We present the functional and structural characterization of the first archaeal thermostable NADP-dependent aldehyde dehydrogenase AlDHPyr1147. In vitro, AlDHPyr1147 catalyzes the irreversible oxidation of short aliphatic aldehydes at 60–85°С, and the affinity of AlDHPyr1147 to the NADP+ at 60°С is comparable to that for mesophilic analogues at 25°С. We determined the structures of the apo form of AlDHPyr1147 (3.04 Å resolution), three binary complexes with the coenzyme (1.90, 2.06, and 2.19 Å), and the ternary complex with the coenzyme and isobutyraldehyde as a substrate (2.66 Å). The nicotinamide moiety of the coenzyme is disordered in two binary complexes, while it is ordered in the ternary complex, as well as in the binary complex obtained after additional soaking with the substrate. AlDHPyr1147 structures demonstrate the strengthening of the dimeric contact (as compared with the analogues) and the concerted conformational flexibility of catalytic Cys287 and Glu253, as well as Leu254 and the nicotinamide moiety of the coenzyme. A comparison of the active sites of AlDHPyr1147 and dehydrogenases characterized earlier suggests that proton relay systems, which were previously proposed for dehydrogenases of this family, are blocked in AlDHPyr1147, and the proton release in the latter can occur through the substrate channel

The objective was to conduct a comparative analysis of the dynamics of biomarkers of apoptosis and autophagy in the eripheral blood of patients in the acute period of ischemic stroke (IS) and compare it with the dynamics of the severity of neurological deficit according to the NIHSS (National Institutes of Health Stroke Scale) and the volume of the brain lesion according to the results of magnetic resonance imaging (MRI). To assess the role of the studied parameters in the prognosis of the outcome of the acute period of the disease. Methods and material. We examined 56 patients in the acute period of newly developed atherothrombotic IS. The control group consisted of 29 healthy donors. Patients underwent a dynamic clinical and neurological examination on the 1st, 7th and 14th days from the onset of the disease. At the same time intervals, the dynamics of the expression of biomarkers of apoptosis and autophagy in peripheral blood was evaluated by flow cytometry and compared with neurological status indicators on the 1st, 7th, and 14th  days, respectively, using correlation analysis.Results. A statistically significant increase in the expression of annexin V and caspase-3 compared with the control group was observed throughout the study in all populations of leukocytes with a maximum increase in the first 24 hours. Increased expression levels of annexin V and caspase-3 positively correlated with the severity of neurological deficit and the amount of brain damage on the 1st and 7th days. A direct correlation was found between increased values of autophagy biomarkers LC3, Cyto-ID, the amount of brain damage, and the severity of neurological deficit on the 7th day.Conclusion. A statistically significant increase in biomarkers of apoptosis and autophagy in the peripheral blood in the acute period of IS correlates with the severity of clinical and neurological parameters. The role of a significant increase in caspase-3 as a predictor of adverse disease outcome requires further study.

Dear Colleagues! We are glad to present the 10th Edition (revised) of the Standards of Specialized Diabetes Care. These evidence-based guidelines were designed to standardize and facilitate diabetes care in all regions of the Russian Federation. The Standards are updated on the regular basis to incorporate new data and relevant recommendations from national and international clinical societies, including World Health Organization Guidelines (WHO, 2011, 2013), International Diabetes Federation (IDF, 2011, 2012, 2013), European Association for the Study of Diabetes (EASD 2018, 2019), American Diabetes Association (ADA, 2018, 2019, 2021), American Association of Clinical Endocrinologists (AACE, 2020, 2021), International Society for Pediatric and Adolescent Diabetes (ISPAD, 2018) and Russian Association of Endocrinologists (RAE, 2019). Current edition of the “Standards” also integrates results of completed randomized clinical trials (ADVANCE, ACCORD, VADT, UKPDS, SAVOR, TECOS, LEADER, EXAMINE, ELIXA, SUSTAIN, DEVOTE, EMPA-REG OUTCOME, CANVAS, DECLARE, CARMELINA, REWIND, CREDENCE, CAROLINA, DAPA-CKD, DAPA-HF, EMPEROR-Reduced trial, VERIFY, VERTIS CV, PIONEER, etc.), as well as findings from the national studies of diabetes mellitus (DM), conducted in close partnership with a number of Russian hospitals.Latest data indicates that prevalence of DM in the world increased during the last decade more than two-fold, reaching some 537 million patients by the end of 2021. According to the current estimation by the International Diabetes Federation, 643 million patients will be suffering from DM by 2030 and 784 million by 2045.Like many other countries, Russian Federation experiences a sharp rise in the prevalence of DM. According to Russian Federal Diabetes Register, there are at least 4 871 863 patients with DM in this country on 01.01.2021 (3,34% of population) with 92,3% (4 498 826)–Type 2 DM, 5,6% (271 468)–Type 1 DM and 2,1% (101 569)–other types of DM, including 9 729 women with gestational DM. However, these results underestimates real quantity of patients, because they consider only registered cases. Results of Russian epidemiological study (NATION) confirmed that only 54% of Type 2 DM are diagnosed. So real number of patients with DM in Russia is 10 million patients (about 7% of population). This is a great long-term problem, because a lot of patients are not diagnosed, so they don’t receive any treatment and have high risk of vascular complications.Severe consequences of the global pandemic of DM include its vascular complications: nephropathy, retinopathy, coronary, cerebral and peripheral vascular disease. These conditions are responsible for the majority of cases of diabetes-related disability and death. In сurrent edition of the “Standards”:New goals of glycemic control for continuous glucose monitoring (time in range, below range and above range, glucose variability) are given. It also features updated guidelines on stratification of treatment in newly diagnosed Type 2 diabetes.In the recommendations for the personalization of the choice of antidiabetic agents, it is taken into account that in certain clinical situations (the presence of atherosclerotic cardiovascular diseases and their risk factors, chronic heart failure, chronic kidney disease, obesity, the risk of hypoglycemia) certain classes of hypoglycemic agents (or individual drugs) have proven advantages.Indications for the use of antidiabetic agents in chronic kidney disease are expanded.Information about insulin pump therapy is added.Recommendations on vaccination are added. An algorithm for replacing some insulin preparations with others is given. This text represents a consensus by the absolute majority of national experts, achieved through a number of fruitful discussions held at national meetings and forums. These guidelines are intended for endocrinologists, primary care physicians, pediatricians and other medical professionals involved in the treatment of DM.Compared with previous edition of the Standards of Specialized Diabetes Care edited by Dedov I.I., Shestakova M.V., Mayorov A.Yu., 10th edition, Moscow, 2021 (signed for printing on 10.09.2021) a number of changes have been made.On behalf of the Working Grou

Accretion disks with optical depth transition and advection

Abstract. We consider the effects of advection and radial gradients of pressure and radial drift velocity on the structure of accretion disks around black holes. We concentrated our efforts on highly viscous disk with large accretion rate. Contrary to disk models neglecting advection, we find that continuous solutions extending from the outer disk regions to the inner edge exist for all accretion rates we have considered. We show that the sonic point moves outward with increasing accretion rate. Despite the importance of advection on it's structure, the disk remains geometrically thin. Global solutions of advective accretion disks, which describes continuously the transition between optically thick and optically thin disk regions are constructed and analyzed. Key words. Accretion, accretion disks -black hole physics The Model and the Method of Solution In this paper we will consider the full set of solutions to the disk structure equations with advection including the optically thick, optically thin and the intermediate zones in accretion disks. We use from now on geometric units with G = 1, c = 1, use r as the radial coordinate scaled to r g = M, and scale all velocities to c. We work with the pseudo-Newtonian potential proposed by We use the same set of equations, ingredients and boundary conditions in our models as in The following equations are therefore modified

Paleoenvironment changes in the NW Okhotsk Sea for the last 18 kyr determined with micropaleontological, geochemical, and lithological data

Lithological, geochemical, and micropaleontological data indicate that the Late Glacial of the northwestern Okhotsk Sea (OS) is characterised by severe climatic and environmental conditions with mainly perennial sea ice coverage and low productivity accompanied by weak deep-water ventilation and a temperate formation of the upper Sea of Okhotsk Intermediate Water (SOIW). The age model of the studied core sediments was constructed by AMS 14C dating. The most severe environmental conditions occurred during the period 15.8–14.8 kyr, synchronous with cold Heinrich event 1. Insignificant regional environmental amelioration accompanied by an increase of productivity and ice weakening during summer occurred almost simultaneously with the Bølling–Allerøed (BA) warming. The obtained results distinguished both the Bølling and Allerøed warmings as having different environmental conditions. Oxygen content in the surface sediment was low, as seen from the production of the benthic foraminifera (BF) species. During 12.6–11.1 kyr, synchronous with the Younger Dryas (YD) cold event, the regional environment conditions were cold, but not as severe as the glacial ones. Some climatic warming since the Preboreal has stimulated sea ice melting and surface amelioration during the summer season, which in turn led to a productivity rise and changes in the water column and bottom environment. Some increase in the surface water stratification and the intensified oceanic diatom and surface radiolarian production is parallel with the development of a mesopelagic regime of productivity. The surface sediment condition favours BF abundance and domination by BF species tolerant to oxygen deficiencies. During the Boreal period more stable surface conditions were accompanied by continuously high productivity and an intensifying of its mesopelagic regime. Significant regional climate warming since the Atlantic (9 kyr ago) strongly intensified the summer sea ice melting in the OS, and this created considerable surface environment amelioration with the preferential transport of bacteria and phytodetritus into the SOIW. Further considerable warming of the regional climate from 6 kyr ago contributed to slight sea ice changes, surface water warming, and the enhancement of its stratification; all typical for most of the OS. Along with a high nutrient supply from the Amur River, the NW OS experienced a strong diatom production increase with the maximum amount occurring during the last 3.6 kyr. This changed the productivity type and organic matter export into the water column while increasing the feeding of the ‘‘productive’’ Plagoniidae spp. group and decreasing the microbial biomass supply into the upper SOIW. Some sea surface water cooling or saltier conditions at the beginning of the Subatlantic (2.4–1.8 kyr) was followed by its warming or freshening 1.5–1.0 kyr ago, which likely correlated with the Medieval Warm Period. In turn, that probably led to strong surface water stratification, productivity deterioration and considerable changes in the overall NW OS environment. The established sequence of the northwestern OS environmental changes during the Late Glacial–Holocene is related to the Northern Hemispheric climate changes and was likely forced by atmospheric teleconnection in line with the polar circulation index variability

The aim of this research paper was a sanitary-toxicological study of the effectiveness and safety of developed functional property products, such as chopped semi-finished products from chicken with wheat bran and shortbread cake with cottage cheese and sea buckthorn, in preclinical studies on laboratory animals. Methods — The effect of new products on the experimental rats was studied using physiological, biochemical, pathomorphological and histological research methods. As a result, it was discovered that the inclusion of foods with wheat bran and fresh sea buckthorn into the diet of experimental animals had a positive effect on the behavioral reactions of rats, the increase of appetite and the rapid growth of animals. Results — The improvement in the metabolic processes physiology of the studied rats was noted in the biochemical and morphological blood parameters. In the experimental group of rats where carbohydrates were partially substituted with cottage cheese shortbread cake and sea buckthorn, a significant decrease in the total bilirubin content was noted; the level of cholesterol in the blood serum of experimental animals from group which received chicken cutlets with bran was 1.4-1.7 times lower than the cholesterol content in animals of the control and other experimental groups. At the same time, statistically significant differences in the number of red blood cells, white blood cells, platelets and hemoglobin level, the activity of alanine and aspartate aminotransferases, alkaline phosphatase and amylase, the level of total protein and the creatinine content were not revealed in the group of experimental rats compared with the control group of rats, receiving a standard diet. The histological data showed that the liver structure of the experimental animals had a more pronounced beam and capillary structure compared with the control group, and the condition of the villi and epithelium of the small intestine showed the positive physiological effect of the studied herbal additives in food technology with functional properties. Conclusion — The research results allow us to conclude that the developed products are functional, intended for the systematic use in the composition of food rations by all age groups of a healthy population, which reduces the risk of gastrointestinal and liver diseases
