    TCC (graduação) - Universidade Federal de Santa Catarina, Centro Sócio Econômico, Curso de Administração.Este trabalho refere-se à análise da influência exercida do Centro de Cultura e Eventos da Universidade Federal de Santa Catarina sobre o mercado de eventos na região metropolitana de Florianópolis. Esta pesquisa é considerada como descritiva, com o intuito de analisar a influência de um fenômeno, contudo, sem aprofundar no seu procedimento. A coleta de dados primários foi realizada por meio de duas entrevistas não-estruturadas com dois funcionários de alto escalão das organizações CentroSul e Centro de Cultura e Eventos da UFSC. Em um segundo momento, parte significante dos dados primários foram obtidos de outros trabalhos acadêmicos. Analisando os dados populacionais recolhidos foi possível perceber uma tendência de crescimento no movimento de turistas, assim como um incremento na receita estimada para os próximos anos. Como parte do estudo de turismo de eventos, foi realizada uma análise das condições políticas, econômicas, científicas e industriais da cidade de Florianópolis, sendo identificada grande diversidade de opções turísticas oferecidas. Assim como as condições de estrutura turística, tendo como base instituições como hotéis e resorts de alto padrão. Por fim, este trabalho foca no relacionamento entre a realização de eventos e a Universidade Federal de Santa Catarina (UFSC).     Generation of Stratified Squamous Epithelial Progenitor Cells from Mouse Induced Pluripotent Stem Cells

    Background: Application of induced pluripotent stem (iPS) cells in regenerative medicine will bypass ethical issues associated with use of embryonic stem cells. In addition, patient-specific IPS cells can be useful to elucidate the pathophysiology of genetic disorders, drug screening, and tailor-made medicine. However, in order to apply iPS cells to mitotic tissue, induction of tissue stem cells that give rise to progeny of the target organ is required. Methodology/Principal Findings: We induced stratified epithelial cells from mouse iPS cells by co-culture with PA6 feeder cells (SDIA-method) with use of BMP4. Clusters of cells positive for the differentiation markers KRT1 or KRT12 were observed in KRT14-positive colonies. We successfully cloned KRT14 and p63 double-positive stratified epithelial progenitor cells from iPS-derived epithelial cells, which formed stratified epithelial sheets consisting of five- to six-polarized epithelial cells in vitro. When these clonal cells were cultured on denuded mouse corneas, a robust stratified epithelial layer was observed with physiological cell polarity including high levels of E-cadherin, p63 and K15 expression in the basal layer and ZO-1 in the superficial layer, recapitulating the apico-basal polarity of the epithelium in vivo.     Deficiency of triad junction and contraction in mutant skeletal muscle lacking junctophilin type 1

    In skeletal muscle excitation–contraction (E–C) coupling, the depolarization signal is converted from the intracellular Ca2+ store into Ca2+ release by functional coupling between the cell surface voltage sensor and the Ca2+ release channel on the sarcoplasmic reticulum (SR). The signal conversion occurs in the junctional membrane complex known as the triad junction, where the invaginated plasma membrane called the transverse-tubule (T-tubule) is pinched from both sides by SR membranes. Previous studies have suggested that junctophilins (JPs) contribute to the formation of the junctional membrane complexes by spanning the intracellular store membrane and interacting with the plasma membrane (PM) in excitable cells. Of the three JP subtypes, both type 1 (JP-1) and type 2 (JP-2) are abundantly expressed in skeletal muscle. To examine the physiological role of JP-1 in skeletal muscle, we generated mutant mice lacking JP-1. The JP-1 knockout mice showed no milk suckling and died shortly after birth. Ultrastructural analysis demonstrated that triad junctions were reduced in number, and that the SR was often structurally abnormal in the skeletal muscles of the mutant mice. The mutant muscle developed less contractile force (evoked by low-frequency electrical stimuli) and showed abnormal sensitivities to extracellular Ca2+.     Hydrogen-bond-assisted isotactic-specific radical polymerization of N-vinyl-2-pyrrolidone with tartrate additives in toluene at low temperatures : high-resolution 1H NMR analysis

    A diethyl L-tartrate (L-EtTar)-assisted radical polymerization of N-vinyl-2-pyrrolidone has been developed as the first reported example of the synthesis of isotactic-rich poly(N-vinyl-2-pyrrolidone) (PVP). The addition of L-EtTar in toluene at temperatures of –40°C and lower led to a significant increase in the polymer yield by one order of magnitude compared with the reaction in the absence of L-EtTar. Decreasing the polymerization temperature led to increases in the isotacticity of the PVP, with the mm triad reaching 66.4% at −93 °C. 1H NMR measurement at 920 MHz was conducted to establish a reliable strategy for quantifying the triad tacticities. High-temperature NMR measurements at 250 °C were performed using a specially-designed NMR probe, which led to dramatic narrowing of the 1H line width

    Image-guided core needle biopsy for musculoskeletal lesions

    Background: Image-guided percutaneous core needle biopsy (CNB) has been an important diagnostic procedure for musculoskeletal lesions. Here we surveyed the variety of diagnostic strategies available and assessed the clinical usefulness and limitations of image-guided CNB carried out by a multidisciplinary team comprising specialists in various fields. Methods: We conducted a retrospective study of 284 image-guided CNBs among 1899 consecutive biopsy procedures carried out at our institution for musculoskeletal tumorous conditions, focusing on their effectiveness including diagnostic accuracy and utility for classification of specimens according to malignant potential and histological subtype as well as their correlation with biopsy routes. Results: Among the 284 studied biopsies, 252 (88.7%) were considered clinically “effective”. The sensitivity for detection of malignancy was 94.0% (110/117) and the specificity was 95.3% (41/43). The diagnostic accuracy for detection of malignancy was 94.4% (151/160) and that for histological subtype was 92.3% (48/52). The clinical effectiveness of the procedure was correlated with the complexity of the biopsy route (P = 0.015); the trans-pedicular, trans-retroperitoneal and trans-sciatic foramen approaches tended to yield ineffective results. Repeat biopsy did not have a significant impact on the effectiveness of image-guided CNB (P = 0.536). Conclusions: The diagnostic accuracy rates of image-guided CNB performed at multidisciplinary sarcoma units were usable even for patients who have variety of diagnostic biopsy procedures. It is important to establish and implement diagnostic strategies based on an understanding that complicated routes, especially for spine and pelvic lesions, may be associated with ineffectiveness and/or complications

    An 82-year-old man had systemic rash and itching. He was treated with antihistamines and steroids, but did not improve. We continued treatment at our hospital, but there was no improvement.The deck-chair sign was observed during treatment, and we diagnosed papuloerythroderma. We considered the possibility of merged malignant tumors and carried out a detailed examination.The final diagnosis was clinical stage I rectal cancer. The patient underwent laparoscopic low anterior resection. The refractory systemic rash completely improved by the 23th postoperative day. At present, the rectal cancer has not relapsed for 4 years after the operation

    Structure and expression of two highly related genes encoding SCM-1/human lymphotactin

    AbstractSCM-1/lymphotactin is a chemokine-like molecule produced selectively, if not exclusively, by activated CD8+ T cells. Here we report that there are two highly homologous SCM-1 genes, which we designate as SCM-1α and SCM-1β. Both genes have three exons and two introns. The 1st intron of SCM-1α contains a pseudogene of the ribosomal large subunit L7a. In SCM-1β, a 1.5-kb region including about a quarter of the L7a pseudogene is deleted from the 1st intron. Otherwise, the two genes are highly homologous including the 5′ and 3′ flanking regions. Both genes were mapped to human chromosome 1q23. The two genes were similarly induced in peripheral blood mononuclear cells by mitogenic stimulation. Primer extension and RNase protection revealed several transcription initiation sites. The biological activities of SCM-1α and SCM-1β, which have two amino acid differences at positions 7 and 8 in the mature proteins, remain to be compared
