Gene Transfer of Calcitonin Gene-Related Peptide Inhibits Macrophages and Inflammatory Mediators in Vein Graft Disease
Vein graft disease is a chronic inflammatory disease and limits the late results of coronary revascularization. Calcitonin gene-related peptide (CGRP) inhibits macrophages infiltrated and inflammatory mediators, we hypothesized that transfected CGRP gene inhibits macrophages infiltrated and inflammatory mediators in vein graft disease. Autologous rabbit jugular vein grafts were incubated ex vivo in a solution of mosaic adeno-associated virus vectors containing CGRP gene (AAV2/1.CGRP) 、escherichia coli lac Z gene (AAV2/1.LacZ) or saline and then interposed in the carotid artery. Intima/media ratio were evaluated at postoperative 4 weeks, Macrophages were marked with CD68 antibody by immunocytochemistry. Inflammatory mediators were mensurated with real-time PCR. Neointimal thickening was significantly suppressed in AAV2/1.CGRP group. Macrophages infiltrated and inflammatory mediators monocyte chemoattractant protein-1 (MCP-1)、tumor necrosis factorα(TNF-α)、inducible nitricoxide synthase (iNOS)、matrix metalloproteinase-9 (MMP-9) was significantly suppressed in AAV2/1.CGRP group.Gene transfected AAV2/1.CGRP suppressed neointimal hyperplasia in vein graft disease by suppressed macrophages infiltrated and inflammatory mediators
A Network Celebrity Identification and Evaluation Model Based on Hybrid Trust Relation
Trust-based celebrity user identification is the key to the industry\u27s reputation for electronic word of mouth. However, trust and mistrust are independent and coexistent concepts. In this context, we need to consider the existence of the two kinds of user relations brought about by the impact. This paper analyzes the characteristics of trust and distrust in social networks, and gives formal descriptions of trust networks, untrusted networks, and mixed trust networks. Based on the indicators such as degree distribution, correlation coefficient, and matching coefficient, the structural properties of mixed trust networks are studied. Based on the PageRank algorithm, the HTMM metrics affecting users under the mixed trust network environment are proposed. Finally, the validity of HTMM is verified through a real data set containing trust and distrust. Experimental results show that the results of HTMM\u27s celebrity user identification method still have a low level of trust
Boosting Continuous Control with Consistency Policy
Due to its training stability and strong expression, the diffusion model has
attracted considerable attention in offline reinforcement learning. However,
several challenges have also come with it: 1) The demand for a large number of
diffusion steps makes the diffusion-model-based methods time inefficient and
limits their applications in real-time control; 2) How to achieve policy
improvement with accurate guidance for diffusion model-based policy is still an
open problem. Inspired by the consistency model, we propose a novel
time-efficiency method named Consistency Policy with Q-Learning (CPQL), which
derives action from noise by a single step. By establishing a mapping from the
reverse diffusion trajectories to the desired policy, we simultaneously address
the issues of time efficiency and inaccurate guidance when updating diffusion
model-based policy with the learned Q-function. We demonstrate that CPQL can
achieve policy improvement with accurate guidance for offline reinforcement
learning, and can be seamlessly extended for online RL tasks. Experimental
results indicate that CPQL achieves new state-of-the-art performance on 11
offline and 21 online tasks, significantly improving inference speed by nearly
45 times compared to Diffusion-QL. We will release our code later.Comment: 18 pages, 9 page
WS-mediator for improving dependability of service composition
Web Services and service-oriented architectures (SOAs) represent a new paradigm for building distributed computing applications. In recent years, they have started to play a critical role in numerous e-Science and e-Commerce applications. The advantages of Web Services, such as their loosely coupled architecture and standardized interoperability, make them a desirable platform, especially for developing large-scale applications such as those based on cross-organizational service composition. However, the Web Service technology is now facing many serious issues that need to be addressed, one of the most important ones being the dependability of their composition. Web Service composition relies on individual component services and computer networks, particularly the Internet. As the component services are autonomous, prior to use their dependability is unknown. In addition to that, computer networks are inherently unreliable media: from the user's perspective, network failures may undermine the dependability of Web Services. Consequently, failures of individual component services and of the network can undermine the dependability of the entire application relying on service composition. Our research is intended to contribute to achieving higher dependability of Web Service composition. We have developed a novel solution, called WS-Mediator system, implementing resilience-explicit computing and fault tolerance mechanisms to improve the dependability of Web Service composition. It consists of a number of subsystems, called Sub-Mediators, which are deployed at various geographical locations across the Internet to monitor Web Services and dynamically generate Web Service dependability metadata in order to make resilience-explicit decisions. In addition to applying the fault tolerance mechanisms that deal with various kinds of faults during the service composition, the resilience-explicit reconfiguration mechanism dynamically selects the most dependable Web Services to achieve higher service composition dependability fault tolerance. A specific instance of the WS-Mediator architecture has been developed in the Java Web Service technology. A series of experiments with real-world Web Services, in particular in the bioinformatics domain, have been carried out using the Java WS- Mediator.
