    INTRACELLULAR CENTRIFUGAL SEPARATION OF ORGANELLES IN PHYCOMYCES

    Live sporangiophores of Phycomyces blakesleeanus were centrifuged at 35,000 rpm. The cell contents sedimented into distinct layers, and each layer was studied with an electron microscope and with cytochemical methods. The following layers were found (their volumes and their densities are shown in Fig. 3): 1. polyphosphates; 2. polyphosphates and protein crystals; 3. glycogen; 4. yellow layer with ferritin; 5. ribosomes; 6. protein crystals; 7. mitochondria; 8. mitochondria and fibrils; 9. nuclei; 10. endoplasmic reticulum; 11. vesicles, membranes, and reticulum; 12. vacuole; 13. lipoproteins, membranes; 14. fat droplet. The densities of the various layers were determined by the injection of droplets of inert oils of known density into the sporangiosphores before centrifugation. Sedimented cell organelles could be isolated. Centrifuged nuclei of a lycopene-producing mutant were injected into the intact sporangiophore of an albino host where they induced color formation. The ensuing spores, when plated, gave a mixture of white and colored colonies. It was concluded that cell organelles, sedimented by centrifugation of living sporangiophores, remain alive and can be used for biochemical studies. Microspectrophotometric examination of the layers indicated the presence of cytochromes and flavines in the mitochondria and of cytochromes in the nuclei. No pigments corresponding to the action spectrum for the light growth response were found

    The writer discusses negative and positive reactions to nostalgia. He illustrates the creative view with the literary work and religious thought of J. C. Biernatzky (1795-1840), a German poet, writer and pastor of Polish descent. As a third-generation immigrant he still felt a deep nostalgia for Poland, the homeland of his grandfather. Regressive sequels, disturbances of higher cerebral activity and other abnormal behaviors are examined in relation to stress caused by immigration and the new language environment. The importance of the native language is stressed. In addition, the added pressure to assimilate imposed by monolingual (usually English-speaking) cultures is discussed

    Three-dimensional building modeling from lidar data based on the case of the town Domžale

    In the diploma thesis the procedure of creating a 3D model of buildings from lidar data is\ud treated. The goal is to present in a simple and efficient way the actual state of buildings on a\ud field and check the usability of automatic vectorisation of buildings from lidar data with\ud program package TerraSolid. Entire procedure, from data acquisition to final product is\ud presented. In theoretical part the basics of airborne laser scanning (ALS) and processing of\ud data, acquired with ALS technology, are described. Practical part was made on the case of the\ud town Domţale. Within this part the measuring equipment, used for realization of air mission,\ud recording of data, data processing and use of the program package TerraSolid for the 3D\ud building modeling are presented. We found out that automatic building modeling, based on\ud lidar data, with TerraSolid is quite simple, but for defining the accurate shape of the roofs, a\ud lot of manual work was still needed. 3D models are mostly used for comparison with realestate\ud records, in the spatial planning, town presentation and tourism
