    Uncovering the candidate genes related to sheep body weight using multi-trait genome-wide association analysis

    In sheep, body weight is an economically important trait. This study sought to map genetic loci related to weaning weight and yearling weight. To this end, a single-trait and multi-trait genome-wide association study (GWAS) was performed using a high-density 600 K single nucleotide polymorphism (SNP) chip. The results showed that 43 and 56 SNPs were significantly associated with weaning weight and yearling weight, respectively. A region associated with both weaning and yearling traits (OARX: 6.74–7.04 Mb) was identified, suggesting that the same genes could play a role in regulating both these traits. This region was found to contain three genes (TBL1X, SHROOM2 and GPR143). The most significant SNP was Affx-281066395, located at 6.94 Mb (p = 1.70 × 10−17), corresponding to the SHROOM2 gene. We also identified 93 novel SNPs elated to sheep weight using multi-trait GWAS analysis. A new genomic region (OAR10: 76.04–77.23 Mb) with 22 significant SNPs were discovered. Combining transcriptomic data from multiple tissues and genomic data in sheep, we found the HINT1, ASB11 and GPR143 genes may involve in sheep body weight. So, multi-omic anlaysis is a valuable strategy identifying candidate genes related to body weight

    The granary weevil Sitophilus granarius (L.) is one of the most serious primary insect pests of stored products. When S. granarius present in grains, various volatile organic compounds are released as chemical signals which can be used to detect the insects. In this study, volatile chemical compounds released from S. granarius were analyzed using the headspace solid phase micro-extraction (HS-SPME) coupled with gas chromatography (GC)–mass spectrometry (MS) techniques. Two key compounds, 3-hydroxy-2-butanone and 1-pentadecene, were identified from mixed gender of S. granarius adults at high density. Moreover, both male and female adults showed dose-dependent electroantennography (EAG) responses to 3-hydroxy-2-butanone. In behavioral assays, S. granarius was attracted to 3-hydroxy-2-butanone at 0.001 µg/10 µL but repelled at 10 µg/10 µL or higher. S. granarius was consistently repelled by 1-pentadecene at concentrations at 0.001 and 1000 µg/10 µL. 3-hydroxy-2-butanone and 1-pentadecene have considerable potential to offer in the development of new approaches for the monitoring and management of this destructive stored grain insect pest

    Identification of Biomarker Volatile Organic Compounds Released by Three Stored-Grain Insect Pests in Wheat

    Monitoring and early detection of stored-grain insect infestation is essential to implement timely and effective pest management decisions to protect stored grains. We report a reliable analytical procedure based on headspace solid-phase microextraction coupled with gas chromatography–mass spectrometry (HS-SPME-GC-MS) to assess stored-grain infestation through the detection of volatile compounds emitted by insects. Four different fibre coatings were assessed; 85 µm CAR/PDMS had optimal efficiency in the extraction of analytes from wheat. The headspace profiles of volatile compounds produced by Tribolium castaneum (Herbst), Rhyzopertha dominica (Fabricius), and Sitophilus granarius (Linnaeus), either alone or with wheat, were compared with those of non-infested wheat grains. Qualitative analysis of chromatograms showed the presence of different volatile compound profiles in wheat with pest infestation compared with the wheat controls. Wheat-specific and insect-specific volatile compounds were identified, including the aggregation pheromones, dominicalure-1 and dominicalure-2, from R. dominica, and benzoquinones homologs from T. castaneum. For the first time, the presence of 3-hydroxy-2-butanone was reported from S. granarius, which might function as an alarm pheromone.     Investigation of flow boiling heat transfer characteristic of mixture refrigerant L-41B in a horizontal smooth tube

    The saturated flow boiling heat transfer characteristics of L-41b, R1234ze(E)/R32 (27/73 mass%), inside an 8 mm ID horizontal tube were investigated. The experiment were carried out at the saturation temperature of 10 to 20°C with heat flux ranging from 5 to 10 kW/m2 and mass flux ranging from 200-500 kg/m2s. The influence of mass flux, heat flux, and quality on the heat transfer coefficients were examined and discussed. The experimental data of local heat transfer coefficients were compared with four well-known correlations available in literatures. Additionally, a new vapour-phase multiplier correlation to predict the local heat transfer coefficients of L-41b flow boiling inside smooth tubes was developed. The results show that the deviations of the new correlation are within –24.98% to +14.68% to the experimental data and that the 95% prediction values are within ±15%
