    Occurrence of False Positive Results for the Detection of Carbapenemases in Carbapenemase-Negative Escherichia coli and Klebsiella pneumoniae Isolates

    Adequate detection of the production of carbapenemase in Enterobacteriaceae isolates is crucial for infection control measures and the appropriate choice of antimicrobial therapy. In this study, we investigated the frequency of false positive results for the detection of carbapenemases in carbapenemase-negative Escherichia coli and Klebsiella pneumoniae clinical isolates by the modified Hodge test (MHT). Three hundred and one E. coli and K. pneumoniae clinical isolates were investigated. All produced extended spectrum β-lactamases (ESBLs) but were susceptible to carbapenems. Antimicrobial susceptibility testing was performed by the disk diffusion and agar dilution methods. The MHT was performed using the standard inoculum of test organisms recommended by the CLSI. Genes that encoded ESBLs and carbapenemases were identified by PCR and DNA sequencing. Among the 301 clinical isolates, none of the isolates conformed to the criteria for carbapenemase screening recommended by the CLSI. The susceptibility rates for imipenem, meropenem, and ertapenem all were 100.0%, 100.0%, and 100.0%, respectively. Of the 301 E. coli and K. pneumoniae isolates, none produced carbapenemase. The MHT gave a positive result for 3.3% (10/301) of the isolates. False positive results can occur when the MHT is used to detect carbapenemase in ESBL-producing isolates and clinical laboratories must be aware of this fact

This review outlines the main structures, preparation methods, physicochemical properties, potential applications, and future prospects of PPy nanomaterials. The preparation approaches include the soft micellar template method, hard physical template method and templateless method. Due to their excellent electrical conductivity, biocompatibility, environmental stability and reversible redox properties, PPy nanomaterials have potential applications in the fields of energy storage, biomedicine, sensors, adsorption and impurity removal, electromagnetic shielding, and corrosion resistant. Finally, the current difficulties and future opportunities in this research area are discussed

    Pathogenic Implication of a Fibrinogen-Binding Protein of Staphylococcus epidermidis in a Rat Model of Intravascular-Catheter-Associated Infectionâ–¿

    The involvement of Fbe, a fibrinogen-binding protein of Staphylococcus epidermidis, in the pathogenesis of catheter-associated infection was investigated. An fbe (gene encoding Fbe protein) mutant was constructed by allelic replacement, wherein an erythromycin resistance gene replaced a portion of the A region of fbe. Meanwhile, a rat central venous catheter (CVC) infection model was established to assess the importance of Fbe in the pathogenesis of CVC-associated infection due to S. epidermidis. Fbe-positive S. epidermidis strain HB was significantly more likely to cause a CVC-associated infection resulting in bacteremia and metastatic disease than its isogenic Fbe-deficient mutant (100% versus 20%, P < 0.01). These results confirm the importance of adherence associated with Fbe in the pathogenesis of CVC-associated infection caused by S. epidermidis

    Morphology, Crystallization and Thermal Behaviors of PLA-Based Composites: Wonderful Effects of Hybrid GO/PEG via Dynamic Impregnating

    In this paper, a dynamic impregnating device, which can generate supersonic vibration with the vacuum-adsorbing field, was used to prepare the hybrid graphene oxide (GO)/polyethylene glycol (PEG). Interestingly, the hybrid GO/PEG under dynamic impregnating and/or internal mixing was introduced into poly-(lactic acid) (PLA) matrix via melting-compounding, respectively. On one hand, compared with the internal mixing, the hybrid GO/PEG with the different component ratio using dynamic impregnation had a better dispersed morphology in the PLA matrix. On the other hand, compared with the high molecular weight (Mw) of PEG, the hybrid GO/PEG with low Mw of PEG had better an exfoliated morphology and significantly improved the heat distortion temperature (HDT) of the PLA matrix. Binding energies results indicate that low Mw of PEG with GO has excellent compatibility. Dispersed morphologies of the hybrid GO/PEG show that the dynamic impregnating had stronger blending capacity than the internal mixing and obviously improved the exfoliated morphology of GO in the PLA. Crystallization behaviors indicate that the hybrid GO/PEG with the low Mw of PEG based on dynamic impregnating effectively enhanced the crystallinity of PLA, and the cold crystallization character of PLA disappeared in the melting process. Moreover, the storage modulus and loss factor of the PLA-based composites were also investigated and their HDT was improved with the introduction of hybrid GO/PEG. Furthermore, a physical model for the dispersed morphology of the hybrid GO/PEG in the PLA matrix was established. Overall, the unique blending technique of hybrid GO/PEG via dynamic impregnating is an effective approach to enhance the property range of PLA and is suitable for many industrial applications

    Volatile Profiles and Sensory Characteristics of Cabernet Sauvignon Dry Red Wines in the Sub-Regions of the Eastern Foothills of Ningxia Helan Mountain in China

    To elucidate the effects of the different terroir on wine aroma in six sub-regions of Eastern Foothills of Helan Mountain in Ningxia, a premium wine-producing region in China, 71 Cabernet Sauvignon wines were investigated by gas chromatography-mass spectrometry (GC-MS), check-all-that-apply (CATA), and quantitative descriptive analysis (QDA). The bidirectional orthogonal partial least squares-discriminant analysis (O2PLS-DA) results showed that the Cabernet Sauvignon dry red wines from Xixia (XX) and Yongning (YN) had similar volatile profiles due to their geographical proximity and were characterized by higher concentrations of esters, higher alcohols, and volatile phenols because the similar aromatic profiles were detected in their dry red wines. Shizuishan (SZS) and Hongsipu (HSP) wines showed clear differences compared to the wines of the other four sub-regions, being mainly characterized by relatively higher phenolic aldehydes and volatile phenols. The concentrations of methoxypyrazines and norisoprenoids varied mainly depending on the climate diversity of the sub-regions. The highest 3-isobutyl-2-methoxypyrazine (IBMP) concentration was presented in the Helan (HL) wines. The Qingtongxia (QTX) wines have the highest &beta;-damascenone, which might be influenced by the fact that QTX has the lowest effective accumulated temperature and the highest sunshine duration among the five sub-regions. Esters including ethyl octanoate, ethyl decanoate, ethyl butanoate, ethyl hexanoate, and isoamyl acetate were the highest in HL. Additionally, the herbaceous, black berry, and red berry notes in HL and QTX were the most outstanding

    Comparison of adhesin genes and antimicrobial susceptibilities between uropathogenic and intestinal commensal Escherichia coli strains.

    The presence of adhesins is arguably an important determinant of pathogenicity for Uropathogenic Escherichia coli (UPEC). Antimicrobial susceptibilities were tested by agar dilution method, fifteen adhesin genes were detected by polymerase chain reaction, and multilocus sequence typing (MLST) was analyzed in 70 UPEC isolates and 41 commensal E. coli strains. Extended-spectrum β-lactamase (ESBL) was determined with confirmatory test. The prevalence of ESBL-producers in UPEC (53%, 37/70) was higher than the commensal intestinal isolates (7%, 3/41), and 97% (36/37) of the ESBL-producing UPEC harbored bla CTX-M genes. afa was present in 36% (10/28) UPEC isolates from recurrent lower urinary tract infection (UTI), and none in the acute pyelonephritis, acute uncomplicated cystitis or commensal strains (P<0.0001). papG was detected in 28% (20/70) of UPEC isolates, while 5% (2/41) of the commensal strains were papG positive (P = 0.0025), and the prevalence of papG was significantly higher in acute pyelonephritis group (71%) than the other two UTI groups (P<0.0001). The prevalence of flu, yqi, yadN and ygiL was significantly higher in UPEC isolates than in the commensal strains. ESBL-producing UPEC showed a lower prevalence of adhesin genes compared with non-ESBL-producing strains. The MLST profiles were different between UPEC and commensal strains, with ST131 (19%, 13/70) and ST10 (20%, 8/41) being the most common MLSTs, respectively. This study demonstrated that several adhesin genes were more prevalent in UPEC isolates than in commensal E. coli, and afa may be associated with recurrent lower UTI whereas papG is more frequently associated with acute pyelonephritis

    High Performance Anti-Corrosion Coatings of Poly (Vinyl Butyral) Composites with Poly <i>N</i>-(vinyl)pyrrole and Carbon Black Nanoparticles

    Zinc is widely used in battery negative electrodes and steel coatings for automotive industries. The anti-corrosion property of zinc is the most important factor determining the performance and lifetime of the products. In this paper, both size-controlled poly N-(vinyl)pyrrole (PNVPY) nanoparticles and carbon black (CB) nanoparticles were compounded with poly (vinyl butyral) (PVB) binder developing a series of composite coatings covered on the zinc substrates using a spin-coating technique. The morphologies of the surface and cross section of the PNVPY/CB/PVB coatings indicate that the PNVPY and CB nanoparticles are uniformly distributed in the matrix. The corrosion resistance of the composite coatings was tested by electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization in a 3.5% NaCl solution. It is found that the coating with 1.9 wt.% PNVPY and 2.3 wt.% CB nanoparticles shows a remarkably high resistance value (Rc) and corrosion protection efficiency (99.99%). Meanwhile, the immersion results also reveal its superior corrosion resistance. It is considered that the nanoscale dispersion of PNVPY and carbon in PVB matrix and the strong interface action between the nanoparticles and PVB result in the uniform microstructure of the composites which endues the superior corrosion properties of the coatings

    The complete chloroplast genome sequence of Trikeraia hookeri

    Trikeraia hookeri is an alpine grass with significant ecological value. Here, the complete chloroplast genome sequence of T. hookeri using Illumina sequencing data was reported. The size of the whole cp genome was 137,696 bp in length, consisting of a pair of inverted repeats (IR 13,755 bp), a large single-copy region (LSC 81,613 bp), and a small single-copy region (SSC 28,568 bp). The T. hookeri chloroplast genome encodes 119 genes: 81 mRNA genes, 34 tRNA genes and 4 rRNA genes. The GC content of T. hookeri chloroplast genome was 38.8% and those in LSC, SSC, and IR regions were 36.9, 40.8, and 42.3%, respectively. The maximum-likelihood phylogenetic analysis demonstrated that T. hookeri was most closely related to Stipa lipskyi (NC028444) and Stipa purpurrea (NC029390). Our findings provide fundamental information for further evolutionary and phylogenetic researches of T. hookeri
