    Impact of body dose parameters on circulating immune cells in locally advanced nasopharyngeal carcinoma patients: a retrospective cohort study

    Background and purpose: The implementation of intensity-modulated radiotherapy (IMRT) has significantly enhanced the survival outcomes for patients with nasopharyngeal carcinoma (NPC). However, this therapeutic approach still falls short in meeting the prognostic requirements of individuals with locally advanced NPC (LANPC). Therefore, it is imperative to identify effective prognostic markers to enhance the efficacy of radiotherapy and achieve personalized treatment. Given the potential predictive value demonstrated in previous studies regarding radiotherapy-related body dose parameters and immune blood cells, this study aimed to investigate the correlation between body dose parameters and reduced immune cells and patient prognosis during radiotherapy in LANPC patients. Methods: Clinical data of 423 patients with LANPC (stage Ⅲ-Ⅳa) treated in Fudan University Shanghai Cancer Center from Jan.1, 2012 to Dec. 31, 2016 were retrospectively analyzed. Percentage changes of different immune blood cells during radiotherapy were also collected. Cox proportional hazard model was used to determine prognostic factors for overall survival (OS), locoregional recurrence-free survival (LRFS) and distant metastasis-free survival (DMFS). Body dose-based parameters were extracted from dose-volume histograms (DVHs). Logistic regression was applied to determine parameters that could predict white blood cells reduction. Results: High ΔLYM% (ΔLYM%≥7 7.0%) and high ΔMONO% (ΔMONO%≥2 8.5%) were identified as two adverse prognostic factors in LANPC patients. In multivariable analysis, high ΔLYM% was found to be a significant predictor of worse OS (HR = 1.672, P = 0.012), LRFS (HR = 1.712, P = 0.006), and DMFS (HR = 1.971, P = 0.001). High ΔMONO% was associated with worse OS (HR = 1.355, P = 0.015) and DMFS (HR = 1.704, P = 0.003). The change of ΔLYM% was influenced by the integral body dose (IBD) (OR = 1.004, P = 0.037) and body V60 (OR = 1.046, P = 0.036). ΔMONO% was significantly affected by body V55 (OR = 1.144, P = 0.009) and V70 (OR = 0.734, P = 0.022). Conclusion: Integral body dose, V60, and V55, V70 can serve as dose-volume constraints to retain sufficient immune cell populations to improve prognosis

    Exploring the plankton bacteria diversity and distribution patterns in the surface water of northwest pacific ocean by metagenomic methods

    The study of marine microbial communities is crucial for comprehending the distribution patterns, adaptations to the environment, and the functioning of marine microorganisms. Despite being one of the largest biomes on Earth, the bacterioplankton communities in the Northwest Pacific Ocean (NWPO) remain understudied. In this research, we aimed to investigate the structure of the surface bacterioplankton communities in different water masses of the NWPO. We utilized metagenomic sequencing techniques and cited previous 16S rRNA data to explore the distribution patterns of bacterioplankton in different seasons. Our results revealed that Cyanobacteria, Proteobacteria, Bacteroidetes, and Actinobacteria dominated the microbial communities, accounting for over 95% of the total. During spring, we observed significant differentiation in community structure between the different water masses. For instance, Prochlorococcus and Pseudoalteromonas were primarily distributed in the nutrient-deficient subtropical countercurrent zone, while Flavobacteriaceae and Rhodobacteraceae were found in the Kuroshio-Oyashio mixing zone. During summer, the surface planktonic bacteria communities became homogenized across regions, with Cyanobacteria becoming the dominant group (68.6% to 84.9% relative abundance). The metabolic processes of the microorganisms were dominated by carbohydrate metabolism, followed by amino acid transport and metabolism. However, there was a low relative abundance of functional genes involved in carbohydrate metabolism in the Kuroshio-Oyashio mixing zone. The metagenomic data had assembled 37 metagenomic-assembled genomes (MAGs), which belong to Proteobacteria, Bacteroidetes, and Euryarchaeota. In conclusion, our findings highlight the diversity of the surface bacterioplankton community composition in the NWPO, and its distinct geographic distribution characteristics and seasonal variations

    Association between daily screen time and risk of stroke among middle-aged and elderly people: research based on China health and nutrition survey

    BackgroundWe aimed to explore the independent associations between screen time and the risk of stroke among Chinese adults based on the China Health and Nutrition Survey (CHNS).MethodsData on Chinese adults aged older than 40 years from the CHNS in during 2004–2009 were selected. A total of 4,587 individuals were included in 2009, including screen time and the risk of stroke. Simultaneously, we traced the previous screen time to 2004 for those with outcome measures in 2009 (n = 2,100). Basic information, lifestyle, and screen behavior were obtained through face-to-face interviews and self-completed questionnaires. Anthropometric data collected included blood pressure, body weight, height, hip circumference, and waist circumference. Fasting blood was obtained for measurements of lipid and glucose levels. Cross-sectional analysis and cohort analysis were both performed using multivariate logistic regression.ResultsOf all participants, 3,004 (65.49%) participants spent more than 2 h per day on screen time. Taking the men who spent less than 2 h on screen per day as reference, the crude odds ratio (OR) of the high risk of stroke was 1.53 [95% confidence interval (CI), 1.20–1.95] for the men who spent 2–3 h per day on screen and 2.37 (95% CI, 1.78–3.16) for the men who spent more than 3 h per day on screen. This difference remained significant after adjusting for confounding factors. No association was observed among women. However, in the cohort analysis with screen time in 2006 as the independent variable, the association between screen time and stroke risk was found both in men [OR, 1.83 (95% CI, 1.19–2.82)] and women [OR, 1.48 (95% CI, 1.10–1.99)]).ConclusionWe found that the high screen time was associated with an increased stroke risk, which was pronounced in men, warranting a universal need to limit screen time in order to improve health

    Identification, Characterization, and Expression of a Novel P450 Gene Encoding CYP6AE25 from the Asian Corn Borer, Ostrinia furnacalis

    An allele of the cytochrome P450 gene, CYP6AE14, named CYP6AE25 (GenBank accession no. EU807990) was isolated from the Asian com borer, Ostrinia fumacalis (Guenée) (Lepidoptera: Pyralidae) by RT-PCR. The cDNA sequence of CYP6AE25 is 2315 bp in length and contains a 1569 nucleotides open reading frame encoding a putative protein with 523 amino acid residues and a predicted molecular weight of 59.95 kDa and a theoretical pI of 8.31. The putative protein contains the classic heme-binding sequence motif F××G×××C×G (residues 451–460) conserved among all P450 enzymes as well as other characteristic motifs of all cytochrome P450s. It shares 52% identity with the previously published sequence of CYP6AE14 (GenBank accession no. DQ986461) from Helicoverpa armigera. Phylogenetic analysis of amino acid sequences from members of various P450 families indicated that CYP6AE25 has a closer phylogenetic relationship with CYP6AE14 and CYP6B1 that are related to metabolism of plant allelochemicals, CYP6D1 which is related to pyrethroid resistance and has a more distant relationship to CYP302A1 and CYP307A1 which are related to synthesis of the insect molting hormones. The expression level of the gene in the adults and immature stages of O. furnacalis by quantitative real-time PCR revealed that CYP6AE25 was expressed in all life stages investigated. The mRNA expression level in 3rd instar larvae was 12.8- and 2.97-fold higher than those in pupae and adults, respectively. The tissue specific expression level of CYP6AE25 was in the order of midgut, malpighian tube and fatty body from high to low but was absent in ovary and brain.     Vitamin B12 Levels in Methamphetamine Addicts

    Objective: It has been established that reduced vitamin B12 serum levels are associated with cognitive decline and mental illness. The chronic use of methamphetamine (MA), which is a highly addictive drug, can induce cognitive impairment and psychopathological symptoms. There are few studies addressing the association of MA with vitamin B12 serum levels. This study examined whether the serum levels of B12 are associated with MA addiction.Methods: Serum vitamin B12, homocysteine (Hcy), glucose and triglyceride concentrations were measured in 123 MA addicts and 108 controls. In addition, data were collected on their age, marital status, level of education and Body Mass Index (BMI) for all participants. In the patient group, the data for each subject were collected using the Fagerstrom Test for Nicotine Dependence (FTND), the Alcohol Use Disorders Identification Test (AUDIT), and a drug use history, which included the age of onset, total duration of MA use, the number of relapses and addiction severity.Results: Our results showed that MA addicts had lower vitamin B12 levels (p < 0.05) than those of healthy controls, but Hcy levels were not significantly different between the two groups (p > 0.05). Serum B12 levels were negatively correlated with the number of relapses in the MA group. Furthermore, binary logistics regression analysis indicated that the B12 was an influencing factor contributing to addiction severity.Conclusion: The findings of this study suggest that some MA addicts might have vitamin B12 deficiency, and serum B12 levels may be involved in the prognosis of MA addiction

    Associations between genetic variations in the FURIN gene and hypertension

    <p>Abstract</p> <p>Background</p> <p>Hypertension is a complex disease influenced by multiple genetic and environmental factors. The Kazakh ethnic group is characterized by a relatively high prevalence of hypertension. Previous research indicates that the FURIN gene may play a pivotal role in the renin-angiotensin system and maintaining the sodium-electrolyte balance. Because these systems influence blood pressure regulation, we considered FURIN as a candidate gene for hypertension. The purpose of this study was to systematically investigate the association between genetic variations in the FURIN gene and essential hypertension in a Xinjiang Kazakh population.</p> <p>Methods</p> <p>We sequenced all exons and the promoter regions of the FURIN gene in 94 hypertensive individuals to identify genetic variations associated with the disorder. Genotyping was performed using the TaqMan polymerase chain reaction method for four representative common single nucleotide polymorphisms (SNPs, -7315C > T, 1970C > G, 5604C > G, 6262C > T) in 934 Kazakh Chinese people. One SNP (1970C > G) was replicated in 1,219 Uygur Chinese people.</p> <p>Results</p> <p>Nine novel and seven known single nucleotide polymorphisms were identified in the FURIN gene. The results suggest that 1970C > G was associated with a hypertension phenotype in Kazakh Chinese (additive model, <it>P </it>= 0.091; dominant model, <it>P = </it>0.031, allele model, <it>P </it>= 0.030), and after adjustment with logistic regression analysis, ORs were 1.451 (95%CI 1.106-1.905, <it>P </it>= 0.008) and 1.496 (95% 1.103-2.028, <it>P </it>= 0.01) in additive and dominant models, respectively. In addition, the association between 1970C > G and hypertension was replicated in Uygur subjects (additive model, <it>P </it>= 0.042; dominant model, <it>P </it>= 0.102; allele model, <it>P </it>= 0.027) after adjustment in additive and dominant models, ORs were 1.327 (95% 1.07-1.646), <it>P </it>= 0.01 and 1.307 (95%CI 1.015-1.681, <it>P </it>= 0.038), respectively. G allele carriers exhibited significant lower urinary Na<sup>+ </sup>excretion rate than non-carriers in the Kazakh Chinese population (152.45 ± 76.04 uM/min vs 173.33 ± 90.02 uM/min, <it>P </it>= 0.007).</p> <p>Conclusion</p> <p>Our results suggest that the FURIN gene may be a candidate gene involved in human hypertension, and that the G allele of 1970C > G may be a modest risk factor for hypertension in Xinjiang Kazakh and Uygur populations.</p

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Enterprise Low-Carbon Behavior, Financial Performance and Economic Transformation——Data from Listed Companies in China

    The low-carbon development of enterprises is an important breakthrough in Chinese economic transformation and the optimization and upgrading of the industrial structure. Based on a sample of Chinese listed companies involved in the low-carbon industry from 2010 to 2018, this paper empirically analyzes the correlation between the low carbon behavior, economic transformation and financial performance of listed companies. The results show that a company’s carbon intensity and financial performance are negatively related, and this relation is more significant when the financial performance is measured using the ROA (return on asset) of listed companies. The level of economic transformation in places where enterprises are located can significantly strengthen the positive relationship between enterprise low-carbon behavior and financial performance, including in central and western areas, where positive relationships are strengthened, and areas with heavy polluting industries, where positive relationships are weakened. Therefore, it is necessary to strengthen carbon emission supervision for non-heavy polluting industries and enterprises in the central and western regions
