    Rapid and Efficient Extraction and HPLC Analysis of Sesquiterpene Lactones from Aucklandia lappa Root.

    The root of Aucklandia lappa Decne, family Asteraceae, is widely used in Asian traditional medicine due to its sesquiterpene lactones. The aim of this study was the development and optimization of the extraction and analysis of these sesquiterpene lactones. The current Chinese Pharmacopoeia reports a monograph for "Aucklandiae Radix", but the extraction method is very long and tedious including maceration overnight and ultrasonication. Different extraction protocols were evaluated with the aim of optimizing the maceration period, solvent, and shaking and sonication times. The optimized method consists of only one hour of shaking plus 30 minutes of sonication using 100% MeOH as solvent. 1H NMR spectroscopy was used as a complementary analytical tool to monitor the residual presence of sesquitepene lactones in the herbal material. A suitable LC-DAD method was set up to quantify the sesquiterpene lactones. Recovery was ca. 97%, but a very high instability of constituents was found after powdering the herbal drug. A loss of about 20% of total sesquiterpenes was found after 15–20 days; as a consequence, it is strongly endorsed to use fresh powdered herbal material to avoid errors in the quantification

    Experimental Infection of Rabbits with Rabbit and Genotypes 1 and 4 Hepatitis E Viruses

    Background: A recent study provided evidence that farmed rabbits in China harbor a novel hepatitis E virus (HEV) genotype. Although the rabbit HEV isolate had 77-79% nucleotide identity to the mammalian HEV genotypes 1 to 4, their genomic organization is very similar. Since rabbits are used widely experimentally, including as models of infection, we investigated whether they constitute an appropriate animal model for human HEV infection.Methods: Forty-two SPF rabbits were divided randomly into eleven groups and inoculated with six different isolates of rabbit HEV, two different doses of a second-passage rabbit HEV, and with genotype 1 and 4 HEV. Sera and feces were collected weekly after inoculation. HEV antigen, RNA, antibody and alanine aminotransferase in sera and HEV RNA in feces were detected. The liver samples were collected during necropsy subject to histopathological examination.Findings: Rabbits inoculated with rabbit HEV became infected with HEV, with viremia, fecal virus shedding and high serum levels of viral antigens, and developed hepatitis, with elevation of the liver enzyme, ALT. The severity of disease corresponded to the infectious dose (genome equivalents), with the most severe hepatic disease caused by strain GDC54-18. However, only two of nine rabbits infected with HEV genotype 4, and none infected with genotype 1, developed hepatitis although six of nine rabbits inoculated with the genotype 1 HEV and in all rabbits inoculated with the genotype 4 HEV seroconverted to be positive for anti-HEV IgG antibody by 14 weeks post-inoculation.Conclusions: These data indicate that rabbits are an appropriate model for rabbit HEV infection but are not likely to be useful for the study of human HEV. The rabbit HEV infection of rabbits may provide an appropriate parallel animal model to study HEV pathogenesis

    A Novel High-Throughput Vaccinia Virus Neutralization Assay and Preexisting Immunity in Populations from Different Geographic Regions in China

    Background: Pre-existing immunity to Vaccinia Tian Tan virus (VTT) resulting from a large vaccination campaign against smallpox prior to the early 1980s in China, has been a major issue for application of VTT-vector based vaccines. It is essential to establish a sensitive and high-throughput neutralization assay to understand the epidemiology of Vaccinia-specific immunity in current populations in China. Methodology/Principal Findings: A new anti-Vaccinia virus (VACV) neutralization assay that used the attenuated replication-competent VTT carrying the firefly luciferase gene of Photinus pyralis (rTV-Fluc) was established and standardized for critical parameters that included the choice of cell line, viral infection dose, and the infection time. The current study evaluated the maintenance of virus-specific immunity after smallpox vaccination by conducting a non-randomized, crosssectional analysis of antiviral antibody-mediated immune responses in volunteers examined 30–55 years after vaccination. The rTV-Fluc neutralization assay was able to detect neutralizing antibodies (NAbs) against Vaccinia virus without the ability to differentiate strains of Vaccinia virus. We showed that the neutralizing titers measured by our assay were similar to those obtained by the traditional plaque reduction neutralization test (PRNT). Using this assay, we found a low prevalence of NAb to VTT (7.6%) in individuals born before 1980 from Beijing and Anhui provinces in China, and when present, anti-VTT NAb titers were low.     Biophysical Property and Broad Anti-HIV Activity of Albuvirtide, a 3-Maleimimidopropionic Acid-Modified Peptide Fusion Inhibitor

    Albuvirtide (ABT) is a 3-maleimimidopropionic acid (MPA)-modified peptide HIV fusion inhibitor that can irreversibly conjugate to serum albumin. Previous studies demonstrated its in vivo long half-life and potent anti-HIV activity. Here, we focused to characterize its biophysical properties and evaluate its antiviral spectrum. In contrast to T20 (Enfuvirtide, Fuzeon), ABT was able to form a stable α-helical conformation with the target sequence and block the fusion-active six-helix bundle (6-HB) formation in a dominant-negative manner. It efficiently inhibited HIV-1 Env-mediated cell membrane fusion and virus entry. A large panel of 42 HIV-1 pseudoviruses with different genotypes were constructed and used for the antiviral evaluation. The results showed that ABT had potent inhibitory activity against the subtypes A, B and C that predominate the worldwide AIDS epidemics, and subtype B′, CRF07_BC and CRF01_AE recombinants that are currently circulating in China. Furthermore, ABT was also highly effective against HIV-1 variants resistant to T20. Taken together, our data indicate that the chemically modified peptide ABT can serve as an ideal HIV-1 fusion inhibitor

    Development of a World Health Organization International Reference Panel for different genotypes of hepatitis E virus for nucleic acid amplification testing.

    Globally, hepatitis E virus (HEV) is a major cause of acute viral hepatitis. Epidemiology and clinical presentation of hepatitis E vary greatly by location and are affected by the HEV genotype. Nucleic acid amplification technique (NAT)-based assays are important for the detection of acute HEV infection as well for monitoring chronic cases of hepatitis E. The aim of the study was to evaluate a panel of samples containing different genotypes of HEV for use in nucleic NAT-based assays. The panel of samples comprises eleven different members including HEV genotype 1a (2 strains), 1e, 2a, 3b, 3c, 3e, 3f, 4c, 4g as well as a human isolate related to rabbit HEV. Each laboratory assayed the panel members directly against the 1 World Health Organization (WHO) International Standard (IS) for HEV RNA (6329/10) which is based upon a genotype 3 a strain. The samples for evaluation were distributed to 24 laboratories from 14 different countries and assayed on three separate days. Of these, 23 participating laboratories returned a total of 32 sets of data; 17 from quantitative assays and 15 from qualitative assays. The assays used consisted of a mixture of in-house developed and commercially available assays. The results showed that all samples were detected consistently by the majority of participants, although in some cases, some samples were detected less efficiently. Based on the results of the collaborative study the panel (code number 8578/13) was established as the "1st International Reference Panel (IRP) for all HEV genotypes for NAT-based assays" by the WHO Expert Committee on Biological Standardization. This IRP will be important for assay validation and ensuring adequate detection of different genotypes and clinically important sub-genotypes of HEV

    UB-LSTM: A Trajectory Prediction Method Combined with Vehicle Behavior Recognition

In order to make an accurate prediction of vehicle trajectory in a dynamic environment, a Unidirectional and Bidirectional LSTM (UB-LSTM) vehicle trajectory prediction model combined with behavior recognition is proposed, and then an acceleration trajectory optimization algorithm is proposed. Firstly, the interactive information with the surrounding vehicles is obtained by calculation, then the vehicle behavior recognition model is established by using LSTM, and the vehicle information is input into the behavior recognition model to identify vehicle behavior. Then, the trajectory prediction model is established based on Unidirectional and Bidirectional LSTM, and the identified vehicle behavior and the input information of the behavior recognition model are input into the trajectory prediction model to predict the horizontal and vertical speed and coordinates of the vehicle in the next 3 seconds. Experiments are carried out with NGSIM data sets, and the experimental results show that the mean square error (MSE) between the predicted trajectory and the actual trajectory obtained by this method is 0.124, which is 97.2% lower than that of the method that does not consider vehicle behavior and directly predicts the trajectory. The test loss is 0.000497, which is 95.68% lower than that without considering vehicle behavior. The predicted trajectory is obviously optimized, closer to the actual trajectory, and the performance is more stable

    This paper designs the integrated charging station of PV and hydrogen storage based on the charging station. The energy storage system includes hydrogen energy storage for hydrogen production, and the charging station can provide services for electric vehicles and hydrogen vehicles at the same time. To improve the independent energy supply capacity of the hybrid charging station and reduce the cost, the components are reasonably configured. To minimize the configuration cost of the integrated charging station and the proportion of power purchase to the demand of the charging station, the energy flow strategy of the integrated charging station is designed, and the optimal configuration model of optical storage capacity is constructed. The NSGA-II algorithm optimizes the non-inferior Pareto solution set, and a fuzzy comprehensive evaluation evaluates the optimal configuration
