    OCT for non-destructive examination of the internal biological structures of mosquito specimen

    The Study of mosquitoes and their behavioral analysis are of crucial importance to control the alarmingly increasing mosquito-borne diseases. Conventional imaging techniques use either dissection, exogenous contrast agents. Non-destructive imaging techniques, like x-ray and microcomputed tomography uses ionizing radiations. Hence, a non-destructive and real-time imaging technique which can obtain high resolution images to study the anatomical features of mosquito specimen can greatly aid researchers for mosquito studies. In this study, the three-dimensional imaging capabilities of optical coherence tomography (OCT) for structural analysis of Anopheles sinensis mosquitoes has been demonstrated. The anatomical features of An. sinensis head, thorax, and abdomen regions along with internal morphological structures like foregut, midgut, and hindgut were studied using OCT imaging. Two-dimensional (2D) and three-dimensional (3D) OCT images along with histology images were helpful for the anatomical analysis of the mosquito specimens. From the concurred results and by exhibiting this as an initial study, the applicability of OCT in future entomological researches related to mosquitoes and changes in its anatomical structure is demonstrated

    The First Case of X-linked Alpha-thalassemia/Mental Retardation (ATR-X) Syndrome in Korea

    Mutation of the ATRX gene leads to X-linked alpha-thalassemia/mental retardation (ATR-X) syndrome and several other X-linked mental retardation syndromes. We report the first case of ATR-X syndrome documented here in Korea. A 32-month-old boy came in with irritability and fever. He showed dysmorphic features, mental retardation and epilepsy, so ATR-X syndrome was considered. Hemoglobin H inclusions in red blood cells supported the diagnosis and genetic studies confirmed it. Mutation analysis for our patient showed a point mutation of thymine to cytosine on the 9th exon in the ATRX gene, indicating that Trp(C), the 220th amino acid, was replaced by Ser(R). Furthermore, we investigated the same mutation in family members, and his mother and two sisters were found to be carriers

    Protistan community structure and the influence of a branch of Kuroshio in the northeastern East China Sea during the late spring

    The northeastern East China Sea is an ecologically important marine ecosystem influenced by warm water derived from the Kuroshio Current. However, relatively little is known about the spatial variation of protist communities and their regulating factors from this region’s ecosystem during the spring season. Here, we investigated protistan community structures using a complementary approach combining 18S rRNA gene amplicon sequencing and light microscopy cell counts at nine stations from the northeastern East China Sea to the west of Jeju Island. The vertical profiles of physicochemical properties revealed that the Jeju Warm Current water mass, flowing from the southeast towards the northwest, created a thermohaline front dividing the region in two. These two regions had similar planktonic biomass, but the protistan communities differed significantly: dinoflagellates accounted for higher proportions of the protistan communities in the warm and saline waters, particularly at stations E35, E44, and E45, while the relative abundances of diatoms and picochlorophytes were higher in the low-density water of the western stations (E32 and E42). Furthermore, higher species richness and Shannon Diversity Index values in the warm and saline waters suggests that the Jeju Warm Current, a branch of the Kuroshio, increases protistan taxonomic diversity in the northeastern East China Sea during the late spring. Seed populations of harmful algal bloom-causing species were discovered in the warm and saline water originating from the Kuroshio, which is particularly important as it indicates that these waters could introduce harmful species that may spread to the Yellow Sea and Korea Strait. Taken together, the study suggests that potential changes to the current systems in the region could dramatically alter the structure of its protistan community

    Genomic profile analysis of diffuse-type gastric cancers

    Background: Stomach cancer is the third deadliest among all cancers worldwide. Although incidence of the intestinal-type gastric cancer has decreased, the incidence of diffuse-type is still increasing and its progression is notoriously aggressive. There is insufficient information on genome variations of diffuse-type gastric cancer because its cells are usually mixed with normal cells, and this low cellularity has made it difficult to analyze the genome. Results: We analyze whole genomes and corresponding exomes of diffuse-type gastric cancer, using matched tumor and normal samples from 14 diffuse-type and five intestinal-type gastric cancer patients. Somatic variations found in the diffuse-type gastric cancer are compared to those of the intestinal-type and to previously reported variants. We determine the average exonic somatic mutation rate of the two types. We find associated candidate driver genes, and identify seven novel somatic mutations in CDH1, which is a well-known gastric cancer-associated gene. Three-dimensional structure analysis of the mutated E-cadherin protein suggests that these new somatic mutations could cause significant functional perturbations of critical calcium-binding sites in the EC1-2 junction. Chromosomal instability analysis shows that the MDM2 gene is amplified. After thorough structural analysis, a novel fusion gene TSC2-RNF216 is identified, which may simultaneously disrupt tumor-suppressive pathways and activate tumorigenesis. Conclusions: We report the genomic profile of diffuse-type gastric cancers including new somatic variations, a novel fusion gene, and amplification and deletion of certain chromosomal regions that contain oncogenes and tumor suppressors.open121

    <p>Abstract</p> <p>Background</p> <p>Second-generation drug-eluting stents (DES) have raised the bar of clinical performance. These stents are mostly made from cobalt chromium alloy. A newer generation DES has been developed from platinum chromium alloy, but clinical data regarding the efficacy and safety of the platinum chromium-based everolimus-eluting stent (PtCr-EES) is limited, with no comparison data against the cobalt chromium-based zotarolimus-eluting stent (CoCr-ZES). In addition, an antiplatelet regimen is an integral component of medical therapy after percutaneous coronary intervention (PCI). A 1-week duration of doubling the dose of clopidogrel (double-dose antiplatelet therapy (DDAT)) was shown to improve outcome at 1 month compared with conventional dose in acute coronary syndrome (ACS) patients undergoing PCI. However in Asia, including Korea, the addition of cilostazol (triplet antiplatelet therapy (TAT)) is used more commonly than doubling the dose of clopidogrel in high-risk patients.</p> <p>Methods</p> <p>In the 'Harmonizing Optimal Strategy for Treatment of coronary artery stenosis - sAfety & effectiveneSS of drug-elUting stents & antiplatelet REgimen' (HOST-ASSURE) trial, approximately 3,750 patients are being prospectively and randomly assigned in a 2 × 2 factorial design according to the type of stent (PtCr-EES vs CoCr-ZES) and antiplatelet regimen (TAT vs DDAT). The first primary endpoint is target lesion failure at 1 year for the stent comparison, and the second primary endpoint is net clinical outcome at 1 month for comparison of antiplatelet therapy regimen.</p> <p>Discussion</p> <p>The HOST-ASSURE trial is the largest study yet performed to directly compare the efficacy and safety of the PtCr-EES versus CoCr-ZES in an 'all-comers' population. In addition, this study will also compare the clinical outcome of TAT versus DDAT for 1-month post PCI.</p> <p>Trial registration</p> <p>ClincalTrials.gov number <a href="http://www.clinicaltrials.gov/ct2/show/NCT01267734">NCT01267734</a>.</p
