    Effects of binary solvent extraction system, extraction time and extraction temperature on phenolic antioxidants and antioxidant capacity from mengkudu (Morinda citrifolia).

    An investigation into the effects of ethanol concentration (0–100%, v/v), extraction time (20–120 min) and extraction temperature (25–65 °C) on the extraction of phenolic antioxidants from mengkudu (Morinda citrifolia) was performed using a single-factor experiment. Total phenolic content (TPC) and total flavonoid content (TFC) assays were used for determination of phenolic compounds. Antioxidant capacity was evaluated by measuring the scavenging effect on 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radicals. Experimental results showed that extraction conditions had significant effect on extraction of phenolic compounds and antioxidant capacities. The optimised conditions were 40% ethanol for 80 min at 65 °C, with values of 919.95 mg GAE/100 g DW for TPC, 472.73 mg CE/100 g DW for TFC, 791.71 μmol TEAC/100 g DW for ABTS and 1928.5 μmol TEAC/100 g DW for DPPH. TPC was significantly correlated with DPPH under the effects of ethanol concentration (r = 0.932) and extraction time (r = −0.938)

    Four new chamigrane derivatives, laurecomin A (1). laurecomin B (2), laurecomin C (3), and laurecomin D (4), one new naturally occurring sesquiterpene, 2,10-dibromo-3-chloro-7-chamigren-9-ol acetate (5), and three known halogenated structures, deoxyprepacifenol (6), 1-bromoselin-4(14),11-diene (7), and 9-bromoselin-4(14).11-diene (8), were isolated from the marine red alga Laurencia cornposita collected from Pingtan Island, China. The structures of these compounds were unambiguously established by 1D, 2D NMR and mass spectroscopic techniques.     Light reflection from a metal surface with subwavelength cavities

    The interaction of light with the localized/delocalized system, i.e., a metal surface with rectangular cavities of finite depth, has been studied. Reflection spectrum has been measured in the optical frequencies and resonant minima have been observed. We have developed an analytical model, which agrees well with the experiment. The localized waveguide resonance and delocalized surface resonance have been identified and discussed. The results may be useful for manipulating the coupling between light and matters.Comment: 11 pages, 3 figure

    Constraints on the cosmological parameters with three-parameter correlation of Gamma-ray bursts

    As one of the most energetic and brightest events, gamma-ray bursts (GRBs) can be treated as a promising probe of the high-redshift universe. Similar to type Ia supernovae (SNe Ia), GRBs with same physical origin could be treated as standard candles. We select GRB samples with the same physical origin, which are divided into two groups. One group is consisted of 31 GRBs with a plateau phase feature of a constant luminosity followed by a decay index of about -2 in the X-ray afterglow light curves, and the other has 50 GRBs with a shallow decay phase in the optical light curves. For the selected GRB samples, we confirm that there is a tight correlation between the plateau luminosity L0L_0, the end time of plateau tbt_b and the isotropic energy release Eγ,isoE_{\gamma,iso}. We also find that the L0tbEγ,isoL_0-t_b-E_{\gamma,iso} correlation is insensitive to the cosmological parameters and no valid limitations on the cosmological parameters can be obtained using this correlation. We explore a new three-parameter correlation L0L_0, tbt_b, and the spectral peak energy in the rest frame Ep,iE_{p,i} (L0tbEp,iL_0-t_b-E_{p,i}), and find that this correlation can be used as a standard candle to constrain the cosmological parameters. By employing the optical sample only, we find the constraints of Ωm=0.6970.278+0.402(1σ)\Omega_m = 0.697_{-0.278}^{+0.402}(1\sigma) for a flat Λ\LambdaCDM model. For the non-flat Λ\LambdaCDM model, the best-fitting results are Ωm=0.7130.278+0.346\Omega_m = 0.713_{-0.278}^{+0.346}, ΩΛ=0.9810.580+0.379(1σ)\Omega_{\Lambda} = 0.981_{-0.580}^{+0.379}(1\sigma). For the combination of the X-ray and optical smaples, we find Ωm=0.3130.125+0.179(1σ)\Omega_m = 0.313_{-0.125}^{+0.179}(1\sigma) for a flat Λ\LambdaCDM model, and Ωm=0.3440.112+0.176\Omega_m = 0.344_{-0.112}^{+0.176}, ΩΛ=0.7700.416+0.366(1σ)\Omega_{\Lambda} = 0.770_{-0.416}^{+0.366}(1\sigma) for a non-flat Λ\LambdaCDM model.Comment: Accepted for publication in The Astrophysical Journal, 13 pages, 9 figures and 2 table

    The inhibitory effect against collagen-induced arthritis by Schistosoma japonicum infection is infection stage-dependent

    <p>Abstract</p> <p>Background</p> <p>A long-term existing schistosome infection can aid in maintaining immuno-homeostasis, thus providing protection against various types of autoimmune diseases to the infected host. Such benefits have often been associated with acute or egg stage infection and with the egg-induced Th2 response. However, since schistosome infection undergoes different stages, each associated with a specific induction of Th responses, the requirements for the ability of the different stages of schistosome infection to protect against autoimmune disease has not been elucidated. The present study was designed to study whether different stages of schistosome infection offer unique protection in collagen-induced arthritis and its mechanisms.</p> <p>Results</p> <p>Arthritis susceptible strain DBA/1 male mice were infected with <it>Schistosoma japonicum </it>for either 2 weeks resulting in early stage infection or for 7 weeks resulting in acute or egg stage infection. Following <it>Schistosoma japonicum </it>infection, collagen II was administered to induce collagen-induced arthritis, an animal model for human rheumatoid arthritis. Infection by <it>Schistosoma japonicum </it>significantly reduced the severity and the incidence of experimental autoimmune collagen-induced arthritis. However, this beneficial effect can only be provided by a pre-established acute stage of infection but not by a pre-established early stage of the infection. The protection against collagen-induced arthritis correlated with reduced levels of anti-collagen II IgG, especially the subclass of IgG2a. Moreover, in protected mice increased levels of IL-4 were present at the time of collagen II injection together with sustained higher IL-4 levels during the course of arthritis development. In contrast, in unprotected mice minimal levels of IL-4 were present at the initial stage of collagen II challenge together with lack of IL-4 induction following <it>Schistosoma japonicum </it>infection.</p> <p>Conclusion</p> <p>The protective effect against collagen-induced arthritis provided by <it>Schistosoma japonicum </it>infection is infection stage-dependent. Furthermore, the ability of schistosomiasis to negatively regulate the onset of collagen-induced arthritis is associated with a dominant as well as long-lasting Th2 response at the initiation and development of autoimmune joint and systemic inflammation.</p

    Radio Plateaus in Gamma-Ray Burst Afterglows and Their Application in Cosmology

    The plateau phase in the radio afterglows has been observed in very few gamma-ray bursts (GRBs), and 27 radio light curves with plateau phase were acquired from the published literature in this article. We obtain the related parameters of the radio plateau, such as temporal indexes during the plateau phase (α1\alpha_1 and α2\alpha_2), break time (\Tbz) and the corresponding radio flux (FbF_{\rm b}). The two parameter Dainotti relation between the break time of the plateau and the corresponding break luminosity (\Lbz) in radio band is \Lbz \propto \Tbz^{-1.20\pm0.24}. Including the isotropic energy \Eiso and the peak energy \Epi, the three parameter correlations for the radio plateaus are written as \Lbz \propto \Tbz^{-1.01 \pm 0.24} \Eiso^{0.18 \pm 0.09} and \Lbz \propto \Tbz^{-1.18 \pm 0.27} \Epi^{0.05 \pm 0.28}, respectively. The correlations are less consistent with that of X-ray and optical plateaus, implying that radio plateaus may have a different physical mechanism. The typical frequencies crossing the observational band may be a reasonable hypothesis that causes the breaks of the radio afterglows. We calibrate GRBs empirical luminosity correlations as standard candle for constraining cosmological parameters, and find that our samples can constrain the flat Λ\LambdaCDM model well, while are not sensitive to non-flat Λ{\Lambda}CDM model. By combining GRBs with other probes, such as SN and CMB, the constraints on cosmological parameters are \om = 0.297\pm0.006 for the flat Λ{\Lambda}CDM model and \om = 0.283\pm0.008, \oL = 0.711\pm0.006 for the non-flat Λ{\Lambda}CDM model, respectively.Comment: 16 pages, 6 figures and 6 tables, accepted for publication in Ap

    Pro-Angiogenic Activity of Monocytic-Type Myeloid-Derived Suppressor Cells from Balb/C Mice Infected with Echinococcus Granulosus and the Regulatory Role of miRNAs

    Background/Aims: This study aims to predict the pro-angiogenic functions of monocytic-type myeloid-derived suppressor cells (M-MDSCs) derived from mice infected with Echinococcus granulosus. Methods: M-MDSCs were collected from Balb/c mice infected with E. granulosus and normal mice (control) and cultured in vitro. Human umbilical vein endothelial cells (HUVECs) were stimulated with the cell supernatant, and angiogenesis was investigated and analysed by the Angiogenesis module of the software NIH Image J. RNA was extracted from fresh isolated M-MDSCs and analysed with miRNA microarray; differentially expressed miRNAs and their potential functions were analysed through several bioinformatics tools. Finally, quantitative PCR was used to confirm the results of microarray analysis. Results: M-MDSCs from mice infected with E. granulosus could promote the formation of tubes from HUVECs in vitro. Moreover, vascular endothelial growth factor (VEGF) showed significantly high expression, whereas soluble fms-like tyrosine kinase-1 (sFlt-1) showed low expression at the transcriptional level in M-MDSCs from mice infected with E. granulosus. Microarray analysis of miRNAs showed that 28 miRNAs were differentially expressed in M-MDSCs from the two experimental mice groups, and 272 target genes were predicted using the microRNA databases TargetScan, PITA and microRNAorg. These target genes were mainly involved in the biological processes of intracellular protein transport, protein targeting to the lysosome and protein transport, and mainly located in the cytoplasm, neuronal cell body and membrane. Moreover, they were mainly involved in the molecular functions of protein binding, metal ion binding and SH3 domain binding. Further, the differentially expressed miRNAs were mainly enriched in the endocytosis, Wnt and axon guidance pathways, as well as the MAPK, focal adhesion, PI3K-Akt, cAMP, mTOR and TGF-β signalling pathways, which are linked to immunoregulation and angiogenesis based on the results of bioinformatics analysis with DIANA-miRPath 3.0. In addition, the expression of eight miRNAs was randomly verified by quantitative PCR independently in three mice infected with E. granulosus and three normal mice. Conclusion: M-MDSCs have a potential angiogenic role during E. granulosus infection, and miRNAs may play a role in the immune response and angiogenesis functions of M-MDSCs through regulation of the identified signalling pathways

    Distinct roles of NMB and GRP in itch transmission

    A key question in our understanding of itch coding mechanisms is whether itch is relayed by dedicated molecular and neuronal pathways. Previous studies suggested that gastrin-releasing peptide (GRP) is an itch-specific neurotransmitter. Neuromedin B (NMB) is a mammalian member of the bombesin family of peptides closely related to GRP, but its role in itch is unclear. Here, we show that itch deficits in mice lacking NMB or GRP are non-redundant and Nmb/Grp double KO (DKO) mice displayed additive deficits. Furthermore, both Nmb/Grp and Nmbr/Grpr DKO mice responded normally to a wide array of noxious stimuli. Ablation of NMBR neurons partially attenuated peripherally induced itch without compromising nociceptive processing. Importantly, electrophysiological studies suggested that GRPR neurons receive glutamatergic input from NMBR neurons. Thus, we propose that NMB and GRP may transmit discrete itch information and NMBR neurons are an integral part of neural circuits for itch in the spinal cord
