Remarkable inhibitory effects of hybrid liposomes on growth of human colon cancer cells through induction of cell cycle arrest along with apoptosis

In this study, we investigated the effects of hybrid liposomes on the growth of human colon cancer cells in vitro.Methods: Hybrid liposomes (HL-n, n = 21, 23, 25) composed of L-α-dimyristoylphosphatidylcholine (DMPC) and polyoxyethylene(n) dodecyl ethers (C12(EO)n, n = 21, 23, 25) were prepared by the sonication method and their inhibitory effects on growth of human colon cancer HCT116 cells were examined in vitro.Results: Significant growth inhibition of HCT116 cells was observed in the presence of HL-n. The fifty percent inhibitory concentration (IC50) of HL-n was less than half that of DMPC liposomes. Furthermore, fluorescence microscopic and flow cytometric analyses indicated that the markedly inhibitory effects of HL-n on the growth of HCT116 cells could be attained through the induction of cell cycle arrest at the G0/G1 phase along with apoptotic cell death.Conclusion: It was found for the first time that HL-n can induce both cell cycle arrest and apoptosis in colon cancer cells. Psychological state in cancer patients

The psychological state and changes over time of cancer patients in the active treatment stage were classified into emotions by the speech and behavior of the patient described in the medical record article of the cancer psychological interview record, and the analysis of the “emotional state map” was attempted. In all cases, positive / negative emotions were mixed and appeared with variation, but a relatively large number of positive emotions, including , , and , were manifested, and the same was true in patients who experienced stressful treatment events. In the background, the existence of appropriate support from medical professionals and psychological characteristics peculiar to the stage of active treatment was inferred, such as the active treatment of the target patient, the hospitalization environment in which mental and physical pain promptly appealed to medical professionals, and the influence of psychological interviews. Cancer patients during active treatment perceived and expressed changes in the body and pain caused by the disease, and after responses from medical professionals and family members, they were conscious of their physical condition and emotions. It is suggested that this analysis method helps to objectively understand and explain the invisible and ever-changing psychological state of cancer patients in the active treatment stage

Triplet Chemotherapy with Cisplatin, Docetaxel, and Irinotecan for Patients with Recurrent or Refractory Non-small Cell Lung Cancer

We examined the feasibility of triplet chemotherapy using cisplatin, docetaxel, and irinotecan for patients with recurrent or refractory non-small cell lung cancer (NSCLC), retrospectively. Twenty-five patients (21 men and 4 women) with NSCLC and good performance status who were &#60;70 years old were analyzed. The median age was 58 years. Most patients had performance status 1 (16/25), stage IV disease (18/25) and adenocarcinoma-histology (16/25). Cisplatin and docetaxel were given on day 1 and irinotecan on day 2;the cycle was repeated every 3 weeks. The objective response rate was 39.1% (95% confidence interval:18.7-59.5%). The median survival time and actual 2-, 3-, and 5-year survival rates were 14.3 months, 32%, 20%, and 8%, respectively. Of note, only 6 patients were treated with gefitinib at the recurrence after triplet chemotherapy;of these, 4 (67%) achieved a partial response, which might result in favorable survival. Grade 3/4 toxicities consisted of neutropenia (100%), neutropenic fever (56%), nausea/vomiting (40%), and diarrhea (16%);no cases of treatment-related death occurred. Triplet chemotherapy showed impressive survival data in our clinical trial, but proved too toxic for use in treating patients with NSCLC in the clinical practice.</p

Characteristics and psychological consequences of Japanese adolescents\u27 personal autonomy

Previous studies on the concept of parental authority during adolescence have reported that the mental health of adolescents tend to deteriorate when parents control their behavior in their personal domain. Three surveys were conducted with Japanese early, middle, and late adolescents to examine whether adolescents that underestimate parental authority and exhibit personal autonomy are mentally healthy. Study 1: six-grade elementary school students (N=276), Study 2: junior high school students (N=246)and senior high school students (N=357), and Study 3: university students (N=148).In each study, judgments of adolescents about personal freedoms involved in moral, conventional, multifaced,and prudential issues that would be regulated in Japanese society were examined, and correlations with their self-formation and adaptation were analyzed. The results of in every study, adolescents could distinguish situations in which they could demonstrate their personal freedom from those in which their personal freedom is regulated.Furthermore, cluster analysis was conducted on judgments about personal freedom. The results indicated that in every study, there were two types of young people, other than those that made a judgment consistent with characteristics of each domain. One type had excessive feelings of freedom and tended to judge that personal freedom should be valued even in conventional and prudential situations. The other type excessively suppressed personal freedoms and tended to suppress it even in personal settings. The former type had relatively bad relationships with their parents and showed lower adaptation to school (Study 1). They tended to have a moral self with a sense of helplessness and social avoidance (Study 2). Moreover, their mental health deteriorated significantly (Study 3). Based on the above results and those of previous studies, it is suggested that too much control by parents and too strong feelings of personal freedom might have negative effects on the development of the self in adolescence

Is Adenosine Deaminase in Pleural Fluid a Useful Marker for Differentiating Tuberculosis from Lung Cancer or Mesothelioma in Japan, a Country with Intermediate Incidence of Tuberculosis?

The objective of this study was to evaluate the utility of the determination of adenosine deaminase (ADA) level in pleural fluid for the differential diagnosis between tuberculous pleural effusion (TPE) and malignant pleural effusion (MPE) in Japan, a country with intermediate incidence of tuberculosis (TB). We retrospectively reviewed the clinical records of 435 patients with pleural effusion and investigated their pleural ADA levels as determined by an auto analyzer. ROC analysis was also performed. The study included patients with MPE (n＝188), TPE (n＝124), benign nontuberculous pleural effusion (n＝94), and pleural effusion of unknown etiology (n＝29). The median ADA level in the TPE group was 70.8U/L, which was significantly higher than that in any other groups (p＜0.05). The area under the curve (AUC) in ROC analysis was 0.895. With a cut-off level for ADA of 36U/L, the sensitivity, specificity, positive predictive value, and negative predictive value were 85.5%, 86.5%, 69.7%, and 93.6%, respectively. As many as 9% of patients with lung cancer and 15% of those with mesothelioma were false-positive with this ADA cutoff setting. Although the ADA activity in pleural fluid can help in the diagnosis of TPE, it should be noted that some cases of lung cancer or mesothelioma show high ADA activity in geographical regions with intermediate incidence of TB, in contrast to high prevalence areas
