Investigating political risk in the German energy industry
This study analyses the phenomenon of political risk in the German energy industry. Political risk can be understood as “the probability that factors caused or influenced by the (in) action or reactions of stakeholders within a political system to events outside or within the country” (Brink 2004). This study asks for determinants and characteristics of political risk, its origination, its occurrence, and its impact. The focus of this study lies in the German energy industry, where politically initiated drastic changes are the predominant challenge for utilities (Roland Berger, 2013). The industry levels investigated include strategic and risk company’s issues and are focused on the senior management and senior politicians’ insights. It looks for the identification and analysis of the linkage of political risk and their effect on utility companies in Germany of different sizes and active within different sectors.
While political risk is not limited to Germany or the energy industry, there have been enormous changes in this field recently: the German government recently decided to shut down all nuclear-fuelled power plants as part of a programme called energy turnaround (Energiewende). The content of this programme will change the industry structure radically. Renewable energy is now heavily favoured with a consequent decline in nuclear energy usage.
This study analyses political risk combining two approaches: an outside-in and an inside-out analysis. Two important perspectives are captured, evaluated and compared with each other. The first group of interviews focuses on senior political experts, who are strongly connected with the energy industry. This “inside” information comes from experts including members of the state and federal parliament, as well as European parliament and one former federal minister of economics. The second group of interviews therefore seeks “outside” information from industry experts, senior managers of the German energy industries, who are daily confronted with the impact of political risk.
It is enriching to combine these two sides, to develop an understanding of the phenomenon of political risk in the German energy industry. The approach of having two interview series with conflicting expert opinion generates a new view of this topic. The findings illustrate these ideas, thoughts, and opinions together, which helps to explain the different sides of political risk in the German energy industry and generates approaches for the utility companies to take to mitigate political risk.
The analyses of the experts’ insights generated the following results
(1) identification and description of the definitional tensions of understanding political risk in the German energy industry
(2) analysis of the political risks the industry is facing
(3) evaluation of the potential impact of these risks on the industry and its companies
(4) development of a conceptual approach for political risk management in the German energy industry.
In detail, it has been derived from the analysis that there are huge differentiations in the understanding of political risk between politicians and managers. There is also no common understanding within the group of politicians. The different perception of political risk was also evident for the types of political risk that were discussed in this study.
Despite the observed influence of political decisions on single enterprises in Germany in recent history, there are also differentiations in the evaluation of the impact of political risk within practice. CLONING OF THE 1.4-kb mRNA SPECIES OF HUMAN COMPLEMENT FACTOR H REVEALS A NOVEL MEMBER OF THE SHORT CONSENSUS REPEAT FAMILY RELATED TO THE CARBOXY TERMINAL OF THE CLASSICAL 150-kDa MOLECULE
Three factor H mRNA species of 4.3 kb, 1.8 kb,
and 1.4 kb are constitutively expressed in human
liver. Having previously characterized full-length
cDNA clones derived from the 4.3-kb and 1.8-kb
factor mRNA, we report here the isolation and eucaryotic
expression of full-length cDNA clones coding
for the 1.4-kbm RNA species. The 1266-bp cDNA
codes for a polypeptide of 330 amino acids and
contains two polyadenylation signals and a short
poly(A)+tailT. he protein is composed of a leader
peptide followed by five short consensus repeat domains.
It shows a hybrid structure with the last
three domains being almost identical to the carboxy-
terminal of thcel assical 1 BO-kDa factor H molecule
and the two first domains representing unique
short consensus repeat structures. Eucaryotic
expression in COS7 cells revealed two polypeptides
derived from one cDNA clone that area lso found in
human serum. Differences between the cDcNloAn es
within the last three domains indicate two distinct,
possibly allelic sequences that, in addition, differ
from the authentic 150-kDa factor H sequence.
Southern blot results support the notion that the
4.3-kb factor H and the 1.4-kb factor H-related
mRNA are transcribed from two separate but highly
homologous genes.

Human complement factor H
We isolated cDNA clones coding for the functionally important tryptic N-terminal38-
kDa fragment of human complement control protein factor H using polyclonal and
monoclonal antibodies to screen a human liver cDNA library cloned in a bacterial
expression vector, PEX-1. By testing the reactivity of antibodies specific for the
recombinant proteins produced by individual clones with proteolytic fragments of
serum H the exact position of these cDNA clones within H was mapped. One clone,
H-19, coding for the 38-kDa fragment of H was sequenced and found to code for 289
amino acids derived from the 38-kDa N-terminal fragment as well as for the first 108
amino acids belonging to the complementary 142-kDa tryptic fragment. The derived
protein sequence could be arranged in 6 highly homologous repeats of about 60 amino
acids each, the homology between the repeats being determined by the characteristic
position of cysteine, proline, glycine, tyrosine and tryptophane residues. The region
coding for the epitope recognized by one of our monoclonal antibodies was localized
by subcloning restriction fragments of H-19 into the expression plasmid and testing
for the expression of this epitope
CD44 in Differentiated Embryonic Stem Cells: Surface Expression and Transcripts Encoding Multiple Variants
Expression of the surface-adhesion molecule CD44 was investigated during the in vitro
differentiation of the embryonic stem (ES) cell line D3. By immunofluorescence analysis,
totipotent, undifferentiated ES cells did not show surface expression of CD44, although two
transcripts of approximately 1.6 and 3.3 kb were detected on Northern blots. Following 1
week of differentiation in either suspension or substrate-attached cultures, CD44 appeared
on the surface of some D3 cells, and synthesis of an additional 4.5 kb mRNA species was
detected on Northern blots. At this stage, at least three distinct transcripts encoding CD44
variants were induced within the cultures, resulting from alternative splicing of additional
exons in the variable domains of CD44. From PCR analysis, they all appeared to contain the
variable exon v10, and two of them in addition contained v6. Taken together, these results
suggest that CD44 may play a role in cell migration and adhesion in the early development
of the mouse embryo

This strain has hampered programs and affects the health of the urban forest. As our globalized market increases, invasive species become a growing threat that U&CF programs struggle with in their debilitating state. The invasive species, emerald ash borer, has the ability to cause catastrophic harm to Minnesota communities due to the high percentage of ash on public owned property. In an effort to provide assistance, and build capacity for dealing with emerald ash borer, the Emerald Ash Borer: Rapid Response project was developed. This project utilized volunteers and community gravel beds to build U&CF infrastructure to increase the ability of a community to manage not just emerald ash borer, but develop a successful and sustaining U&CF program. The intensive and direct assistance method used in the project provides new ways for states and other organizations to consider building and tracking U&CF capacity in communities
Speichel als alternatives Untersuchungsmaterial zur Drogenanalytik mittels Immunoassay: Ein Vergleich zu verschiedenen Nachweisverfahren im Urin in drogentherapeutischen Einrichtungen
Dem Nachweis von Drogen und psychotropen Substanzen im Speichel kann bei verschiedenen klinisch-forensischen Fragestellungen, zum Beispiel im Bereich der Verkehrsmedizin, eine große Bedeutung zukommen. Die Reliabilität des so genannten Drogenscreenings im Speichel mit Hilfe des CEDIA-Verfahren sowie mit dem EMIT-Verfahren (für Cannabinoide) untersucht. Für die Speicheltestung wurde durch Vorversuche eine untere Nachweisgrenze (LOD) festgelegt. Die Festlegung des jeweiligen Cut-offs der einzelnen Analyte erfolgte nach Auswertung der ROC-Kurve. Sämtliche Urinproben wurden mit dem CEDIA-Verfahren sowie mit Urinstäbchen (GLORIA-Verfahren) untersucht. Zur Auswertung kamen 96 Speichel- und 103 Urinproben von insgesamt 31 opiatabhängigen Patienten. Es zeigte sich, dass im Speicheltestverfahren nur für Methadon vergleichbar gute Ergebnisse wie im Urintestverfahren erreicht werden konnten. Nur mäßige Testergebnisse hinsichtlich der Aussagefähigkeit der Immunoassayverfahren im Speichel ergaben sich für die meisten anderen untersuchten Drogen wie Amphetamine und Barbiturate, Kokain und Opiate. Für Cannabinoide und Benzodiazepine erscheint diese Methode nicht geeignet zu sein. Dagegen zeigten sich bis auf eine niedrige Sensitivität bei Kokain bei allen Analyten mit Urinstäbchen gute Nachweisergebnisse. Die Kombination von einem Immunoassayverfahren und Bestätigungsanalysen, wie zum Beispiel Gaschromatographie (Massenspektroskopie) im Urin, erscheint daher weiterhin als die Methode der ersten Wahl bei nicht invasiven Testverfahren. Dagegen ist der Einsatz von Speicheltestverfahren, etwa in dem Bereich der Verkehrsmedizin eher kritisch zu werten und erscheint am ehesten bei Substitutionsbehandlungen als zusätzliches Testverfahren sinnvoll
Molekulare Identifizierung und funktionelle Charakterisierung der K+-Kanal-Ausstattung auditorischer Haarsinneszellen
Hintergrund: Die präzise Verarbeitung von auditiven Reizen wird durch äußere (ÄHZ) und innere Haarsinneszellen (IHZ) im Corti´schen Organ gewährleistet. Da die physiologischen Aufgaben (kochleäre Verstärkung und synaptische Übertragung) dieser Zellen vom Rezeptorpotential angetrieben werden, sind Form und Amplitude dieser Potentialänderungen essentiell für die Funktion der Sinneszellen. Die Charakteristika der Rezeptorpotentiale hängen dabei von der Aktivität der apikalen mechano-elektrischen Transduktionskanäle und von basolateralen K+-Leitfähigkeiten ab. Diese Leitfähigkeiten wurden aufgrund ihrer biophysikalischen Eigenschaften als IK,f, IK,n und IK,s klassifiziert. Die Ionenkanäle und deren Interaktionspartner, die diese K+-Leitfähigkeiten hervorbringen, sind weitestgehend unbekannt.
Ziele der Arbeit: Die Ziele dieser Doktorarbeit waren es, (I.) die Relevanz eines möglichen Interaktionspartners (das Enzym β-Sekretase 1; BACE1) der K+-Kanäle in ÄHZ zu untersuchen, und (II.) die K+-Kanaluntereinheiten, welche die IK,s Komponente in IHZ von Mäusen ausmachen, zu entschlüsseln, sowie deren physiologische Aufgaben anhand von Computermodellen zu bestimmen.
Ergebnisse zu BACE1: Ich konnte zeigen, dass der signifikante Hörverlust der BACE1-/- Mäuse durch Desorganisation der synaptischen Verbindungen zwischen IHZ und auditorischen Nervenfasern und durch eine stark verringerte Myelinisierung dieser Nervenfasern hervorgerufen wird. Dabei ist BACE1 für die Entwicklung der kochleären Nervenfasern, nicht jedoch für die Aufrechterhaltung der Myelinisierung von Bedeutung. Außerdem konnte ich Neuregulin-1 als BACE1-Substrat identifizieren, das die Organisation der Nervenfasern in der Cochlea vermittelt. BACE1 ist also entgegen unserer Ausgangshypothese kein Interaktionspartner von K+-Kanälen in Haarsinneszellen, jedoch unabkömmlich für sensitives Hören. Da die Inhibition von BACE1 ein populärer Ansatz zur Therapie der Alzheimer-Krankheit ist, war es wichtig zu prüfen, ob entsprechende Medikamente Auswirkungen auf die Hörleistung haben. Die Behandlung von Mäusen mit dem BACE1-Inhibitor NB-360 beeinträchtigte jedoch nicht das Hörvermögen der Tiere. Diese Ergebnisse lassen vermuten, dass Hörverlust keine Nebenwirkung der Therapie der Alzheimer-Krankheit darstellt.
Ergebnisse zur K+-Kanalausstattung der Haarsinneszellen: In einem Hypothesen-basierten Kandidatenansatz ist es mir gelungen, die K+-Kanalausstattung von IHZ zu entschlüsseln. Dafür habe ich aus publizierten Einzelzell-Transkriptom-Analysen von IHZ Kandidatenkanäle extrahiert und deren Expression in der Cochlea mittels molekularbiologischen und immunhistochemischen Techniken bestätigt. Mittels elektrophysiologischer Methoden konnte ich dann überprüfen, ob diese Kanäle in den IHZ aktiv sind. Für den funktionellen Nachweis konnte ich zum einen auf etablierte Protokolle zurückgreifen, andere Kandidatenkanäle musste ich erst im Expressionssystem eingehend charakterisieren und selbst geeignete Messprotokolle und ein pharmakologisches Profil zum Nachweis in nativem Gewebe entwickeln. Diese Untersuchungen zeigten, dass sich trotz enger phylogenetischer Verwandtschaft und darauf aufbauenden Ähnlichkeiten in der Gesamtarchitektur, die pharmakologischen Eigenschaften von K+-Kanälen doch signifikant unterscheiden können. Aufbauend auf den Arbeiten im Expressionssystem konnte ich dann zeigen, dass Kv1.8-, Kv11.1- und Kv12.1-Kanäle in IHZ aktiv sind. Somit konnte ich Kv11.1-Kanälen erstmals eine Rolle im sensorischen System, und Kv1.8- und Kv12.1-Kanälen überhaupt erstmalig eine physiologische Bedeutung zuweisen. Die Relevanz dieser außergewöhnlichen Ausstattung aus fünf K+-Kanaluntereinheiten in IHZ (Kv1.8-, Kv7.4-, Kv11.1-, Kv12.1- und BKCa-Kanäle) wurde dann anhand von Computermodellen eingehend untersucht. Diese Simulationen zeigten, dass alle Kanäle gemeinsam die Präzision der Schallverarbeitung in IHZ optimieren und die Verarbeitung komplexer Schallereignisse und somit sensitives Hören überhaupt erst ermöglichen, während IHZ mit vereinfachter Kanalzusammensetzung in der Verarbeitung von Schallreizen eingeschränkt sind
