Tubgcp3 Is Required for Retinal Progenitor Cell Proliferation During Zebrafish Development
The centrosomal protein γ-tubulin complex protein 3 (Tubgcp3/GCP3) is required for the assembly of γ-tubulin small complexes (γ-TuSCs) and γ-tubulin ring complexes (γ-TuRCs), which play critical roles in mitotic spindle formation during mitosis. However, its function in vertebrate embryonic development is unknown. Here, we generated the zebrafish tubgcp3 mutants using the CRISPR/Cas9 system and found that the tubgcp3 mutants exhibited the small eye phenotype. Tubgcp3 is required for the cell cycle progression of retinal progenitor cells (RPCs), and its depletion caused cell cycle arrest in the mitotic (M) phase. The M-phase arrested RPCs exhibited aberrant monopolar spindles and abnormal distributed centrioles and γ-tubulin. Moreover, these RPCs underwent apoptosis finally. Our study provides the in vivo model for the functional study of Tubgcp3 and sheds light on the roles of centrosomal γ-tubulin complexes in vertebrate development
Molecular cloning and analysis of zebrafish voltage-gated sodium channel beta subunit genes: implications for the evolution of electrical signaling in vertebrates
<p>Abstract</p> <p>Background</p> <p>Action potential generation in excitable cells such as myocytes and neurons critically depends on voltage-gated sodium channels. In mammals, sodium channels exist as macromolecular complexes that include a pore-forming alpha subunit and 1 or more modulatory beta subunits. Although alpha subunit genes have been cloned from diverse metazoans including flies, jellyfish, and humans, beta subunits have not previously been identified in any non-mammalian species. To gain further insight into the evolution of electrical signaling in vertebrates, we investigated beta subunit genes in the teleost <it>Danio rerio </it>(zebrafish).</p> <p>Results</p> <p>We identified and cloned single zebrafish gene homologs for beta1-beta3 (<it>zbeta1-zbeta3</it>) and duplicate genes for beta4 (<it>zbeta4.1, zbeta4.2</it>). Sodium channel beta subunit loci are similarly organized in fish and mammalian genomes. Unlike their mammalian counterparts, <it>zbeta1 </it>and <it>zbeta2 </it>subunit genes display extensive alternative splicing. Zebrafish beta subunit genes and their splice variants are differentially-expressed in excitable tissues, indicating tissue-specific regulation of <it>zbeta1-4 </it>expression and splicing. Co-expression of the genes encoding zbeta1 and the zebrafish sodium channel alpha subunit Nav1.5 in Chinese Hamster Ovary cells increased sodium current and altered channel gating, demonstrating functional interactions between zebrafish alpha and beta subunits. Analysis of the synteny and phylogeny of mammalian, teleost, amphibian, and avian beta subunit and related genes indicated that all extant vertebrate beta subunits are orthologous, that beta2/beta4 and beta1/beta3 share common ancestry, and that beta subunits are closely related to other proteins sharing the V-type immunoglobulin domain structure. Vertebrate sodium channel beta subunit genes were not identified in the genomes of invertebrate chordates and are unrelated to known subunits of the <it>para </it>sodium channel in <it>Drosophila</it>.</p> <p>Conclusion</p> <p>The identification of conserved orthologs to all 4 voltage-gated sodium channel beta subunit genes in zebrafish and the lack of evidence for beta subunit genes in invertebrate chordates together indicate that this gene family emerged early in vertebrate evolution, prior to the divergence of teleosts and tetrapods. The evolutionary history of sodium channel beta subunits suggests that these genes may have played a key role in the diversification and specialization of electrical signaling in early vertebrates.</p
A general model for collaboration networks
In this paper, we propose a general model for collaboration networks.
Depending on a single free parameter "{\bf preferential exponent}", this model
interpolates between networks with a scale-free and an exponential degree
distribution. The degree distribution in the present networks can be roughly
classified into four patterns, all of which are observed in empirical data. And
this model exhibits small-world effect, which means the corresponding networks
are of very short average distance and highly large clustering coefficient.
More interesting, we find a peak distribution of act-size from empirical data
which has not been emphasized before of some collaboration networks. Our model
can produce the peak act-size distribution naturally that agrees with the
empirical data well.Comment: 10 pages, 10 figure
Solutions to cultural, organizational, and technical challenges in developing PECAS models for the cities of Shanghai, Wuhan, and Guangzhou
Massive construction of transportation infrastructure and fast growth of private car ownership have brought unprecedented changes in land use and transportation systems to cities and regions in many developing countries. Traditional “four-step” travel demand models, which are not designed to assess transport policies under the case of rapid land-use change, cannot be used to achieve coordinated planning of transport and land use. Therefore, there is a pressing need to develop and use integrated land-use transport models (ILUTMs), which consider interactions among socioeconomic activities, urban land use, and transportation development, for policy analysis and for guiding the progressive urbanization process taking place in many parts of these countries. In light of this, efforts have been invested in developing production, exchange, and consumption allocation system (PECAS) models for the cities of Shanghai, Wuhan, and Guangzhou in mainland China. This paper presents the cultural, organizational, and technical challenges encountered in the development of PECAS models for the cities of Shanghai, Wuhan, and Guangzhou and the mitigating solutions from the development teams for taking up or working around them. The solutions and discussions presented in this paper should be interesting to researchers and practitioners for developing ILUTMs in the context of a developing country like China
Droplet-like Fermi surfaces in the anti-ferromagnetic phase of EuFeAs, an Fe-pnictide superconductor parent compound
Using angle resolved photoemission it is shown that the low lying electronic
states of the iron pnictide parent compound EuFeAs are strongly
modified in the magnetically ordered, low temperature, orthorhombic state
compared to the tetragonal, paramagnetic case above the spin density wave
transition temperature. Back-folded bands, reflected in the orthorhombic/
anti-ferromagnetic Brillouin zone boundary hybridize strongly with the
non-folded states, leading to the opening of energy gaps. As a direct
consequence, the large Fermi surfaces of the tetragonal phase fragment, the low
temperature Fermi surface being comprised of small droplets, built up of
electron and hole-like sections. These high resolution ARPES data are therefore
in keeping with quantum oscillation and optical data from other undoped
pnictide parent compounds.Comment: 4 figures, 6 page
Determination of the gamma emission probabilities of 239
239Np is an important nuclide as the decay daughter of 239U and it decays to 239Pu by emitting beta particles and gamma rays with a half life of 2.356 days. The data of the emission probabilities of its gamma-rays in the open references are consistent except for the main gamma-ray of 106.1 keV, the emission probability of which varies from 25.9% to 27.2%. To verify the emission probability of 106.1 keV gamma-ray of 239Np, a N-type coaxial HPGe detector was calibrated using 241Am, 133Ba, 60Co, 152Eu and 155Eu reference gamma sources to get the accurate efficiency of the 106.1 keV gamma-ray. 239Np was purified from solution containing 243Am, where 239Np is the alpha decay daughter of 243Am. The specific activity of 239Np solution was determined by a 4πβ (PC)-γ coincidence counting device. There were 6 gamma sources prepared to measure with the HPGe detector, and the activity of 239Np in each gamma source was calculated with the weights of the solution contained in it. The emission probability of 106.1 keV of 239Np is measured to be (25.4 ± 0.3)%, which is consistent with 25.34%, the value evaluated in 2014
Pairing symmetry and properties of iron-based high temperature superconductors
Pairing symmetry is important to indentify the pairing mechanism. The
analysis becomes particularly timely and important for the newly discovered
iron-based multi-orbital superconductors. From group theory point of view we
classified all pairing matrices (in the orbital space) that carry irreducible
representations of the system. The quasiparticle gap falls into three
categories: full, nodal and gapless. The nodal-gap states show conventional
Volovik effect even for on-site pairing. The gapless states are odd in orbital
space, have a negative superfluid density and are therefore unstable. In
connection to experiments we proposed possible pairing states and implications
for the pairing mechanism.Comment: 4 pages, 1 table, 2 figures, polished versio
