    The Solid Phase Microextraction (SPME) method was originally developed to extract volatile and semivolatile compounds from wastewater samples but has since been applied to flavour compounds in foods and beverages. Research using the HS-SPME in related areas such as cheese and skim milk powder has been carried out but, to date, no work has been done on yoghurt flavours. The main objective of this study was to devise a methodology for the Headspace Solid Phase Microextraction (HS-SPME) technique to investigate and quantify six flavour analytes in natural, set yoghurts made from recombined milk. The relevant literature was reviewed and from it, a research proposal for this work on yoghurts was drawn. The first step in analysing and quantifying the yoghurt volatiles was to set up a working methodology for the HS-SPME method. The 100 μm polydimethylsiloxane (PDMS) fibre was chosen along with 20 minutes being the optimum fibre adsorption time. General equipment, materials and methods used throughout this thesis are also detailed. The external standard (ES) method was used to calibrate the GC and quantify the analyte concentrations in this study. The internal standard (IS) method was not used as a quantitative tool in this study. Once the HS-SPME methodology had been set up for the analysis of yoghurts, the classical Static Headspace (SH) method was compared with the HS-SPME method for extraction efficiency. The results suggested that the two methods were complementary in that the SH method extracted the more volatile compounds (acetaldehyde, acetone and 2-butanone) whereas, the HS-SPME method extracted the semi- to non-volatile compounds (ethanol, diacetyl and acetoin) more readily. However, the HS-SPME was found to be the more sensitive and effective method of the two techniques tested. The next step in the thesis was to investigate the presence of the six analytes in milk and cultured yoghurt. The effects of the sample matrix, fat levels and incubation on the volatile concentrations were also examined. The results suggested that the six analytes were inherently present in milks but at low concentrations. No conclusive effects were found for the sample matrix, fat levels and incubation. However, it was evident that fermentation of the milks using bacterial starter cultures resulted in a large increase in some of the volatiles being investigated. Following this, the effects of fat levels, storage time and storage temperature on the six volatiles in yoghurts were examined. The results indicated that significant fat level effects were only seen for diacetyl and acetoin, while temperature effects were only observed for ethanol. In both trials, only general trends for the analytes concentrations were drawn because the data varied from day to day. The results suggested that most of the compounds decreased with time except for diacetyl, which seemed to increase. The final part of this study looked at applying the devised HS-SPME methodology to a series of commercial yoghurts as a preliminary trial, with a view to investigating a potential application for the HS-SPME method. Fourteen commercial yoghurts were analysed and the six analytes quantified. The data obtained was analysed using Principle Component Analysis (PCA), which divided the yoghurts into groups based on their analyte concentrations. From these groupings, eight yoghurts were selected and fresh samples were analysed using HS-SPME and PCA. This was carried out parallel with an untrained consumer panel, which had to distinguish differences between the yoghurts in a series of triangle tests by smelling the headspace on opening the yoghurt containers. The conclusions drawn were that, unlike the HS-SPME method with PCA, the average consumer could not differentiate the yoghurts based on smell alone. PCA also showed that the HS-SPME results obtained were fairly reproducible. In conclusion, the HS-SPME method was shown to be a useful analytical technique, which can be used to analyse and quantify flavour compounds in natural, set yoghurts. This area of investigation has a lot of scope, with the results from this study providing a basis or starting point for further investigations in this area. Future studies may lead to potential applications for the HS-SPME method, one of which may be quality control where correlation of sensory data with HS-SPME analytical data is required

    A Dynamics and Stability Framework for Avian Jumping Take-off

    Jumping take-off in birds is an explosive behaviour with the goal of providing a rapid transition from ground to airborne locomotion. An effective jump is predicated on the need to maintain dynamic stability through the acceleration phase. The present study concerns understanding how birds retain control of body attitude and trajectory during take-off. Cursory observation suggests that stability is achieved with relatively little cost. However, analysis of the problem shows that the stability margins during jumping are actually very small and that stability considerations play a significant role in selection of appropriate jumping kinematics. We use theoretical models to understand stability in prehensile take-off (from a perch) and also in non-prehensile take-off (from the ground). The primary instability is tipping, defined as rotation of the centre of gravity about the ground contact point. Tipping occurs when the centre of pressure falls outside the functional foot. A contribution of the paper is the development of graphical tipping stability margins for both centre of gravity location and acceleration angle. We show that the nose-up angular acceleration extends stability bounds forward and is hence helpful in achieving shallow take-offs. The stability margins are used to interrogate simulated take-offs of real birds using published experimental kinematic data from a guinea fowl (ground take-off) and a diamond dove (perch take-off). For the guinea fowl the initial part of the jump is stable, however simulations exhibit a stuttering instability not observed experimentally that is probably due to absence of compliance in the idealised joints.     Wadge Bank trawl fishery studies. Pt. 2. The effect of trawling on the catch per hour from 1945 to 1960

    The object of this series of papers has been given in Part 1 (see Additional informations for details) which deals with the first known commercial fishery on the Wadge Bank from 1928 to 1935. There is no recorded trawling on the Bank between 1936 and 1944. This paper deals with the changes in the total catch (i.e. all species combined) per hour of trawling in relation to the changes in fishing intensity from 1945, when the present trawling activities started, to 1960. The effect of trawling on individual categories or varieties will be presented later

    Wadge Bank trawl fishery studies. Pt. 5. Rational exploitation of the resident demersal stock

    The general history of the trawl fishery of the Wadge Bank off Cape Comorin, South India (Fig. 1), the nature and composition of its demersal fish population, and the present state of its fishery has been given by various authors (Malpas 1926, Pearson and Malpas 1926, Sivalingam and Medcof 1957, Medcof 1963, Mendis 1965a, 1965b, Sivalingam 1966a, 1966b, 1969a and 1969b). It has been shown earlier (Sivalingam and Medcof 1957, Sivalingam 1969a) that the Wadge Bank stock is made up of two groups. The resident stock which is present on the fishing grounds throughout the year and the migrant stock that appears on the fishing grounds only during the southwest monsoon months. The object of this paper is to discuss the effect of fishing on the resident stock between 1945 and 1962 and based on the information available; assess the maximum sustained yield of the resident stock. The "Bigfish" of the resident stock is the mainstay of the Wadge Bank trawl fishery (Sivalingam 1969a) and it will be shown that this stock has been overexploited from 1953 to 1957. The first sign of recovery was evident in 1960 and continued till 1962. The data since 1962 are not available to the author for analysis. It has been reported by Mendis (1965b) that considerable expansion of the trawler fleet was anticipated in 1966, and if so, the history of the fishery from 1953 to 1957 may be repeated. The assessment presented in this paper should form a rational basis for management programs

    Wadge Bank trawl fishery studies. Pt. 4. An analysis of the length frequency measurements of the sea bream (Lethrinus nebulosus) made in 1949 and 1953 to 1958

    Length frequency distributions of the sea bream collected during the period 1953 to 1958 have been analysed. The increase in average sizes of the sea bream with depth suggests a movement to deeper waters with increase in size. By numbers, the sea bream is more abundant between 21 and 30 fathoms than in deeper areas. The recruitment was continuous and regular. There is no sign of entry or progression of a dominant brood throughout the period under study. Length frequency distribution shows three distinct modes. The first mode occurs regularly but does not progress beyond 40cm, recruitment being balanced by natural and fishing mortality. The other two which are not regular are probably the result of fishing outside regular areas. Short sections of “growth” lines which fit into one another when extrapolated, are evident. The larger lines obtained by extrapolation are parallel to one another. These tentative "growth lines" indicate that this species which enters the fishing grounds, when 15 cm or larger in length are exploited by the trawl fishery for a period of three to four years. This species appears to be six months old when it enters the fishing grounds and increases in length by about 37.5 cm in the next 30 months. Later growth slows down. The average size of the specimens sampled continued to get smaller from 1953 till 1957. It is shown that this reduction in size is due to increased fishing effort

    DNA Barcoding of Endangered Sacred Plant (Stereospermum suavelons) in Cuddalore District, Tamil Nadu, India

    In the present investigation, DNA barcodings were made for the sacred trees of Main (Big) Temples in Cuddalore district, namely, Stereospermum suavelons to document their reliable identification, conservation, discrimination, similarities and evolutionary trend among them and with their related taxa for future use.  DNA isolation from leaf samples of present study species was carried out by using a modified CTAB method and good isolation was got for the species studied.Gradient PCR amplification was performed for the isolated DNA using matK gene and the primers matK472F & matK1248R.  The amplification success was 90-95%. PCR amplification was tested with 1 % agarose gel electrophoresis using ethidium bromide and the products were confirmed.  The PCR products were sent to Xcelris Labs Ltd. Sydney House, Premchand Nagar Road, Bodakdev, Ahmedabad 380054, India, for DNA sequencing and sequences were got for the species with the success rate of 95 %.  Pairwise sequence alignments were made with BLAST and multiple sequence alignments are made with ClustalW, and based on the sequence alignments, dentrograms were constructed using software Mega 5 and Neighbor joining method to study the phylogenetic aspects of the species studied and with their related taxa.
