    Influence of Ketotifen, Cromolyn Sodium, and Compound 48/80 on the survival rates after intestinal ischemia reperfusion injury in rats

    <p>Abstract</p> <p>Background</p> <p>Mast cells were associated with intestinal ischemia-reperfusion injury, the study was to observe the influence of Ketotifen, Cromolyn Sdium(CS), and Compound 48/80(CP) on the survival rates on the third day after intestinal ischemia-reperfusion injury in rats.</p> <p>Methods</p> <p>120 healthy Sprague-Dawley rats were randomly divided into 5 groups, Sham-operated group (group S), model group (group M), group K, group C and group CP. Intestinal damage was triggered by clamping the superior mesenteric artery for 75 minutes, group K, C, and CP were treated with kotifen 1 mg·kg<sup>-1</sup>, CS 50 mg·kg<sup>-1</sup>, and CP 0.75 mg·kg<sup>-1 </sup>i.v. at 5 min before reperfusion and once daily for three days following reperfusion respectively. Survival rate in each group was recorded during the three days after reperfusion. All the surviving rats were killed for determining the concentration of serum glutamic-oxaloacetic transaminase(AST), glutamic pyruvic transaminase(ALT), the ratio of AST compare ALT(S/L), total protein(TP), albumin(ALB), globulin(GLB), the ratio of ALB compare GLB(A/G), phosphocreatine kinase(CK), lactate dehydrogenase(LDH), urea nitrogen(BUN) and creatinine(CRE) at the 3<sup>rd </sup>day after reperfusion. And ultrastructure of IMMC, Chiu's score, lung histology, IMMC counts, the levels of TNF-α, IL-1β, IL-6 and IL-10 of the small intestine were detected at the same time.</p> <p>Results</p> <p>Intestinal ischemia-reperfusion injury reduced the survival rate. The concentrations of TP, ALB and level of IL-10 in intestine in group M decreased significantly while the concentrations of S/L, LDH and the levels of IL-6 and TNF-α in intestine increased significantly compared with group S (<it>P </it>< 0.05). Treatment with Ketotifen and CS increased the survival rate compared with group M (<it>P </it>< 0.05), attenuated the down-regulation or up-regulation of the above index (<it>P </it>< 0.05). Treatment with CP decreased the survival rate on the 3<sup>rd </sup>day after reperfusion compared with group M(<it>P </it>< 0.05). Group K and C had better morphology in IMMC in the small intestine and in the lungs than in group M and CP, although the Chiu's score and IMMC counts remained the same in the five groups(<it>P </it>> 0.05).</p> <p>Conclusion</p> <p>Mast cell inhibition after ischemia prior to reperfusion and following reperfusion may decrease the multi-organ injury induced by intestine ischemia reperfusion, and increase the survival rates.</p

    Quantum-Dot Light-Emitting Diodes with Nitrogen-Doped Carbon Nanodot Hole Transport and Electronic Energy Transfer Layer

    Electroluminescence efficiency is crucial for the application of quantum-dot light-emitting diodes (QD-LEDs) in practical devices. We demonstrate that nitrogen-doped carbon nanodot (N-CD) interlayer improves electrical and luminescent properties of QD-LEDs. The N-CDs were prepared by solution-based bottom up synthesis and were inserted as a hole transport layer (HTL) between other multilayer HTL heterojunction and the red-QD layer. The QD-LEDs with N-CD interlayer represented superior electrical rectification and electroluminescent efficiency than those without the N-CD interlayer. The insertion of N-CD layer was found to provoke the Forster resonance energy transfer (FRET) from N-CD to QD layer, as confirmed by time-integrated and - resolved photoluminescence spectroscopy. Moreover, hole-only devices (HODs) with N-CD interlayer presented high hole transport capability, and ultraviolet photoelectron spectroscopy also revealed that the N-CD interlayer reduced the highest hole barrier height. Thus, more balanced carrier injection with sufficient hole carrier transport feasibly lead to the superior electrical and electroluminescent properties of the QD-LEDs with N-CD interlayer. We further studied effect of N-CD interlayer thickness on electrical and luminescent performances for high-brightness QD-LEDs. The ability of the N-CD interlayer to improve both the electrical and luminescent characteristics of the QD-LEDs would be readily exploited as an emerging photoactive material for high-efficiency optoelectronic devices.ope

    Relationships Linking Amplification Level to Gene Over-Expression in Gliomas

    Background: Gene amplification is thought to promote over-expression of genes favouring tumour development. Because amplified regions are usually megabase-long, amplification often concerns numerous syntenic or non-syntenic genes, among which only a subset is over-expressed. The rationale for these differences remains poorly understood. Methodology/Principal Finding: To address this question, we used quantitative RT-PCR to determine the expression level of a series of co-amplified genes in five xenografted and one fresh human gliomas. These gliomas were chosen because we have previously characterised in detail the genetic content of their amplicons. In all the cases, the amplified sequences lie on extra-chromosomal DNA molecules, as commonly observed in gliomas. We show here that genes transcribed in nonamplified gliomas are over-expressed when amplified, roughly in proportion to their copy number, while non-expressed genes remain inactive. When specific antibodies were available, we also compared protein expression in amplified and nonamplified tumours. We found that protein accumulation barely correlates with the level of mRNA expression in some of these tumours. Conclusions/Significance: Here we show that the tissue-specific pattern of gene expression is maintained upon amplification in gliomas. Our study relies on a single type of tumour and a limited number of cases. However, it strongly suggests that, even when amplified, genes that are normally silent in a given cell type play no role in tumour progression

    A master equation is constructed to treat the nucleon transfer process in heavy ion fusion reactions to form superheavy nucleus. The relative motion concerning the energy, the angular momentum and the fragment deformation relaxations is explicitly treated to couple with the diffusion process. The nucleon transition probabilities, which are derived microscopically, are thus time dependent. The calculated evaporation residue cross-sections for both cold and hot fusion are in good agreement with the known experimental data

    Questioning the effectiveness of vaping on smoking cessation success among young people: re-analysis of the 2019 Australian National Drug Strategy Household Survey

    Aims We critically reexplored a recent claim that smoking cessation success rates were higher among people who used e-cigarettes. In particular, we investigated the consistency of this effect among different age groups. In addition, we explored the relationships between vaping, smoking cessation and youth mental health. Methods Cross-sectional analyses of data from the 2013, 2016, and 2019 Australian National Drug Strategy Household Surveys were undertaken. A successful smoking cessation attempt was defined as having abstained from smoking for at least one month over the past year. The Kessler Psychological Distress Scale (K10) was used to measure psychological distress. Multivariable logistic regression analyses were conducted to examine interactions between age and use of e-cigarettes on smoking cessation and youth mental health, adjusting for demographic factors. Results The previous observation that a higher percentage of people quit smoking using e-cigarettes (than without e-cigarettes) was only observed in adults, but not among youth aged 14-25. Moreover, among those that quit smoking using e-cigarettes, a large majority were currently vaping. Youths had the highest rates of mental ill-health and the highest rates of ever-vaping. Discussion &amp; Conclusions Vaping rates are rising rapidly, especially among youths. Given the physical health effects of vaping and its association with higher mental health, we propose a precautionary approach when assessing the complex relationships between vaping and smoking cessation. E-cigarettes as an aid for smoking cessation should only be used when medically prescribed, under close supervision, and only when all other safer smoking cessation aids have been exhausted

    Neutron and proton diffusion in fusion reactions for the synthesis of superheavy nuclei

    The restriction of the one dimensional (1D) master equation (ME) with the mass number of the projectile-like fragment as a variable is studied, and a two-dimensional (2D) master equation with the neutron and proton numbers as independent variables is set up, and solved numerically. Our study showed that the 2D ME can describe the fusion process well in all projectile-target combinations. Therefore the possible channels to synthesize super-heavy nuclei can be studied correctly in wider possibilities. The available condition for employing 1D ME is pointed out
