Metal-Ion-Binding Oligonucleotides: High-Affinity Probes for Nucleic Acid Sequences
Small non-coding RNAs have numerous biological functions in cell and are divided into different classes such as: microRNA, snoRNA, snRNA and siRNA. MicroRNA (miRNA) is the most studied non-coding RNA to date and is found in plants, animals and some viruses. miRNA with short sequences is involved in suppressing translation of target genes by binding to their mRNA post-transcriptionally and silencing it. Their function besides silencing of the viral gene, can be oncogenic and therefore the cause of cancer. Hence, their roles are highlighted in human diseases, which increases the interest in using them as biomarkers and drug targets. One of the major problems to overcome is recognition of miRNA. Owing to a stable hairpin structure, chain invasion by conventional Watson-Crick base-pairing is difficult. One way to enhance the hybridization is exploitation of metal-ion mediated base-pairing, i. e. oligonucleotide probes that tightly bind a metal ions and are able to form a coordinative bonds between modified and natural nucleobases. This kind of metallo basepairs containing short modified oligonucleotides can also be useful for recognition of other RNA sequences containing hairpin-like structural motives, such as the TAR sequence of HIV. In addition, metal-ion-binding oligonucleotides will undoubtedly find applications in DNA-based nanotechnology. In this study, the 3,5-dimethylpyrazol-1-yl substituted purine derivatives were successfully incorporated within oligonucleotides, into either a terminal or non-terminal position. Among all of the modified oligonucleotides studied, a 2-(3,5-dimethylpyrazol-1-yl)-6-oxopurine base containing oligonucleotide was observed to bind most efficiently to their unmodified complementary sequences in the presence of both Cu2+ or Zn2+. The oligonucleotide incorporating 2,6-bis(3,5-dimethylpyrazol-1-yl)purine base also markedly increased the stability of duplexes in the presence of Cu2+ without losing the selectivity.Siirretty Doriast
Combination therapy in chronic periodontitis: a case report
Background: The aim of periodontal treatment is to provide healthy and functional dentition for the whole life.Cases Report: A 42 year old female with sever chronic periodontitis, treated medically and surgically, is reported. She initially received antibiotic, Scaling and root planning in addition to oral hygiene instruction. After four weeks, periodontal Surgery, root canal Therapy, extraction of excess tooth and restoration of some teeth were performed, because of remaining residual pockets, and bone loss, flap Surgery and Access flap, with papilla preservation flap method, also modified Widmann flap, were done. After one month, favorable clinical improvement was obtained.Conclusion: It can be concluded that high oral hygiene level in accompanied with Suitable medical and surgical treatment, enhanced the success of periodontal treatment outcomes even in sever disease.
HybridChain: Fast, Accurate, and Secure Transaction Processing with Distributed Learning
In order to fully unlock the transformative power of distributed ledgers and
blockchains, it is crucial to develop innovative consensus algorithms that can
overcome the obstacles of security, scalability, and interoperability, which
currently hinder their widespread adoption. This paper introduces HybridChain
that combines the advantages of sharded blockchain and DAG distributed ledger,
and a consensus algorithm that leverages decentralized learning. Our approach
involves validators exchanging perceptions as votes to assess potential
conflicts between transactions and the witness set, representing input
transactions in the UTXO model. These perceptions collectively contribute to an
intermediate belief regarding the validity of transactions. By integrating
their beliefs with those of other validators, localized decisions are made to
determine validity. Ultimately, a final consensus is achieved through a
majority vote, ensuring precise and efficient validation of transactions. Our
proposed approach is compared to the existing DAG-based scheme IOTA and the
sharded blockchain Omniledger through extensive simulations. The results show
that IOTA has high throughput and low latency but sacrifices accuracy and is
vulnerable to orphanage attacks especially with low transaction rates.
Omniledger achieves stable accuracy by increasing shards but has increased
latency. In contrast, the proposed HybridChain exhibits fast, accurate, and
secure transaction processing, and excellent scalability
A High-throughput and Secure Coded Blockchain for IoT
We propose a new coded blockchain scheme suitable for the Internet-of-Things
(IoT) network. In contrast to existing works for coded blockchains, especially
blockchain-of-things, the proposed scheme is more realistic, practical, and
secure while achieving high throughput. This is accomplished by: 1) modeling
the variety of transactions using a reward model, based on which an
optimization problem is solved to select transactions that are more accessible
and cheaper computational-wise to be processed together; 2) a transaction-based
and lightweight consensus algorithm that emphasizes on using the minimum
possible number of miners for processing the transactions; and 3) employing the
raptor codes with linear-time encoding and decoding which results in requiring
lower storage to maintain the blockchain and having a higher throughput. We
provide detailed analysis and simulation results on the proposed scheme and
compare it with the state-of-the-art coded IoT blockchain schemes including
Polyshard and LCB, to show the advantages of our proposed scheme in terms of
security, storage, decentralization, and throughput
Metal Ion Chelates as Surrogates of Nucleobases for the Recognition of Nucleic Acid Sequences: The Pd2+ Complex of 2,6-Bis(3,5-dimethylpyrazol-1-yl)purine Riboside
A 2,6-bis(3,5-dimethylpyrazol-1-yl)purine ribonucleoside has been prepared and incorporated as a conventionally protected phosphoramidite into a 9-mer 2′-O-methyl oligoribonucleotide. According to 1H NMR spectroscopic studies, this nucleoside forms with Pd2+ and uridine a ternary complex that is stable at a micromolar concentration range. CD spectroscopic studies on oligonucleotide hybridization, in turn, suggest that the Pd2+ chelate of this artificial nucleoside, when incorporated in a 2′-O-methyl-RNA oligomer, is able to recognize thymine within an otherwise complementary DNA strand. The duplex containing thymidine opposite to the artificial nucleoside turned out to be somewhat more resistant to heating than its counterpart containing 2′-deoxycytidine in place of thymidine, but only in the presence of Pd2+. According to UV-melting measurements, replacement of 2′-O-methyladenosine with the artificial nucleoside markedly enhances hybridization with a DNA target, irrespective of the identity of the opposite base and the presence of Pd2+. With the thymidine containing DNA target, the Tm value is 2–4°C higher than with targets containing any other nucleoside opposite to the artificial nucleoside, but the dependence on Pd2+ is much less clear than in the case of the CD studies
Clinical image; Management of Coronary Artery Thrombosis
Background: Acute myocardial infarction is an increasing cause of morbidity and mortality in the world. This condition sometimes happens as a result of coronary arteries blockage due to clot formation, spasm or plaque rupture.
Cases Report: A 31-year-old man referred to our hospital with severe exertional chest pain. Electrocardiogram revealed ST- segment elevation in precordial leads. Coronary arteries angiography revealed a visible clot at proximal part of left anterior descending artery. As the thrombolysis in myocardial infarction (TIMI) flow of the artery was normal and the patient became asymptomatic , conservative management was considered. The patient was commenced on anticoagulant, glycoprotein 2b/3a inhibitor, aspirin, and clopidogrel. Follow-up angiography showed normal flow with no clot existence.
Conclusion: Coronary thrombus is an etiology of acute coronary syndrome. Patients often have varying levels of concurrent atherosclerosis. It’s management is anticoagulation, thrombolysis, and occasionally invasive strategies . Some cases can be managed conservatively, so decision-making is an import part of these patients management
Simultaneous right coronary artery spasm in a patient with Anterior ST-Segment Elevation Myocardial Infarction: a case report

We report simultaneous right ventricular and anterior ST-segment elevation myocardial infarction in a previously healthy young man.The angiographic results demonstrated the simultaneous occlusion of the right and left coronary arteries because of simultaneous occlusion of left anterior descending artery (LADA) and spasm of right coronary artery (RCA). In this patient, we found simultaneous ST elevations in right and precordial leads so everyone should be careful about all leads of the surface electrocardiogram for decision making in the management of a patient
