    This final report covers three years and several phases of work in which instrumentation for the Planetary Instrument Definition and Development Program (PIDDP) were successfully developed. There were two main thrusts to this research: (1) to develop and test methods for electrostatically scanning detector field-of-views, and (2) to improve the mass resolution of plasma mass spectrometers to M/delta M approximately 25, their field-of-view (FOV) to 360 degrees, and their E-range to cover approximately 1 eV to 50 keV.     Imaging Polarimeter Arrays for Near-Millimeter Waves

    An integrated-circuit antenna array has been developed that images both polarization and intensity. The array consists of a row of antennas that lean alternately left and right, creating two interlaced sub-arrays that respond to different polarizations. The arrays and the bismuth bolometer detectors are made by a photoresist shadowing technique that requires only one photolithographic mask. The array has measured polarization at a wavelength of 800 µm with an absolute accuracy of 0.8° and a relative precision of 7 arc min. and has demonstrated nearly diffraction-Iimited resolutiort of a 20° step in polarization

    Time for change: a new training programme for morpho-molecular pathologists?

    The evolution of cellular pathology as a specialty has always been driven by technological developments and the clinical relevance of incorporating novel investigations into diagnostic practice. In recent years, the molecular characterisation of cancer has become of crucial relevance in patient treatment both for predictive testing and subclassification of certain tumours. Much of this has become possible due to the availability of next-generation sequencing technologies and the whole-genome sequencing of tumours is now being rolled out into clinical practice in England via the 100 000 Genome Project. The effective integration of cellular pathology reporting and genomic characterisation is crucial to ensure the morphological and genomic data are interpreted in the relevant context, though despite this, in many UK centres molecular testing is entirely detached from cellular pathology departments. The CM-Path initiative recognises there is a genomics knowledge and skills gap within cellular pathology that needs to be bridged through an upskilling of the current workforce and a redesign of pathology training. Bridging this gap will allow the development of an integrated 'morphomolecular pathology' specialty, which can maintain the relevance of cellular pathology at the centre of cancer patient management and allow the pathology community to continue to be a major influence in cancer discovery as well as playing a driving role in the delivery of precision medicine approaches. Here, several alternative models of pathology training, designed to address this challenge, are presented and appraised

    The distribution of H13CN in the circumstellar envelope around IRC+10216

    H13CN J=8-7 sub-millimetre line emission produced in the circumstellar envelope around the extreme carbon star IRC+10216 has been imaged at sub-arcsecond angular resolution using the SMA. Supplemented by a detailed excitation analysis the average fractional abundance of H13CN in the inner wind (< 5E15 cm) is estimated to be about 4E-7, translating into a total HCN fractional abundance of 2E-5 using the isotopic ratio 12C/13C=50. Multi-transitional single-dish observations further requires the H13CN fractional abundance to remain more or less constant in the envelope out to a radius of about 4E16 cm, where the HCN molecules are effectively destroyed, most probably, by photodissociation. The large amount of HCN present in the inner wind provides effective line cooling that can dominate over that generated from CO line emission. It is also shown that great care needs to be taken in the radiative transfer modelling where non-local, and non-LTE, effects are important and where the radiation field from thermal dust grains plays a major role in exciting the HCN molecules. The amount of HCN present in the circumstellar envelope around IRC+10216 is consistent with predicted photospheric values based on equilibrium chemical models and indicates that any non-equilibrium chemistry occurring in the extended pulsating atmosphere has no drastic net effect on the fractional abundance of HCN molecules that enters the outer envelope.     THE POLITICS OF CHANGE, PRECARIATISED YOUTH UNEMPLOYMENT AND LABOUR MARKET POLICIES IN NIGERIA

    The insecurities suffered by the youth precariat class in Nigeria derive from labour market insecurity, employment insecurity, skill reproduction insecurity, income insecurity and representation insecurity, all of which point to largely steep economic, social and political insecurities. This labyrinth of insecurities, particularly as it manifests in bulging youth unemployment, further marginalises this class, and exacerbates its precarity. Since members of this class respond in the most vicious way by pulling at the fabric of social cohesion and peace or in the least by running a criminal economy aimed at achieving economic assuagement, serious commitment must be made by the political and economic managers of the state to address youth unemployment crisis in Nigeria. Particularly, a lot is expected from President Muhammadu Buhari whose anti-corruption and people-oriented policy disposition is expected to sire a democratic regime that will address the contradictions in the body polity and economy, paving way for greater youth employment in the country. This study adopts the Political Economy paradigm for its analysis and a qualitative approach in its collection of data to investigate how President Muhammadu Buhari’s Politics of Change can be combined with relevant labour market programmes in giving hope and a sense of belonging to the precariatised youth class in Nigeria. These disenchanted and precarious young Nigerians must be provided with decent livelihoods that can wean them off their resort to criminality and antisocial behaviours which some have adopted as survivalist measures to escape the hard social and economic realities in the countr

    Promoter Binding by the Adr1 Transcriptional Activator May Be Regulated by Phosphorylation in the DNA-Binding Region

    Background: Post-translational modification regulates promoter-binding by Adr1, a Zn-finger transcriptional activator of glucose-regulated genes. Support for this model includes the activation of an Adr1-dependent gene in the absence of Adr1 protein synthesis, and a requirement for the kinase Snf1 for Adr1 DNA-binding. A fusion protein with the Adr1 DNA-binding domain and a heterologous activation domain is glucose-regulated, suggesting that the DNA binding region is the target of regulation. Methodology/Principal Findings: Peptide mapping identified serine 98 adjacent to the Zn-fingers as a phosphorylation site. An antibody specific for phosphorylated serine 98 on Adr1 showed that the level of phosphorylated Adr1 relative to the level of total Adr1 decreased with glucose derepression, in a Snf1-dependent manner. Relative phosphorylation decreased in a PHO85 mutant, and this mutant constitutively expressed an Adr1-dependent reporter. Pho85 did not phosphorylate Adr1 in vitro, suggesting that it affects Adr1 indirectly. Mutation of serine 98 to the phosphomimetic amino acid aspartate reduced in vitro DNA-binding of the recombinant Adr1 DNA-binding domain. Mutation to aspartate or alanine affected activation of a reporter by full-length Adr1, and in vivo promoter binding. Conclusions/Significance: Mutation of Adr1 serine 98 affects in vitro and in vivo DNA binding, and phosphorylation of serine 98 in vivo correlates with glucose availability, suggesting that Adr1 promoter-binding is regulated in part by serine 98 phosphorylation
