    目的  通过分析报告开腹胆囊切除加胆总管切开取石术后诱发甲状腺危象1例,以期引起临床医师的注意,提高诊疗质量。方法  全面汇报患者病例资料及诊疗过程,分析其可能的发生原因。结果  非甲状腺手术诱发甲状腺危象虽不多见,但仍有发生。结论  临床医师应提高意识,详细术前检查,术后短时间内突然出现无法解释的心律失常,血压升高等症状的患者,应考虑甲状腺危象的发生。Objective: To attract the attention of the clinician and to improve the quality of diagnosis by reporting a case of thyroid crisis after cholecystectomy and choledocholithotomy. Methods: Comprehensive report and the process of diagnosis and treatment in patients with clinical data, analyze the possible causes of the incident. Result: It is a rare condition that has no specific clinical manifestations, is difficult to diagnose at the early stage. Conclusion: The clinician should raise awareness and consider thyroid crisis as a possible factor that results in the sudden appearance of arrhythmia, high blood pressure and other symptoms after the operation

    Objective:To explore the effect of yinxingdamo on serum Complement C3、interleukin-6,8 in Patients with Acute Cerebral Infarction(ACI).Methods:136 cases of ACI were equally and randomly divided into two groups,one is the control group,the other group is having both routine treatment and yinxingdamo treatment for 4 weeks. Both the patients of two groups were respectively observed the serum Complement C3、interleukin-6,8 before and after treatment.Meantime,another 40 cases of normal subjects are as reference.Results:Before the treatment,the serum Complement C3、interleukin-6,8 levels of patients with ACI were obviously higher than the reference group(P<0.05),but after treatment,the levels of the treatment group were obviously lower than before.The difference was statistically significant (P<0.05).Conclusion:The treatment of Yinxingdamo injection can lower the level of the serum Complement C3、interleukin-6,8 in Patients with ACI,causing a significant effect of anti-inflammatory.目的  探讨银杏达莫对急性脑梗死(ACI)患者血清补体C3、白细胞介素-6(IL-6)及白细胞介素-8(IL-8)水平变化的影响。方法 将ACI患者136例随机分为常规治疗对照组68例和银杏达莫治疗+常规治疗治疗组68例,分别治疗前及治疗4周后测定血清补体C3、IL-6及IL-8。同期另选40例健康体检者作参考组。结果  ACI患者治疗前血清补体C3、IL-6及IL-8水平均明显高于参考组(P<0.05)。治疗组经过治疗后血清补体C3、IL-6及IL-8水平均明显低于治疗前,并且更低于对照组,差异具有统计学意义(P<0.05);与参考组比较无显著性差异(P<0.05)。对照组患者治疗前后血清补体C3、IL-6及IL-8无显著性差异(P<0.05)。结论  ACI患者应用银杏达莫治疗使补体C3、IL-6及IL-8显著降低,具有明显抗炎作用效果。

    Illuminating cell signaling with genetically encoded FRET biosensors in adult mouse cardiomyocytes.

    FRET-based biosensor experiments in adult cardiomyocytes are a powerful way of dissecting the spatiotemporal dynamics of the complicated signaling networks that regulate cardiac health and disease. However, although much information has been gleaned from FRET studies on cardiomyocytes from larger species, experiments on adult cardiomyocytes from mice have been difficult at best. Thus the large variety of genetic mouse models cannot be easily used for this type of study. Here we develop cell culture conditions for adult mouse cardiomyocytes that permit robust expression of adenoviral FRET biosensors and reproducible FRET experimentation. We find that addition of 6.25 µM blebbistatin or 20 µM (S)-nitro-blebbistatin to a minimal essential medium containing 10 mM HEPES and 0.2% BSA maintains morphology of cardiomyocytes from physiological, pathological, and transgenic mouse models for up to 50 h after adenoviral infection. This provides a 10-15-h time window to perform reproducible FRET readings using a variety of CFP/YFP sensors between 30 and 50 h postinfection. The culture is applicable to cardiomyocytes isolated from transgenic mouse models as well as models with cardiac diseases. Therefore, this study helps scientists to disentangle complicated signaling networks important in health and disease of cardiomyocytes

    The chlamydial periplasmic stress response serine protease cHtrA is secreted into host cell cytosol

    <p>Abstract</p> <p>Background</p> <p>The periplasmic High Temperature Requirement protein A (HtrA) plays important roles in bacterial protein folding and stress responses. However, the role of chlamydial HtrA (cHtrA) in chlamydial pathogenesis is not clear.</p> <p>Results</p> <p>The cHtrA was detected both inside and outside the chlamydial inclusions. The detection was specific since both polyclonal and monoclonal anti-cHtrA antibodies revealed similar intracellular labeling patterns that were only removed by absorption with cHtrA but not control fusion proteins. In a Western blot assay, the anti-cHtrA antibodies detected the endogenous cHtrA in Chlamydia-infected cells without cross-reacting with any other chlamydial or host cell antigens. Fractionation of the infected cells revealed cHtrA in the host cell cytosol fraction. The periplasmic cHtrA protein appeared to be actively secreted into host cell cytosol since no other chlamydial periplasmic proteins were detected in the host cell cytoplasm. Most chlamydial species secreted cHtrA into host cell cytosol and the secretion was not inhibitable by a type III secretion inhibitor.</p> <p>Conclusion</p> <p>Since it is hypothesized that chlamydial organisms possess a proteolysis strategy to manipulate host cell signaling pathways, secretion of the serine protease cHtrA into host cell cytosol suggests that the periplasmic cHtrA may also play an important role in chlamydial interactions with host cells.</p

    BMP9-Induced Osteogenetic Differentiation and Bone Formation of Muscle-Derived Stem Cells

    Efficient osteogenetic differentiation and bone formation from muscle-derived stem cells (MDSCs) should have potential clinical applications in treating nonunion fracture healing or bone defects. Here, we investigate osteogenetic differentiation ability of MDSCs induced by bone morphogenetic protein 9 (BMP9) in vitro and bone formation ability in rabbit radius defects repairing model. Rabbit's MDSCs were extracted by type I collagenase and trypsin methods, and BMP9 was introduced into MDSCs by infection with recombinant adenovirus. Effects of BMP9-induced osteogenetic differentiation of MDSCs were identified with alkaline phosphatase (ALP) activity and expression of later marker. In stem-cell implantation assay, MDSCs have also shown valuable potential bone formation ability induced by BMP9 in rabbit radius defects repairing test. Taken together, our findings suggest that MDSCs are potentiated osteogenetic stem cells which can be induced by BMP9 to treat large segmental bone defects, nonunion fracture, and/or osteoporotic fracture
