    Parametric optimization and heat transfer analysis of a dual loop ORC (organic Rankine cycle) system for CNG engine waste heat recovery

    In this study, a dual loop ORC (organic Rankine cycle) system is adopted to recover exhaust energy, waste heat from the coolant system, and intercooler heat rejection of a six-cylinder CNG (compressed natural gas) engine. The thermodynamic, heat transfer, and optimization models for the dual loop ORC system are established. On the basis of the waste heat characteristics of the CNG engine over the whole operating range, a GA (genetic algorithm) is used to solve the Pareto solution for the thermodynamic and heat transfer performances to maximize net power output and minimize heat transfer area. Combined with optimization results, the optimal parameter regions of the dual loop ORC system are determined under various operating conditions. Then, the variation in the heat transfer area with the operating conditions of the CNG engine is analyzed. The results show that the optimal evaporation pressure and superheat degree of the HT (high temperature) cycle are mainly influenced by the operating conditions of the CNG engine. The optimal evaporation pressure and superheat degree of the HT cycle over the whole operating range are within 2.5–2.9 MPa and 0.43–12.35 K, respectively. The optimal condensation temperature of the HT cycle, evaporation and condensation temperatures of the LT (low temperature) cycle, and exhaust temperature at the outlet of evaporator 1 are kept nearly constant under various operating conditions of the CNG engine. The thermal efficiency of the dual loop ORC system is within the range of 8.79%–10.17%. The dual loop ORC system achieves the maximum net power output of 23.62 kW under the engine rated condition. In addition, the operating conditions of the CNG engine and the operating parameters of the dual loop ORC system significantly influence the heat transfer areas for each heat exchanger

    The annulation reaction between various indoles and 2-alkoxycyclopropanoate esters is reported. Both high efficiency and complete stereochemical control were observed in some cases with this annulation process. A single stereocenter on the cyclopropane controls the diastereoselective formation of up to four new stereocenters. A different reaction course was observed with 3-substituted indole substrates, and an intervening C-3 to C-2-migration process arose that gives synthetically useful C-2 alkylation indole products

    Multi-stage collaborative efficiency measurement of scitech finance: network-DEA analysis and spatial impact research

    Sci-tech and finance plays an increasingly important role and have become an important driving force in economic development. In China, the problem of insufficient financial support for sci-tech innovation is important to enterprises. According to the internal relationship between different stages of Sci-tech and the finance system, this paper is aimed at exploring the efficiency measurement method between sci-tech and finance systems. Firstly the multi-stage collaborative structure of sci-tech finance is built, where the system of sci-tech is divided into three stages including the R&D stage, transformation stage of sci-tech achievements and industrialization stage, and the financing channel is the input of the finance system into the sci-tech system at different stages. The measurement method of the multi-stage collaborative efficiency between sci-tech and finance systems is put forward by the framework of network DEA. Then, taking China as an example, we collect the information of 30 provinces and cities from 2009 to 2016 and measure the efficiency of each system and the collaborative efficiency of the both. The efficiency’s spatial correlation is tested by means of Moran index. Finally, the influencing factors of the collaborative efficiency are analyzed based on the spatial econometric regression model, which considers the financing channels and human capital. To sum up, there are significant differences in the sci-tech finance collaborative efficiency among regions in China. Among them, the collaborative efficiency of Beijing, Shanghai and Jiangsu ranks in the top three. Comparing the different stages of the sci-tech system, the commercialization stage is a weak link in many regions of China. Human capital and financing channels of sci-tech finance have different degrees of positive impact on the sci-tech finance collaborative efficiency. Among them, human capital plays a greater role in promoting the sci-tech finance collaborative development

    Comparison of antigenicity and conformational changes to β-lactoglobulin following kestose glycation reaction with and without dynamic high-pressure microfluidization treatment

    Previous work indicated that conformational changes of β-lactoglobulin (β-LG) induced by dynamic high pressure microfluidization (DHPM) was related to the increase of antigenicity. In this study, β-LG glycated with 1-kestose and combined with DHPM decreased the antigenicity of β-LG. The antigenicity of control, β-LG-kestose (0.1 MPa) and β-LG-kestose (80 MPa) were 100, 79 and 42 μg/mL respectively. The molecular weight of β-LG conjugated to kestose increased from 18.4 to 19.6 kDa and its conformation scarcely changed. Conversely, combined with DHPM treatment (80 MPa), β-LG conjugated to kestose formed two conjugates with molecular weight of 18.8 and 19.8 kDa, respectively. Furthermore, the unfolding of β-LG as a result of the treatments is reflected by a decrease of intrinsic and synchronous fluorescence intensity and changes to the secondary structure. The conformational changes induced by DHPM and glycation treatments synergistically decrease the antigenicity of β-LG due to more masked or disrupted epitopes

    Misregulation of Alternative Splicing in a Mouse Model of Rett Syndrome

    Mutations in the human MECP2 gene cause Rett syndrome (RTT), a severe neurodevelopmental disorder that predominantly affects girls. Despite decades of work, the molecular function of MeCP2 is not fully understood. Here we report a systematic identification of MeCP2-interacting proteins in the mouse brain. In addition to transcription regulators, we found that MeCP2 physically interacts with several modulators of RNA splicing, including LEDGF and DHX9. These interactions are disrupted by RTT causing mutations, suggesting that they may play a role in RTT pathogenesis. Consistent with the idea, deep RNA sequencing revealed misregulation of hundreds of splicing events in the cortex of Mecp2 knockout mice. To reveal the functional consequence of altered RNA splicing due to the loss of MeCP2, we focused on the regulation of the splicing of the flip/flop exon of Gria2 and other AMPAR genes. We found a significant splicing shift in the flip/flop exon toward the flop inclusion, leading to a faster decay in the AMPAR gated current and altered synaptic transmission. In summary, our study identified direct physical interaction between MeCP2 and splicing factors, a novel MeCP2 target gene, and established functional connection between a specific RNA splicing change and synaptic phenotypes in RTT mice. These results not only help our understanding of the molecular function of MeCP2, but also reveal potential drug targets for future therapies

    Short-term outcomes of robot-assisted versus video-assisted thoracoscopic surgery for non-small cell lung cancer patients with neoadjuvant immunochemotherapy: a single-center retrospective study

    BackgroundNeoadjuvant immunochemotherapy has been increasingly applied to treat non-small cell lung cancer (NSCLC). However, the comparison between robotic-assisted thoracoscopic surgery (RATS) and video-assisted thoracoscopic surgery (VATS) in the feasibility and oncological efficacy following neoadjuvant immunochemotherapy is scarce. This study aims to assess the superiorities of RATS over (VATS) concerning short-term outcomes in treating NSCLC patients with neoadjuvant immunochemotherapy.MethodsNSCLC patients receiving RATS or VATS lobectomy following neoadjuvant immunochemotherapy at Shanghai Chest Hospital from 2019 to 2022 were retrospectively identified. Baseline clinical characteristics, perioperative outcomes, and survival profiles were analyzed.ResultsForty-six NSCLC patients with neoadjuvant immunochemotherapy were included and divided into the RATS (n=15) and VATS (n=31) groups. The baseline clinical characteristics and induction-related adverse events were comparable between the two groups (all p>0.050). The 30-day mortality in the RATS and VATS groups were 0% and 3.23%, respectively (p=1.000). Patients undergoing RATS were associated with reduced surgical-related intensive unit care (ICU) stay than those receiving VATS (0.0 [0.0-0.0] vs. 0.0 [0.0-1.0] days, p=0.026). Moreover, RATS assessed more N1 LNs (6.27 ± 1.94 vs 4.90 ± 1.92, p=0.042) and LN stations (3.07 ± 1.03 vs 2.52 ± 0.57, p=0.038) compared with VATS. By comparison, no difference was found in surgical outcomes, pathological results, and postoperative complications between the RATS and VATS groups (all p>0.050). Finally, RATS and VATS achieved comparable one-year recurrence-free survival (82.96% vs. 85.23%, p=0.821) and the timing of central nervous system, LN, and bone recurrences (all p>0.050).ConclusionRATS is safe and feasible for NSCLC patients with neoadjuvant immunochemotherapy, reducing surgical-related ICU stay, assessing increased N1 LNs and stations, and achieving similar survival profiles to VATS

    Isogenic Pairs of Wild Type and Mutant Induced Pluripotent Stem Cell (iPSC) Lines from Rett Syndrome Patients as In Vitro Disease Model

    Rett syndrome (RTT) is an autism spectrum developmental disorder caused by mutations in the X-linked methyl-CpG binding protein 2 (MECP2) gene. Excellent RTT mouse models have been created to study the disease mechanisms, leading to many important findings with potential therapeutic implications. These include the identification of many MeCP2 target genes, better understanding of the neurobiological consequences of the loss- or mis-function of MeCP2, and drug testing in RTT mice and clinical trials in human RTT patients. However, because of potential differences in the underlying biology between humans and common research animals, there is a need to establish cell culture-based human models for studying disease mechanisms to validate and expand the knowledge acquired in animal models. Taking advantage of the nonrandom pattern of X chromosome inactivation in female induced pluripotent stem cells (iPSC), we have generated isogenic pairs of wild type and mutant iPSC lines from several female RTT patients with common and rare RTT mutations. R294X (arginine 294 to stop codon) is a common mutation carried by 5–6% of RTT patients. iPSCs carrying the R294X mutation has not been studied. We differentiated three R294X iPSC lines and their isogenic wild type control iPSC into neurons with high efficiency and consistency, and observed characteristic RTT pathology in R294X neurons. These isogenic iPSC lines provide unique resources to the RTT research community for studying disease pathology, screening for novel drugs, and testing toxicology

    Baichuan 2: Open Large-scale Language Models

    Large language models (LLMs) have demonstrated remarkable performance on a variety of natural language tasks based on just a few examples of natural language instructions, reducing the need for extensive feature engineering. However, most powerful LLMs are closed-source or limited in their capability for languages other than English. In this technical report, we present Baichuan 2, a series of large-scale multilingual language models containing 7 billion and 13 billion parameters, trained from scratch, on 2.6 trillion tokens. Baichuan 2 matches or outperforms other open-source models of similar size on public benchmarks like MMLU, CMMLU, GSM8K, and HumanEval. Furthermore, Baichuan 2 excels in vertical domains such as medicine and law. We will release all pre-training model checkpoints to benefit the research community in better understanding the training dynamics of Baichuan 2.Comment: Baichuan 2 technical report.     Research Progress on Porous Carbon-Based Non-Precious Metal Electrocatalysts

    The development of efficient, stable, and economic electrocatalysts are key to the large-scale application of electrochemical energy conversion. Porous carbon-based non-precious metal electrocatalysts are considered to be the most promising materials to replace Pt-based catalysts, which are limited in large-scale applications due to high costs. Because of its high specific surface area and easily regulated structure, a porous carbon matrix is conducive to the dispersion of active sites and mass transfer, showing great potential in electrocatalysis. This review will focus on porous carbon-based non-precious metal electrocatalysts and summarize their new progress, focusing on the synthesis and design of porous carbon matrix, metal-free carbon-based catalysts, non-previous metal monatomic carbon-based catalyst, and non-precious metal nanoparticle carbon-based catalysts. In addition, current challenges and future trends will be discussed for better development of porous carbon-based non-precious metal electrocatalysts

    The Sliding and Overturning Analysis of Spent Fuel Storage Rack Based on Dynamic Analysis Model

    Spent fuel rack is the key equipment for the storage of spent fuel after refueling. In order to investigate the performance of the spent fuel rack under the earthquake, the phenomena including sliding, collision, and overturning of the spent fuel rack were studied. An FEM model of spent fuel rack is built to simulate the transient response under seismic loading regarding fluid-structure interaction by ANSYS. Based on D’Alambert’s principle, the equilibriums of force and momentum were established to obtain the critical sliding and overturning accelerations. Then 5 characteristic transient loadings which were designed based on the critical sliding and overturning accelerations were applied to the rack FEM model. Finally, the transient displacement and impact force response of rack with different gap sizes and the supporting leg friction coefficients were analyzed. The result proves the FEM model is applicable for seismic response of spent fuel rack. This paper can guide the design of the future’s fluid-structure interaction experiment for spent fuel rack
