    Effects of reducing dietary crude protein and whole grain feeding on performance and amino acid metabolism in broiler chickens offered wheat-based diets

    A total of 336 off-sex, male Ross 308 chicks were offered seven dietary treatments from 7 to 35 days post-hatch; each treatment was offered to eight replicate cages with six birds per cage. Three wheat-based diets were formulated to declining crude protein (CP) levels of 215, 190 and 165 g/kg but with a constant energy density (12.70 MJ/kg), electrolyte balance (250 mEq/kg) and digestible lysine level (11.00 g/kg). In a 2 × 3 factorial arrangement birds were offered either 215 or 165 g/kg CP diets to which 0%, 12.5% and 25.0% whole gain was incorporated post-pelleting. In addition, a ground grain, 190 g/kg CP diet served as a seventh treatment. The assessed parameters included growth performance, relative gizzard, pancreas and abdominal fat-pad weights, nutrient utilisation, concentrations of free amino acid in portal (anterior mesenteric vein) and systemic (brachial vein) plasma and apparent jejunal and ileal amino acid digestibility coefficients and disappearance rates.     Extending daily feed access intervals does not influence lysine HCl utilization but enhances amino acid digestibilities in broiler chickens

    Off-sex, male Ross 308 chickens were offered maize-soy diets without and with 3.5 g/kg lysine monohydrochloride (HCl), which contained 10.0 or 12.8 g/kg digestible lysine, from 7 to 28 D post-hatch. Birds were permitted access to diets at intervals of 12, 16, and 20 h/day. Lysine HCl improved weight gain (1,465 vs. 1,417 g/bird; P 0.35) between lysine HCl and feed access intervals for parameters of growth performance were not observed, it was concluded that feed access intervals do not influence lysine utilization. The implications of these findings are discussed

    Effect of storage time on the characteristics of corn and efficiency of its utilization in broiler chickens

    Corn is one of the staple food and feed ingredients in China, therefore its storage is of particular importance. Corn is typically stored for 2 or more years in national barns before it is sold as a food or feed ingredient. However, the effects of stored corn in national barns on the animal performance and nutrient utilization have not been investigated thus far. This study attempted to determine the effects of storage time on the chemical and physical characteristics of corn and its nutritional value, broiler growth performance, and meat quality. Corn grains used in the present study were stored for 4 different periods, from 2 to 5 yr, under the same conditions in a building at the Beijing National Grain Storage Facility. A total of 240 birds in Exp. 1 and 90 birds in Exp. 2 were used to compare the effects of storage time on the utilization of nutrients of corn, the performance, and meat quality of broilers. The content of starch, crude protein, amino acids, fatty acids, and test weight generally decreased with increasing storage time. Corn stored for over 4 yr showed decreased catalase (CAT) and peroxidase (POD) activities and increased fat acidity. Body weight gain (BWG) and European production index (EPI) of broilers from 0 to 3 wk tended to decrease linearly with storage time (0.05  0.05). The digestibility of histidine and arginine, and C18:2 and C18:3 changed quadratically with storage time (P < 0.05). Collectively, the results suggest that the use of corn stored for 4 yr in animal feed decreased the performance and meat quality of broilers.     AIM2 Co-immunization with VP1 Is Associated with Increased Memory CD8 T Cells and Mounts Long Lasting Protection against Coxsackievirus B3 Challenge

    The recurrent Coxsackievirus B3 (CVB3) infection is the most important cause of intractable myocarditis which often leads to chronic myocarditis and even dilated cardiomyopathy. Therefore, enhanced DNA vaccines capable of memory CD8 T cells are essential for long-lasting immunological protection against CVB3 infection. In this study, absent in melanoma 2 (AIM2) was used as an adjuvant to enhance the induction of memory CD8 T cells elicited by VP1 (viral capsid protein 1) vaccine. Mice were intramuscularly injected with 50 μg AIM2 plasmid and equal amount of VP1 plasmid (pAIM2/pVP1) vaccine 4 times at 2 week-intervals. We observed that the protection of pAIM2/pVP1 vaccine against CVB3 challenge was evidenced by significantly improved cardiac function, reduced myocardial injuries, and increased survival rate when compared with immunization with pVP1. Co-immunization with pAIM2/pVP1 robustly augmented T lymphocytes proliferation and CVB3-specific cytotoxic T lymphocyte responses. Importantly, 16 weeks after the last immunization, pAIM2/pVP1 co-immunization significantly enhanced the expression of Bcl-6, SOCS3, and Sca-1 which are critical for memory CD8 T cells as compared with pVP1 immunization. Notably, CD8 T cells that are likely vaccine-induced memory T cells were responsible for the protective efficacy of pAIM2/pVP1 vaccine by abolition of a CD8 T cell immune response following a lethal dose of CVB3 infection. Our results indicate that AIM2-adjuvanted vaccine could be a potential and promising approach to promote a long-lasting protection against CVB3-induced myocarditis

    Data Grid provides distributed resources for dealing with large-scale applications that generate huge volume data sets, while the Peer-to-Peer architecture provides the maximum autonomy and scalability. Combing their features, we have built a P2P Data Grid prototype as a high-available coordinated platform for storing, processing, and mining the massive Geospatial data. Replication strategy is the most challenging issue in building such systems, which includes the replica production, dissemination, location, selection, updating, synchronization and consistency maintaining.     Abstract Background The effect of amylases combined with exogenous carbohydrase and protease in a newly harvested corn diet on starch digestibility, intestine health and cecal microbiota was investigated in broiler chickens. Methods Two hunderd and eighty-eight 5-day-old female chickens were randomly divided into six treatments: a newly harvested corn-soybean meal diet (control); control supplemented with 1,500 U/g α-amylase (Enzyme A); Enzyme A + 300 U/g amylopectase + 20,000 U/g glucoamylase (Enzyme B); Enzyme B + protease 10,000 U/g (Enzyme C); Enzyme C + xylanase 15,000 U/g (Enzyme D); and Enzyme D + cellulase 200 U/g + pectinase 1,000 U/g (Enzyme E). Growth performance, starch digestibility, digestive organ morphology, and intestinal microbiota were evaluated in the birds at 16 and 23 d of age. Results Compared with the control diet, supplementation with Enzyme A significantly decreased ileum lesion scoring at 16 d of age (P < 0.05); supplementation with Enzyme B or Enzyme C showed positive effects on ileal amylopectin and total starch digestibility (P < 0.05); Broilers fed with a diet supplemented with Enzyme D had a tendency to decrease body weight gain at 23 d. Enzyme E supplementation improved lesion scoring of jejunum and ileum at 16 d (P < 0.05), and increased ileal amylopectin or total starch digestibility at 23 d (P < 0.05). Supplementation of enzymes changed cecal microbiota diversity. High numbers of Campylobacter, Helicobacter and Butyricicoccus, Anaerostipes and Bifidobacterium, Sutterella and Odoribacter were the main genera detected in supplementations with Enzymes B, C, D, and E respectively. Conclusions Supplementation with amylase combined with glucoamylase or protease showed a beneficial effect on starch digestibility and intestinal microbiota diversity, and increased growth of broilers fed with newly harvested corn

    Combination of Xylanase and Debranching Enzymes Specific to Wheat Arabinoxylan Improve the Growth Performance and Gut Health of Broilers

    Arabinoxylan (AX) is the major antinutritional factor of wheat. This study evaluated the synergistic effects of xylanase and debranching enzymes (arabinofuranosidase [ABF] and feruloyl esterase [FAE]) on AX. During in vitro tests, the addition of ABF or FAE accelerated the hydrolysis of water-soluble AX (WE-AX) and water-insoluble AX (WU-AX) and produced more xylan oligosaccharides (XOS) than xylanase alone. XOS obtained from WE-AX stimulated greater proliferation of Lactobacillus brevis and Bacillus subtilis than did fructo-oligosaccharides (FOS) and glucose. During in vivo trials, xylanase increased the average daily growth (ADG), decreased the feed-conversion ratio (FCR), and reduced the digesta viscosity of jejunum and intestinal lesions of broilers fed a wheat-based diet on day 36. ABF or FAE additions further improved these effects. Broilers fed a combination of xylanase, ABF, and FAE exhibited the best growth. In conclusion, the synergistic effects among xylanase, ABF, and FAE increased AX degradation, which improve the growth performance and gut health of broilers

    Tumor-Penetrating Peptide Enhances Antitumor Effects of IL-24 Against Prostate Cancer

    The interleukin-24 (IL-24), a member of the IL-10–related cytokine gene family, is well known for its tumor suppressor activity in a broad spectrum of human tumors without damaging normal cells. However, poor tumor penetration remains a key problem for the efficacy of IL-24 as a treatment. iRGD is a novel tumor-specific peptide with unique tumor-penetrating and cell-internalizing properties. To enhance the tumor-penetrating and antitumor effects of IL-24, we engineered a recombinant protein consisting of the IL-24 fused to iRGD, which was named IL-24-iRGD. The aim of the present study was to investigate the antitumor effects of IL-24-iRGD in prostate cancer cells in vitro and in vivo. It was observed that IL-24-iRGD induced cell apoptosis, suppressed cell growth of PC-3 in vitro, and promoted protein penetration into tumors in vivo, whereas it had no effect on normal cell line RWPE-1. Then, PC-3 cells were subcutaneously injected into nude mice, and these tumor-bearing mice were administered with IL-24, IL-24-iRGD, or PBS via the tail vein. The IL-24– and IL-24-iRGD–treated groups exhibited tumor growth inhibition rates of 38.6% and 65.6%, respectively, when compared with the PBS-treated group. Besides, cell apoptosis was examined by TdT-mediated dUTP nick end labeling, and the expression of cleaved caspase-3 was analyzed by immunohistochemical staining. The results demonstrated that IL-24-iRGD induced apoptosis and inhibited the growth of PC-3 cells to a significantly greater extent when compared with IL-24 treatment alone. It may provide an improved strategy for antitumor therapy and the clinical treatment of prostate cancer
