得了国家自然科学基金、国家科技重大专项的资助

    Evaluation on difference of therapeutic efficacy of Jiawei Xiaoyao Granules and Pills in treatment of emotional disorder during perimenopause based on Greene Climacteric Scale

    目的:在前期加味逍遥丸治疗围绝经期情绪障碍具有比较优势的基础上,基于grEEnE量表探讨其颗粒剂和丸剂不同剂型间疗效差异。方法:经筛选后患有情绪障碍的围绝经期女性75例随机等分为A、b、C3组,分别对应采用帕罗西汀、加味逍遥丸和加味逍遥颗粒剂治疗8周。所有患者在治疗前后各进行1次grEEnE及其子因子评定,并在观察期结束时进行TESS评定。结果:grEEnE总分差及其焦虑、抑郁因子分差:3种治疗方法均能改善,且相互间无显著差异;grEEnE性因子和血管因子分差:后二组均优于A组(P<0.01);TESS评分:与A组比较,b、C组均有统计学意义(P<0.01),评分均小于A组,且后二组间无显著差异。结论:加味逍遥的2种剂型间疗效无显著差异,考虑到帕罗西汀的不良反应,治疗该病加味逍遥的2种剂型都值得推荐。Objective: To investigate the difference in therapeutic efficacy between Jiawei Xiaoyao Granules and pills based on the preliminary research of treating emotional disorder during perimenopause with Jiawei Xiaoyao Pills and Greene Climacteric Scale.Methods: 75 female patients with emotional disorder during perimenopause were slected and randomly divided into 3 groups as group A, B and C.Patients in the 3 groups were treated with paroxetine, Jiawei Xiaoyao Pills and Jiawei Xiaoyao Granules respectively for 8 weeks.Greene and Greene factors assessment were carried out in all the patients before and after treatment, and the TESS assessment was carried out at the end of the observation period.Results: The three therapies all could improve the gap in total score of Greene and its factors as anxiety and depressed, and the difference among these three groups was no significant.The factors of Greene and gap in score of vascular factors of group B and C was better than that of group A(P<0.01).The difference in TESS score between group A and group B and C was significant(P<0.01), while the difference between group B and group C was not significant.Conclusion: The difference in curative effect between the two formulations was not significant.Because of the adverse reactions of paroxetine, in the treatment of emotional disorder during perimenopause, the two formulations of Jiawei Xiaoyao Powder were worth to be recommended.国家自然科学青年基金项目(No.81302960)~

    一种基于深度学习理论的齿轮系统故障诊断方法

    分别建立了基于快速傅里叶变换和深度置信网络的FFT-DBN模型、基于小波变换和深度卷积神经网络的WT-CNN模型以及基于希尔伯特-黄变换和深度卷积神经网络的HHT-CNN模型,通过将3种深度学习模型有机融合,进一步构建了基于深度学习理论的齿轮系统故障诊断综合评判模型。通过搭建功率封闭齿轮系统振动测试试验台,加工不同故障模式的测试齿轮副并提取其振动加速度信号作为样本,将基于深度学习理论的综合评判模型的故障识别效果与其他模型进行了对比。结果表明,基于深度学习理论的综合评判模型能够有效地辨识出多种齿轮故障;与其他模型相比,基于深度学习理论的综合评判模型的故障识别准确度更高

    为了研究船舶齿轮减速器隔振器布置方式对基础振动的影响,以单层隔振的齿轮传动装置为例,建立了齿轮、轴、轴承、箱体、隔振器及基础的柔性阻抗模型,并通过阻抗综合法建立了系统的耦合阻抗模型;研究了隔振器的跨距、非对称性以及数量对隔振的影响。结果表明,轻微减小隔振器跨距有利于隔振;隔振器对称布置并不是最优的减振方案;隔振器数量增加不一定总会增大振动;隔振分析时不能忽略箱体及基础的肋板特征
