    Acute Toxicity Test of Unripe Papaya (<I>Carica papaya</I> L.) Aqueous Extract (UPAE) on The Blood Urea and Creatinine Concentration

    Unripe papaya aqueous extract (UPAE) widely used as lactation stimulator, antidiabetes, antibacterial, and anti-inflammatory. The utilization of papaya is not known for its safety yet, so it is necessary to research its toxicity. The purpose of this study was to investigate the acute toxicity of UPAE on renal function through measurement of blood urea and creatinine levels. This study was conducted in July 2017 in Laboratory of Medical Biology, Faculty of Medicine, Universitas Islam Bandung. This study used pure in vivo experimental design on 11 Swiss Webster mice using the dose of acute toxicity determination based on new recommended methods of 0; 50; 200; 400; 800; 1,000; 1,500; 2,000; 3,000; 4,000; and 5,000 mg/kgBW. After 24 hours, 1 mL blood drawn through the tail examined for blood urea and creatinine levels. The measurement of urea content using kinetic method point and creatinine level using modified Jaffe method. Provision of UPAE at doses of 0, 50, 200, 400, 800, and 1,000 mg/kgBW resulted on blood urea equal to 39, 35, 48, 49, 48, and 32 mg/dL respectively. Blood urea level 23, 22, 28, 34, and 35 mg/dL was obtained at 1,500 UPAE doses; 2,000; 3,000; 4,000; and 5,000 mg/kgBW dosages respectively. After 24 hours of UPAE administration, the creatinine level in various doses using new recommended method of (0–5,000 mg/kgBW) were 0.75, 0.54, 0.53, 0.50, 0.60, 0.54, 0.52, 0.55, 0.42, 0.51, and 0.40 mg/dL. In conclusion, UPAE do not cause acute toxicity on renal function through measurement of blood urea and creatinine levels.   TOKSISITAS AKUT EKSTRAK AIR BUAH PEPAYA (CARICA PAPAYA L.) TERHADAP KADAR UREUM DAN KREATININ DARAH Ekstrak air buah pepaya muda (EABPM) digunakan secara empiris sebagai laktagogum, antidiabetes, antibakteri, dan antiinflamasi. Tingkat keamanannya belum banyak diketahui sehingga perlu dilakukan penelitian uji toksisitas akut. Tujuan penelitian ini adalah mengetahui toksisitas akut EABPM terhadap fungsi ginjal melalui pengukuran kadar ureum dan kreatinin plasma. Penelitian ini dilaksanakan pada bulan Juli 2018 di Laboratorium Biologi Medis, Fakultas Kedokteran, Universitas Islam Bandung. Penelitian ini menggunakan desain eksperimental murni in vivo terhadap 11 ekor mencit betina galur Swiss Webster dengan penentuan dosis sesuai dengan new recommended method: 0, 50, 200, 400, 800, 1.000, 1.500, 2.000, 3.000, 4.000, dan 5.000 mg/kgBB. Setelah 24 jam, diambil darah melalui ekor mencit sebanyak 1 mL untuk diperiksa kadar ureum dan kreatinin plasma. Pengukuran kadar ureum menggunakan point kinetic method dan kadar kreatinin menggunakan metode Jaffe yang dimodifikasi. Pemberian EABPM pada dosis 0, 50, 200, 400, 800, dan 1.000 mg/kgBB didapatkan kadar ureum plasma 39, 35, 48, 49, 48, dan 32 mg/dL secara berurutan. Kadar ureum plasma 23, 22, 28, 34, dan 35 mg/dL didapatkan pada pemberian dosis EABPM sebanyak 1.500, 2.000, 3.000, 4.000, dan 5.000 mg/kgBB. Kadar kreatinin plasma dalam berbagai dosis (0–5.000 mg/kgBB) adalah 0,75; 0,54; 0,53; 0,50; 0,60; 0,54; 0,52; 0,55; 0,42; 0,51; dan 0,40 mg/dL. Simpulan, EABPM tidak menimbulkan tanda toksisitas akut pada fungsi ginjal melalui pengukuran kadar ureum dan kreatinin plasma

    Comparison Between Different Herbal Formulations of Roses and Their Therapeutic Potential

    Roses are one of the most popular flowers in the world, not only because of their beauty but also because they have medicinal properties. Drugs developed from roses must be support from various parties. Roses have active components that can be used to improve public health, such as anthocyanins, tannins and other active compounds. Making the right rose water herbal mixtures will help filter out the active substances according to the expected therapeutic effect. The manufacture of herbal mixtures of roses can vary, namely, infusions, extracts and rose oil. These three variations require different procedures. The three herbal formulations can provide an initial condition of the content of the active substances in the roses. The results of the rose infusion gave a red color in a short time, while the maceration of the extract with ethyl alcohol, methanol and hexsan gave a varied color appearance, while the identification of rose oil using water distillation did not show any filtered oil. The comparison of the three preparations shows that the herbal formulations can be done in a simple way and the formulation that gives the best results is rose water infusions that produces rose water.     Histopathology of Nephrotoxicity Associated with Administered Water Extract Purple Sweet Potato (<i>Ipomoea batatas</i>) in Mice (<i>Mus musculus</i>) in Stratified Phases of Dose

    The main aim of the registered purple sweet potato (Ipomoea batatas) is to provide minimize the adverse chemical drugs, in addition to its anti-inflammatory, antioxidant, and antimicrobial effects. Potentially adverse effects may be observed in laboratory animals in particular, the extent to which this administration can cause toxicity. This study aimed to examine the histopathology of nephrotoxicity associated with administered water extracts of purple sweet potato in mice with stratified doses. The study was conducted at the Biomedical Laboratory, Faculty of Medicine, Universitas Islam Bandung on September 2019. Female mice (Mus musculus) strain Swiss Webster, aged between 6–8 weeks weighing 25 to 30 g, were obtained from Biopharma Laboratory, Bandung. The animal was acclimatized for seven days before being administered water extract purple sweet potato: eleven mice, one control group, and ten treatment groups underwent toxicity doses of purple sweet potato water extract administration. Purple sweet potato variant of Ayamurasaki prepared in various oral doses. The results show in the control group there were no histopathological changes, but in the group administered water extract purple sweet potato from the first phase seems in a mild grade of macrophage accumulation, mild vacuolization of tubular epithelial cells, mild vascular dilatation, and mild hydrophilic degeneration. In the second phase, macrophage accumulation was visible in moderate grades. The LD50 of purple sweet potato extract is greater than 5,000 mg/kgBW. The findings of this study indicate that registration of purple sweet potato extract in confirmatory doses is safe to administer and did not exhibit any mortality.     Feature of the Knowledge of the Posyandu Cadre School Participants Based on the Pattern of the Material Provided

    National health development focuses on preventive and promotive approaches, as well as empowering families and communities through Posyandu (Integrated Health Service Post). Posyandu cadres play a crucial role in improving the health status of the community. It is essential to continuously foster and enhance the capacity of Posyandu cadres, particularly in addressing health issues related to infants and pregnant women, with a holistic approach that empowers the surrounding community. In collaboration with Parongpong District, the Faculty of Medicine at Unisba has developed a program called the Posyandu Cadre School to strengthen the skills and knowledge of Posyandu cadres. This program aims to improve their understanding of their roles at Posyandu, health topics, communication skills, and the role of women in Islam. Through the Posyandu Cadre School, cadres receive training on various topics, with a total of 17 topics covered. The post-test evaluation of the program reveals that cadres have achieved the highest mastery in topics such as the role of women in Islam, nutrition, stunting, youth health, coping with stress management, and skin health (score &gt; 75%). However, there are certain topics where understanding remains relatively low, such as English proficiency and prevention and control of diarrhea (score &lt; 50%). Nevertheless, overall, the participants have demonstrated a good understanding of the basic concepts of Posyandu. These findings highlight the importance of continuous debriefing and competence-building for Posyandu cadres to ensure their readiness and competency.     Cogongrass (<i>Imperata cylindrica</i> L.) Ethanol Extract on Sepsis Mice Model Body Weight and Sepsis Score

    Sepsis causes damage for cells, behavioral phenotype regression, and will end in most patients' death. The ethanol extract of cogongrass (Imperata cylindrica L.)  acts as an antioxidant. This study aimed to observe the effect of giving ECGR to body weight (BW) and the sepsis score of the sepsis mice model by lipopolysaccharide (LPS) induction. This study was an in vivo study with a randomized post-test controlled group design at the Animal Laboratory of Universitas Padjadjaran, 2018. We used 4 (four) groups of male mice (Mus musculus) DDY strains. Group 1 as a control, group 2: LPS 10 μL/kgBW, group 3, and 4: LPS+ECGR (90 mg/kgBW, and a dose of 115 mg/kgBW, respectively). This treatment was performed for two weeks. Every three days, we measured their body weight. After two weeks, group 2, group 3, and 4 were injected with LPS for 8 hours to induce sepsis. Next, we measured body weight and sepsis score using murine sepsis score (MSS). Then statistical analysis was performed using ANOVA and the Kruskal-Wallis test. The results showed no differences in body weight were found in the treatment groups (3 and 4) compared with control, suggesting no effect of ECGR in decreasing mice body weight. The sepsis score was more than 21 in groups treated with LPS (2, 3, and 4), suggesting LPS can induce sepsis. There was a slight decrease in scores in-group 3 and 4 compared with group 2. This study concludes that the treatment of ECGR caused no harm to body weight and slightly decreased sepsis score in the sepsis mice model.   EKSTRAK ETANOL ALANG-ALANG (IMPERATA CYLINDRICA L.) TERHADAP BOBOT BADAN DAN SKOR SEPSIS MENCIT MODEL SEPSIS Sepsis menyebabkan kerusakan sel, regresi fenotipe perilaku, dan akan berakhir kematian pada sebagian besar pasien. Ekstrak etanol akar alang-alang (Imperata cylindrica L.) (ECGR) berperan sebagai antioksidan. Penelitian ini bertujuan mengetahui pengaruh pemberian ECGR terhadap bobot badan (BB) dan skor sepsis pada mencit model sepsis yang diinduksi lipopolisakarida (LPS). Penelitian ini adalah penelitian in vivo dengan desain randomized post-test controlled group di Laboratoium Hewan Universitas Padjadjaran tahun 2018. Kami menggunakan 4 (empat) kelompok mencit jantan (Mus musculus) strain DDY. Kelompok 1 sebagai kontrol, kelompok 2 diinduksi LPS 10 μL/kgBB, kelompok 3 dan 4 diinduksi LPS+ECGR (dosis 90 mg/kgBB dan 115 mg/kgBB masing-masing). Perlakuan ini dilakukan selama 2 minggu. Setiap tiga hari dilakukan pengukuran bobot badan mencit. Setelah dua minggu, kelompok 2, kelompok 3, dan kelompok 4 diinjeksi LPS selama 8 jam untuk menginduksi sepsis. Selanjutnya, diukur bobot badan dan skor sepsis menggunakan murine sepsis score (MSS). Analisis statistik menggunakan ANOVA dan Uji Kruskal-Wallis. Hasil penelitian menunjukkan tidak terdapat perbedaan bobot badan pada kelompok perlakuan (3 dan 4) dibanding dengan kelompok kontrol yang menunjukkan ECGR tidak berpengaruh dalam menurunkan bobot badan mencit. Skor sepsis lebih dari 21 pada kelompok yang diinduksi LPS (2, 3, dan 4) menunjukkan LPS dapat menyebabkan sepsis. Terdapat sedikit penurunan skor pada kelompok 3 dan 4 dibanding dengan kelompok 2. Simpulan penelitian ini, pengobatan ECGR tidak membahayakan bobot badan dan mengakibatkan sedikit penurunan skor sepsis pada mencit model sepsis
