    Effect of hydrolysed fish protein and autolysed yeast as alternative nitrogen sources on gilthead sea bream (Sparus aurata) growth performances and gut morphology

    The aim of the present study was to compare the effects of two different nitrogen-rich ingredients such as hydrolysed fish protein and autolysed yeast, on gilthead sea bream (Sparus aurata) growth performances and histological gut morphology. Animals were allocated to three experimental groups: the first received a fishmeal-based diet (FM), the second and the third received hydrolysed fish protein (HFP) and autolysed yeast (AY), respectively, to replace an equal amount of fishmeal. The experiment lasted 92 days. No significant differences in body weight and mortality were observed among the different groups. Villus branching, intraepithelial lymphocytes and inflammatory infiltrate of the submucosa were more prominent in AY than HFP and FM. The gut absorbent surface ratio was 5.94, 6.44 and 7.28 for group FM, HFP and AY, respectively, with statistical significant difference between FM and AY and between HFP and AY. A significant increment in the goblet cell density was observed for HFP and AY in comparison to FM. A statistically significant increase in small-sized goblet cells was observed in AY compared to FM. All our findings suggest that it is possible to use either HFP or AY, as partial replacer of FM in the S. aurata diet.Highlights Yeast-derived products as a suitable alternative nitrogen source.     Fishmeal replacement with hermetia illucens meal in aquafeeds: Effects on zebrafish growth performances, intestinal morphometry, and enzymology

    Fishmeal (FM) is still the most important protein source in aquafeeds. However, due to the reduction of wild fish stocks used for FM production, its manufacturing it is now unsustainable. Insect meal represents a valid alternative to FM, due to the low carbon footprint of its production and its high nutritional value. The aim of this study was to investigate the potentials of replacing FM with black soldier fly (Hermetia illucens) meal (HIM) in aquafeeds, using zebrafish as the animal model. Four diets were formulated with increasing HIM/FM replacement rate (the control diet contained 20% FM). Mortality rates, growth performances, and feed consumptions were measured, and fish intestine samples were collected for histological and enzymatic analysis. After 49 days, all groups almost tripled their initial body weight (cumulative body weight gain ranged between 143.9 and 155.2 mg), and no statistically significant differences among treatments were observed in relation to growth performances and histological traits. Contrarily, trypsin, alkaline phosphatases, and alpha amylase–glucoamylase activities were significantly reduced when the FM replacement rate was increased. In conclusion, HIM may represent an alternative to FM, since no adverse effects were observed when it was included up to 20% in the zebrafish diet, even when replacing 100% of the FM

    Use of Photocatalysis for Degradation of Glyphosate in Potable Water of CKDu Prevalent Areas

    Glyphosate, which is commercially available as Roundup®, was the most widely used herbicide in Sri Lanka until recently. Recent studies report the presence of glyphosate in different water sources (drinking water, surface water, groundwater) in the Chronic Kidney Disease of unknown etiology (CKDu) prevalent areas containing elevated hardness levels. Additionally, WHO studies conducted in 2013, provide evidence of the presence of glyphosate in trace levels in urine samples of CKDu subjects. Thus, glyphosate is suspected to be one of the causal factors for CKDu and its removal, when present in potable water with high hardness levels, is a challenging engineering task. Photocatalysis is recognised to be one of the promising technologies for degradation of glyphosate. Hence, this study focuses on investigating the effectiveness and efficiency of the photocatalysis process for degradation of glyphosate from potable waters containing high hardness levels. Experiments were conducted with high purity Glyphosate and Roundup® in the absence (0 mg/l as CaCO3) and presence of hardness (1,500 mg/l as CaCO3). A sunlight activated water purifier bag available in the market that consists of a Titanium Dioxide membrane was used for application of the photocatalysis process. A 3.0 L water sample containing Glyphosate/Roundup® (glyphosate concentration 1 mg/L) was continuously exposed to sunlight for six (06) hours. A solar irradiance meter was used to measure the sunlight intensity. Samples from the reaction solution were collected at predetermined time intervals and analysed for glyphosate and amninomethyphosphonic acid (AMPA) using GC/MS and LC/MS in the absence and presence of hardness, respectively. A rapid decrease in glyphosate concentration to levels below 0.7 mg/l (USEPA Maximum Contaminant Level) was observed during the first 60 minutes of reaction time under all experimental conditions studied. The concentration of AMPA detected was below the level of quantification in all experiments implying that complete degradation of Glyphosate has occurred until inorganic phosphate is produced. When using high purity glyphosate or Roundup®, slower degradation rates were observed in the presence of hardness compared to that in the absence of hardness. Retardation of glyphosate degradation in the presence of hardness could be attributed to the enhanced persistence of glyphosate due to the formation of glyphosate-Ca and -Mg complexes when hardness is present in water. Similarly, slower degradation rates were observed with Roundup® compared to that of with high purity glyphosate, both in the absence and presence of hardness. The presence of the major adjuvant surfactant [polyethoxylatedtallowamine, (POEA)], which is introduced as an inert ingredient during Roundup® production seems to have enhanced the persistence of glyphosate in water thus causing retardation of Glyphosate degradation by the photocatalysis process. In conclusion, photocatalysis is an effective and efficient technique that could be used to degrade glyphosate in potable water of CKDu prevalent areas in spite of the possible antagonistic effects observed on Glyphosate degradation due to the presence of hardness and/or surfactants.Keywords: CKDu, Glyphosate, Photocatalysi

    Unexpected combination of acute croup and myocarditis: Case report

    BACKGROUND: Lower vaccination coverage among foreign-born children is of concern because they live in households and communities characterized by more intense exposure to infectious diseases. Because of their higher prevalence rates, there is an increasing occurrence of infectious diseases imported into developed countries. This case report emphasizes the emerging necessity for new clinicians and pathologists of having competence with old infectious disease pathology. CASE PRESENTATION: A three and a half year old girl, who presented with croup history of 5 days and has been in severe respiratory distress, was admitted to the Pediatric Intensive Care Unit in shock and acute respiratory failure. The patient was immediately intubated, and a grayish nonadherent membrane extending through the glottis down into the larynx was apparent during the procedure. Echocardiographic findings, which were consistent with acute myocarditis, confirmed poor left ventricular contractility despite escalating high doses of inotropes. Autopsy showed numerous strains of toxigenic corynobacterium diphtheriae, which also grew on the Loeffler cultures of membranes received during the intubation. CONCLUSION: It is critical that new generations of clinicians and bio-pathologists not only be trained in the subspecialty of infectious disease pathology, but that they also be willing participants in the diagnosis and investigation of infectious diseases

    What Should Vaccine Developers Ask? Simulation of the Effectiveness of Malaria Vaccines

    A number of different malaria vaccine candidates are currently in pre-clinical or clinical development. Even though they vary greatly in their characteristics, it is unlikely that any of them will provide long-lasting sterilizing immunity against the malaria parasite. There is great uncertainty about what the minimal vaccine profile should be before registration is worthwhile; how to allocate resources between different candidates with different profiles; which candidates to consider combining; and what deployment strategies to consider.We use previously published stochastic simulation models, calibrated against extensive epidemiological data, to make quantitative predictions of the population effects of malaria vaccines on malaria transmission, morbidity and mortality. The models are fitted and simulations obtained via volunteer computing. We consider a range of endemic malaria settings with deployment of vaccines via the Expanded program on immunization (EPI), with and without additional booster doses, and also via 5-yearly mass campaigns for a range of coverages. The simulation scenarios account for the dynamic effects of natural and vaccine induced immunity, for treatment of clinical episodes, and for births, ageing and deaths in the cohort. Simulated pre-erythrocytic vaccines have greatest benefits in low endemic settings (<EIR of 10.5) where between 12% and 14% of all deaths are averted when initial efficacy is 50%. In some high transmission scenarios (>EIR of 84) PEV may lead to increased incidence of severe disease in the long term, if efficacy is moderate to low (<70%). Blood stage vaccines (BSV) are most useful in high transmission settings, and are comparable to PEV for low transmission settings. Combinations of PEV and BSV generally perform little better than the best of the contributing components. A minimum half-life of protection of 2–3 years appears to be a precondition for substantial epidemiological effects. Herd immunity effects can be achieved with even moderately effective (>20%) malaria vaccines (either PEV or BSV) when deployed through mass campaigns targeting all age-groups as well as EPI, and especially if combined with highly efficacious transmission-blocking components.We present for the first time a stochastic simulation approach to compare likely effects on morbidity, mortality and transmission of a range of malaria vaccines and vaccine combinations in realistic epidemiological and health systems settings. The results raise several issues for vaccine clinical development, in particular appropriateness of vaccine types for different transmission settings; the need to assess transmission to the vector and duration of protection; and the importance of deployment additional to the EPI, which again may make the issue of number of doses required more critical. To test the validity and robustness of our conclusions there is a need for further modeling (and, of course, field research) using alternative formulations for both natural and vaccine induced immunity. Evaluation of alternative deployment strategies outside EPI needs to consider the operational implications of different approaches to mass vaccination

    A three year descriptive study of early onset neonatal sepsis in a refugee population on the Thailand Myanmar border.

    BACKGROUND: Each year an estimated four million neonates die, the majority in the first week of life. One of the major causes of death is sepsis. Proving the incidence and aetiology of neonatal sepsis is difficult, particularly in resource poor settings where the majority of the deaths occur. METHODS: We conducted a three year observational study of clinically diagnosed early onset (<7 days of age) neonatal sepsis (EONS) in infants born to mothers following antenatal care at the Shoklo Malaria Research Unit clinic in Maela camp for displaced persons on the Thailand-Myanmar border. Episodes of EONS were identified using a clinical case definition. Conventional and molecular microbiological techniques were employed in order to determine underlying aetiology. RESULTS: From April 2009 until April 2012, 187 infants had clinical signs of EONS, giving an incidence rate of 44.8 per 1000 live births (95% CI 38.7-51.5). One blood culture was positive for Escherichia coli, E. coli was detected in the cerebrospinal fluid specimen in this infant, and in an additional two infants, by PCR. Therefore, the incidence of bacteriologically proven EONS was 0.7 per 1000 live births (95% CI 0.1-2.1). No infants enrolled in study died as a direct result of EONS. CONCLUSION: A low incidence of bacteriologically proven EONS was seen in this study, despite a high incidence of clinically diagnosed EONS. The use of molecular diagnostics and nonspecific markers of infection need to be studied in resource poor settings to improve the diagnosis of EONS and rationalise antibiotic use

    Group B streptococcal carriage, serotype distribution and antibiotic susceptibilities in pregnant women at the time of delivery in a refugee population on the Thai-Myanmar border

    <p>Abstract</p> <p>Background</p> <p>Group B Streptococcus (GBS) is the leading cause of neonatal sepsis in the developed world. Little is known about its epidemiology in the developing world, where the majority of deaths from neonatal infections occur. Maternal carriage of GBS is a prerequisite for the development of early onset GBS neonatal sepsis but there is a paucity of carriage data published from the developing world, in particular South East Asia.</p> <p>Methods</p> <p>We undertook a cross sectional study over a 13 month period in a remote South East Asian setting on the Thai-Myanmar border. During labour, 549 mothers had a combined vaginal rectal swab taken for GBS culture. All swabs underwent both conventional culture as well as PCR for GBS detection. Cultured GBS isolates were serotyped by latex agglutination, those that were negative or had a weak positive reaction and those that were PCR positive but culture negative were additionally tested using multiplex PCR based on the detection of GBS capsular polysaccharide genes.</p> <p>Results</p> <p>The GBS carriage rate was 12.0% (95% CI: 9.4-15.0), with 8.6% positive by both culture and PCR and an additional 3.5% positive by PCR alone. Serotypes, Ia, Ib, II, III, IV, V, VI and VII were identified, with II the predominant serotype. All GBS isolates were susceptible to penicillin, ceftriaxone and vancomycin and 43/47 (91.5%) were susceptible to erythromycin and clindamycin.</p> <p>Conclusions</p> <p>GBS carriage is not uncommon in pregnant women living on the Thai-Myanmar border with a large range of serotypes represented.</p
