    TITAN: A Spatiotemporal Feature Learning Framework for Traffic Incident Duration Prediction

    Critical incident stages identification and reasonable prediction of traffic incident duration are essential in traffic incident management. In this paper, we propose a traffic incident duration prediction model that simultaneously predicts the impact of the traffic incidents and identifies the critical groups of temporal features via a multi-task learning framework. First, we formulate a sparsity optimization problem that extracts low-level temporal features based on traffic speed readings and then generalizes higher level features as phases of traffic incidents. Second, we propose novel constraints on feature similarity exploiting prior knowledge about the spatial connectivity of the road network to predict the incident duration. The proposed problem is challenging to solve due to the orthogonality constraints, non-convexity objective, and non-smoothness penalties. We develop an algorithm based on the alternating direction method of multipliers (ADMM) framework to solve the proposed formulation. Extensive experiments and comparisons to other models on real-world traffic data and traffic incident records justify the efficacy of our model

    Ultraviolet Light Responsive N‐Nitroso Polymers for Antibacterial Nitric Oxide Delivery

    AbstractThis study investigates the incorporation of active secondary amine moieties into the polymer backbone by co‐polymerizing 2,4,6‐tris(chloromethyl)‐mesitylene (TCM) with three diamines, namely 1,4‐diaminobutane (DAB), m‐phenylenediamine (MPD), and p‐phenylenediamine (PPD). This process results in the stabilisation of the amine moieties and the subsequently introduced nitroso groups. Charging bioactive nitric oxide (NO) into the polymers is accomplished by converting the amine moieties into N‐nitroso groups. The ability of the polymers to store and release NO depends on their structures, particularly the amounts of incorporated active secondary amines. With grafting photosensitive N‐nitroso groups into the polymers, the derived NO@polymers exhibit photoresponsivity. NO release is completely regulated by adjusting UV light irradiation. These resulting polymeric NO donors demonstrate remarkable bactericidal and bacteriostatic activity, effectively eradicating E. coli bacteria and inhibiting their growth. The findings from this study hold promising implications for combining NO delivery with phototherapy in various medical applications.This article is protected by copyright. All rights reserve

    Changes on the conformational and functional properties of soybean protein isolate induced by quercetin

    The conformational changes and functional properties of SPI induced by quercetin was investigated via fourier transform infrared (FTIR) spectroscopy, fluorescence spectroscopy, circular dichroism (CD) spectroscopy and molecular docking. A decrease in the fluorescence intensity and a blue shift in the maximum wavelength were observed due to the binding process with fluorescent residues. The analysis of Stern-Volmer equation showed that the fluorescence quenching induced by quercetin took the form of static quenching, and the binding stoichiometry between SPI and quercetin was 1:1. The values of ΔH and ΔS were both positive illustrating that hydrophobic interaction was the primary binding force between quercetin and SPI. Results of FTIR and CD indicated that the binding with quercetin changed the secondary structure of SPI, resulting in a partially unfolded and more flexible structure. SDS-PAGE confirmed there was no covalent interaction between the two constituents. Molecular docking demonstrated that there were stable configurations and high matching degrees in both 11S and 7S proteins with quercetin via hydrogen bonds and hydrophobic interactions. Meanwhile, modification by quercetin enhanced the foaming and emulsifying capacities of SPI.     Diminished but Not Abolished Effect of Two His351 Mutants of Anthrax Edema Factor in a Murine Model

    Edema toxin (ET), which is composed of a potent adenylate cyclase (AC), edema factor (EF), and protective antigen (PA), is one of the major toxicity factors of Bacillus anthracis. In this study, we introduced mutations in full-length EF to generate alanine EF(H351A) and arginine EF(H351R) variants. In vitro activity analysis displayed that the adenylyl cyclase activity of both the mutants was significantly diminished compared with the wild-type EF. When the native and mutant toxins were administered subcutaneously in a mouse footpad edema model, severe acute swelling was evoked by wild-type ET, while the symptoms induced by mutant toxins were very minor. Systemic administration of these EF variants caused non-lethal hepatotoxicity. In addition, EF(H351R) exhibited slightly higher activity in causing more severe edema than EF(H351A). Our findings demonstrate that the toxicity of ET is not abolished by substitution of EF residue His351 by alanine or arginine. These results also indicate the potential of the mouse footpad edema model as a sensitive method for evaluating both ET toxicity and the efficacy of candidate therapeutic agents

    Evaluation of Pollution Level, Spatial Distribution, and Ecological Effects of Antimony in Soils of Mining Areas: A Review

    The first global-scale assessment of Sb contamination in soil that was related to mining/smelting activities was conducted based on 91 articles that were published between 1989 and 2021. The geographical variation, the pollution level, the speciation, the influencing factors, and the environmental effects of Sb that were associated with mining/smelting-affected soils were analyzed. The high Sb values mainly occurred in developed (Poland, Italy, Spain, Portugal, New Zealand, Australia) and developing (China, Algeria, Slovakia) countries. Sb concentrations of polluted soil from mining areas that were reported in most countries significantly exceeded the maximum permissible limit that is recommended by WHO, except in Turkey and Macedonia. The soil Sb concentrations decreased in the order of Oceania (29,151 mg/kg) > North Africa (13,022 mg/kg) > Asia (1527 mg/kg) > Europe (858 mg/kg) > South America (37.4 mg/kg). The existing extraction methods for Sb speciation have been classified according to the extractant, however, further research is needed in the standardization of these extraction methods. Modern analytical and characterization technologies, e.g., X-ray absorption spectroscopy, are effective at characterizing chemical speciation. Conditional inference tree (CIT) analysis has shown that the clay content was the major factor that influenced the soil Sb concentration. Non-carcinogenic risks to the public from soil Sb pollution were within the acceptable levels in most regions. An Sb smelter site at the Endeavour Inlet in New Zealand, an abandoned open-pit Sb mine in Djebel Hamimat, Algeria, an old Sb-mining area in Tuscany, Italy, and Hillgrove mine in Australia were selected as the priority control areas. Cynodon dactylon, Boehmeria, Pteris vittata, and Amaranthus paniculatus were found to be potential Sb accumulators. All of the values of bioaccumulation factors for the crops were less than one. However, ingestion of Sb through crop consumption posed potential non-carcinogenic health risks, which should not be neglected. The soil variables (pH, Eh, total sulfur, carbon nitrogen ratio, total organic carbon, and sulfate), the total Sb and the bioavailable Sb, and heavy metal(loid)s (As, Pb, and Fe) were the major parameters affecting the microbial community compositions

    Evaluating the Ecological Services of Roof Greening Plants in Beijing Based on Functional Traits

    Selecting suitable species to enhance ecological functions is crucial for improvements in the planning and design of roof greening and in maintaining sustainable urban development, especially in rapidly urbanized areas. Assisted by field trips to enhance studies, the present project assessed the ecological functions of 207 plant species used for roof greening in Beijing based on their key functional traits. The results indicate that regulating, cultural, supplying, and supporting functions differed significantly among species and families in the study area. Rosaceae species have higher levels of overall ecological functions than other species, and a large number of Compositae species have lower-level functions. Compared to other families, Araliaceae and Nyctaginaceae have higher mean values of cultural and supporting functions and the highest mean overall function value of 37. Ulmaceae, Sapindaceae, Ginkgoaceae, Berberidaceae, and Aceraceae have higher mean regulating, cultural, supporting, and overall function values. Amaranthaceae, Umbelliferae, Lamiaceae, Saxifragaceae, Ericaceae, and Gramineae have lower values. The existing roof greening in Beijing includes some pitfalls with respect to plant composition as well as plant selection that does not consider ecological functions. The following measures could be proposed to increase ecological functions: (1) Increasing the number of plants with shallow roots and with strong adaptation traits to roof conditions; (2) Enriching ecological communities with diverse plants with high ecological functions; and (3) Carrying out rational ecological planning and management based on detailed and objective data on plant species. Future studies should focus on specifying plant functional traits to enhance ecological functions

    Whole-transcriptome analysis highlights adenylyl cyclase toxins-derived modulation of NF-ÎșB and ERK1/2 pathways in macrophages

    Edema toxin (ET), one of the main toxic factors of Bacillus anthracis (B. anthracis), is a kind of potent adenylate cyclase (AC). B. anthracis has adapted to resist macrophage microbicidal mechanisms in part by secreting ET. To date, there is limited information on the pathogenic mechanisms used by ET to manipulate macrophage function, especially at the transcriptome level. We used RNA sequencing to study transcriptional changes in RAW264.7 cells treated with ET. We aimed to identify molecular events associated with the establishment of infection and followed changes in cellular proteins. Our results indicate that ET inhibited TNF-α expression in the RAW264.7 mouse macrophage cell line by activating the cAMP/PKA pathway. ET challenge of macrophages induced a differential expression of genes that participate in multiple macrophage effector functions such as cytokine production, cell adhesion, and the inflammatory response. Furthermore, ET influenced the expression of components of the ERK1/2, as well as the NF-αB signaling pathways. We also showed that ET treatments inhibit the phosphorylation of the ERK1/2 protein. ET also attenuated NF-αB subunit p65 phosphorylation and transcriptional activity of NF-αB via the cAMP/PKA pathway in macrophages. Since the observed modulatory effects were characteristic only of the bacterial exotoxin ET, we propose this may be a mechanism used by B. anthracis to manipulate macrophages and establish systemic infection

    The Impact of Study Setting on Clinical Characteristics in Older Chinese Adults with Subjective Cognitive Decline: Baseline Investigation of Convenience and Population-Based Samples

    Background. Subjective cognitive decline (SCD) is the earliest symptom stage of Alzheimer’s disease (AD). Previous studies have shown that the study setting is an important influence factor of SCD. However, the effect of this factor among a Chinese population with SCD is not clear. Here, we aim to compare the clinical characteristics of SCD between a convenience and a population-based sample in China. Methods. We included a convenience sample of 212 SCD subjects and a population-based sample of 110 SCD subjects. We performed univariate analysis to evaluate the between-group differences in sociodemographic characteristics, neuropsychological performance, psychiatric conditions, different cognitive domains, and the SCD-plus criteria. Multiple linear regression model was established, adjusted for sex, age, and education, and compared the neuropsychological performance between the groups. Results. The convenience sample had more years of education, a higher family history of dementia, and higher neuropsychological and anxiety depression score than the population-based sample. Using sex, age, education, group as the independent variables, and neuropsychological score as the dependent variable, multiple linear regression model was established; a statistically significant neuropsychological score difference (MoCA-B, AVLT-H-N4, AVLT-H-N5, AVLT-H-N7, AFT, and STT-B) was found between the two samples. In the SCD cognitive domains, the population-based sample had more complaints about declines in their language and planning domains. For SCD-plus criteria in memory domain, the convenience sample had more complaints, worry, and cognitive decline within the last 5 years, along with medical help-seeking. Conclusion. There were some different characteristics among SCD individuals between convenience samples and population-based samples in China
