    Jamming Transition of Point-to-Point Traffic Through Cooperative Mechanisms

    We study the jamming transition of two-dimensional point-to-point traffic through cooperative mechanisms using computer simulation. We propose two decentralized cooperative mechanisms which are incorporated into the point-to-point traffic models: stepping aside (CM-SA) and choosing alternative routes (CM-CAR). Incorporating CM-SA is to prevent a type of ping-pong jumps from happening when two objects standing face-to-face want to move in opposite directions. Incorporating CM-CAR is to handle the conflict when more than one object competes for the same point in parallel update. We investigate and compare four models mainly from fundamental diagrams, jam patterns and the distribution of cooperation probability. It is found that although it decreases the average velocity a little, the CM-SA increases the critical density and the average flow. Despite increasing the average velocity, the CM-CAR decreases the average flow by creating substantially vacant areas inside jam clusters. We investigate the jam patterns of four models carefully and explain this result qualitatively.     Multicell Edge Coverage Enhancement Using Mobile UAV-Relay

    Unmanned aerial vehicle (UAV)-assisted communication is a promising technology in future wireless communication networks. UAVs can not only help offload data traffic from ground base stations (GBSs) but also improve the Quality of Service (QoS) of cell-edge users (CEUs). In this article, we consider the enhancement of cell-edge communications through a mobile relay, i.e., UAV, in multicell networks. During each transmission period, GBSs first send data to the UAV, and then the UAV forwards its received data to CEUs according to a certain association strategy. In order to maximize the sum rate of all CEUs, we jointly optimize the UAV mobility management, including trajectory, velocity, and acceleration, and association strategy of CEUs to the UAV, subject to minimum rate requirements of CEUs, mobility constraints of the UAV, and causal buffer constraints in practice. To address the mixed-integer nonconvex problem, we transform it into two convex subproblems by applying tight bounds and relaxations. An iterative algorithm is proposed to solve the two subproblems in an alternating manner. Numerical results show that the proposed algorithm achieves higher rates of CEUs as compared with the existing benchmark schemes

    Natural transformation of Thermotoga sp. strain RQ7

    Background Thermotoga species are organisms of enormous interest from a biotechnological as well as evolutionary point of view. Genetic modifications of Thermotoga spp. are often desired in order to fully release their multifarious potentials. Effective transformation of recombinant DNA into these bacteria constitutes a critical step of such efforts. This study aims to establish natural competency in Thermotoga spp. and to provide a convenient method to transform these organisms. Results Foreign DNA was found to be relatively stable in the supernatant of a Thermotoga culture for up to 6 hours. Adding donor DNA to T. sp. strain RQ7 at its early exponential growth phase (OD600 0.18 ~ 0.20) resulted in direct acquisition of the DNA by the cells. Both T. neapolitana chromosomal DNA and Thermotoga-E. coli shuttle vectors effectively transformed T. sp. strain RQ7, rendering the cells resistance to kanamycin. The kan gene carried by the shuttle vector pDH10 was detected by PCR from the plasmid extract of the transformants, and the amplicons were verified by restriction digestions. A procedure for natural transformation of Thermotoga spp. was established and optimized. With the optimized method, T. sp. strain RQ7 sustained a transformation frequency in the order of 10−7 with both genomic and plasmid DNA
