    Get PDF
At present, the air freight rate does not match with the current competitive environment, and the revenue level of air freight is not ideal. How to improve the level of income management and balance the relationship between quantity and price has become an urgent problem for China’s air freight. In this context, this paper constructs an air freight pricing model based on revenue management. We review the application models of the air freight, and select the parameters for pricing model. Then the pricing model is constructed based on the revenue management and the status quo of China’s air freight industry. Finally, the model is tested by simulation analysis. This paper aims to provide a scientific and reasonable decision-making reference for air freight pricing through the improvement of pricing model

Combination of dexmedetomidine and diazepam in treatment of a patient with severe alcohol withdrawal syndrome complicated with fever

Long-term alcoholic individuals may develop alcohol withdrawal syndrome (AWS) after abrupt cessation or reduction of alcohol intake. Mild symptoms include involuntary limb trembling, sweating, nausea and vomiting, anxiety and disorder of sleep rhythm, and severe signs consist of hallucination, delirium and even coma. AWS is likely to progress into severe AWS when unrecognized or untreated, which is a potential cause of death. Benzodiazepines are the optimal drugs for the treatment of AWS. Nevertheless, large-dose benzodiazepines alone fail to completely control the symptoms of acute-stage AWS, and it should be combined with other drugs when necessary. In this article, we reported one 41-year-old male case of acute AWS complicated with fever who progressed into severe AWS. The disease condition was finally controlled by combined use of dexmedetomidine and diazepam, a widely prescribed benzodiazepine. The diagnosis and treatment of this case prompt that dexmedetomidine can decrease the dosage of diazepam, ease respiratory depression and mitigate the disorder of sleep rhythm

Trefoil Factor 3, Cholinesterase and Homocysteine: Potential Predictors for Parkinson\u27s Disease Dementia and Vascular Parkinsonism Dementia in Advanced Stage

Trefoil factor 3 (TFF3), cholinesterase activity (ChE activity) and homocysteine (Hcy) play critical roles in modulating recognition, learning and memory in neurodegenerative diseases, such as Parkinson\u27s disease dementia (PDD) and vascular parkinsonism with dementia (VPD). However, whether they can be used as reliable predictors to evaluate the severity and progression of PDD and VPD remains largely unknown. METHODS: We performed a cross-sectional study that included 92 patients with PDD, 82 patients with VPD and 80 healthy controls. Serum levels of TFF3, ChE activity and Hcy were measured. Several scales were used to rate the severity of PDD and VPD. Receivers operating characteristic (ROC) curves were applied to map the diagnostic accuracy of PDD and VPD patients compared to healthy subjects. RESULTS: Compared with healthy subjects, the serum levels of TFF3 and ChE activity were lower, while Hcy was higher in the PDD and VPD patients. These findings were especially prominent in male patients. The three biomarkers displayed differences between PDD and VPD sub-groups based on genders and UPDRS (III) scores\u27 distribution. Interestingly, these increased serum Hcy levels were significantly and inversely correlated with decreased TFF3/ChE activity levels. There were significant correlations between TFF3/ChE activity/Hcy levels and PDD/VPD severities, including motor dysfunction, declining cognition and mood/gastrointestinal symptoms. Additionally, ROC curves for the combination of TFF3, ChE activity and Hcy showed potential diagnostic value in discriminating PDD and VPD patients from healthy controls. CONCLUSIONS: Our findings suggest that serum TFF3, ChE activity and Hcy levels may underlie the pathophysiological mechanisms of PDD and VPD. As the race to find biomarkers or predictors for these diseases intensifies, a better understanding of the roles of TFF3, ChE activity and Hcy may yield insights into the pathogenesis of PDD and VPD

Activation of JNK Signaling Mediates Connective Tissue Growth Factor Expression and Scar Formation in Corneal Wound Healing

    Get PDF
Connective Tissue Growth Factor (CTGF) and Transforming growth factor-β1 (TGF-β1) are key growth factors in regulating corneal scarring. Although CTGF was induced by TGF-β1 and mediated many of fibroproliferative effects of TGF-β1, the signaling pathway for CTGF production in corneal scarring remains to be clarified. In the present study, we firstly investigated the effects of c-Jun N-terminal kinase (JNK) on CTGF expression induce by TGF-β1 in Telomerase-immortalized human cornea stroma fibroblasts (THSF). Then, we created penetrating corneal wound model and determined the effect of JNK in the pathogenesis of corneal scarring. TGF-β1 activated MAPK pathways in THSF cells. JNK inhibitor significantly inhibited CTGF, fibronectin and collagen I expression induced by TGF-β1 in THSF. In corneal wound healing, the JNK inhibitor significantly inhibited CTGF expression, markedly improved the architecture of corneal stroma and reduced corneal scar formation, but did not have a measurable impact on corneal wound healing in vivo. Our results indicate that JNK mediates the expression of CTGF and corneal scarring in corneal wound healing, and might be considered as specific targets of drug therapy for corneal scarring

In this research subtopic, an electro-hydraulic servo four-legged heavy load (FLHL) robot has been designed and developed. This paper proposes an integration layout cylinder design scheme for a non-lightweight hydraulic servo four-legged robot with high loads and torques of hip joint, and derives the mathematical element analysis model for a parallel hydraulic servo cylinder system. The multiple inherent characteristics of the parallel-executed cylinder integration system model are further explored. Based on the controllable functional requirements of interconnected joints and weakening the influence of internal force coupling, a design idea of force/position hybrid control scheme for the parallel-executed cylinder is determined, and then the force/position signal module design unit is used to reversely solve the force/position hybrid control. Considering the inherent requirements of the servo-executed cylinder force control unit module, the implementation process of magnetic flux compensation and speed compensation is discussed in detail. The minimum amplitude controller is applied to the servo-executed cylinder force unit module, and the proportional integrated controller has been determined in the servo-executed cylinder position control unit module. A compound control strategy proposed in this paper is verified on a parallel hydraulic servo platform. The experimental verification results reveal that the values of position/force attenuation amplitude and lag phase are not greater than 9 % and 18°, respectively. In addition, the feasibility of the interconnected implementation of the hybrid control scheme proposed in this paper is further deepened. The effective conclusion of this research will be accepted in the application field of FLHL robot control system

Semi-numerical analysis of a two-stage series composite planetary transmission considering IHB and MsP methods

This study conducts an analytical investigation of the dynamic response characteristics of a two-stage series composite system (TsSCS) with a planetary transmission consisting of dual-power branches. An improved incremental harmonic balance (IHB) method, based on the arc length extension technique, is proposed. The results are compared with those of the numerical integration method to verify the feasibility and effectiveness of the improved method. Following that, the multi-scale perturbation (MsP) method is applied to the TsSCS proposed in this paper. The frequency response equations of the primary resonance, subharmonic resonance, and superharmonic resonance are solved to generate the frequency response characteristic curves of the planetary transmission system in this method. A comparison between the results obtained by the MsP method and the numerical integration method proves that the former is ideal and credible in most regions. Considering the parameters of TsSCS excitation frequency and damping, the nonlinear response characteristics of steady-state motion are mutually converted. The effects of the time-varying parameters and the nonlinear deenthing caused by the gear teeth clearance on the amplitude-frequency characteristics of TsSCS components are studied in this special topic

MiR-4319 Suppress the Malignancy of Triple-Negative Breast Cancer by Regulating Self-Renewal and Tumorigenesis of Stem Cells

Background/Aims: High levels of cancer stem cells (CSCs) in patients with triple-negative breast cancer (TNBC) correlate with risk of poor clinical outcome and possibly contribute to chemoresistance and metastasis in patients with highly malignant TNBC. Aberrant microRNA expression is associated with the dysfunction of self-renewal and proliferation in cancer stem cells, while there is little information about the TNBC-specific microRNAs in regulating CSC ability. Methods: Solexa deep sequencing was performed to detect the expression levels of TNBC or non-TNBC stem cells (CSCs) microRNAs. Mammosphere formation assay, qRT-PCR and the xenograft model in nude mice were performed. Bioinformatic analysis and microarray were used to select the target gene, and luciferase reporter assays were used to confirm the binding sites. Results: Solexa sequencing data exhibited differential expression of 193 microRNAs between TNBC and non-TNBC stem cells. The gene ontology analysis and pathways analyses showed that genes were involved in the maintenance of stemness. MiR-4319 could suppress the self-renewal and formation of tumorspheres in TNBC CSCs through E2F2, and also inhibited tumor initiation and metastasis in vivo. Moreover, increased E2F2 could reverse the effect of miR-4319 on the self-renewal in TNBC CSCs. Conclusions: MiR-4319 suppresses the malignancy of TNBC by regulating self-renewal and tumorigenesis of stem cells and might be a remarkable prognostic factor or therapeutic target for patients with TNBC

Background/Aims: Resistance to trastuzumab remains a common challenge to HER-2 positive breast cancer. Up until now, the underlying mechanism of trastuzumab resistance is still unclear. tRNA-derived small non-coding RNAs, a new class of small non-coding RNA (sncRNAs), have been observed to play an important role in cancer progression. However, the relationship between tRNA-derived fragments and trastuzumab resistance is still unknown. Methods: We detected the levels of tRNA-derived fragments expression in normal breast epithelial cell lines, trastuzumab-sensitive and -resistant breast cancer cell lines using high-throughput sequencing. qRT-PCR was conducted to validate the differentially expressed fragments in serums from trastuzumab-sensitive and -resistant patients. A receiver operating characteristic (ROC) curve analysis was performed to evaluate the power of specific tRNA-derived fragments. Progression-free survival (PFS) was analyzed using Cox-regression. Results: Our sequence results showed that tRNA-derived fragments were differentially expressed in the HBL-100, SKBR3, and JIMT-1 cell lines. tRF-30-JZOYJE22RR33 and tRF-27-ZDXPHO53KSN were found significantly upregulated in trastuzumab-resistant patients compared to sensitive individuals, and the ROC analysis showed that tRF-30-JZOYJE22RR33 and tRF-27-ZDXPHO53KSN were correlated with trastuzumab resistance. In a multivariate analysis, higher levels of tRF-30-JZOYJE22RR33 and tRF-27-ZDXPHO53KSN expression were associated with significantly shorter PFS in patients with metastatic HER-2 positive breast cancer. Conclusion: Our results suggest that tRF-30-JZOYJE22RR33 and tRF-27-ZDXPHO53KSN play important roles in trastuzumab resistance. Patients with high levels of tRF-30-JZOYJE22RR33 and tRF-27-ZDXPHO53KSN expression benefitted less from trastuzumab-based therapy than those that express lower-levels of these molecules. tRF-30-JZOYJE22RR33 and tRF-27-ZDXPHO53KSN may be potential biomarkers and intervention targets in the clinical treatment of trastuzumab-resistant breast cancer

<p>Abstract</p> <p>Background</p> <p>Though cupping therapy has been used in China for thousands of years, there has been no systematic summary of clinical research on it.</p> <p>This review is to evaluate the therapeutic effect of cupping therapy using evidence-based approach based on all available clinical studies.</p> <p>Methods</p> <p>We included all clinical studies on cupping therapy for all kinds of diseases. We searched six electronic databases, all searches ended in December 2008. We extracted data on the type of cupping and type of diseases treated.</p> <p>Results</p> <p>550 clinical studies were identified published between 1959 and 2008, including 73 randomized controlled trials (RCTs), 22 clinical controlled trials, 373 case series, and 82 case reports. Number of RCTs obviously increased during past decades, but the quality of the RCTs was generally poor according to the risk of bias of the Cochrane standard for important outcome within each trials. The diseases in which cupping was commonly employed included pain conditions, herpes zoster, cough or asthma, etc. Wet cupping was used in majority studies, followed by retained cupping, moving cupping, medicinal cupping, etc. 38 studies used combination of two types of cupping therapies. No serious adverse effects were reported in the studies.</p> <p>Conclusions</p> <p>According to the above results, quality and quantity of RCTs on cupping therapy appears to be improved during the past 50 years in China, and majority of studies show potential benefit on pain conditions, herpes zoster and other diseases. However, further rigorous designed trials in relevant conditions are warranted to support their use in practice.</p
