    Crystallization of Adenylylsulfate Reductase from Desulfovibrio gigas: A Strategy Based on Controlled Protein Oligomerization

    Adenylylsulfate reductase (adenosine 5ā€²-phosphosulfate reductase, APS reductase or APSR, E.C.1.8.99.2) catalyzes the conversion of APS to sulfite in dissimilatory sulfate reduction. APSR was isolated and purified directly from massive anaerobically grown Desulfovibrio gigas, a strict anaerobe, for structure and function investigation. Oligomerization of APSR to form dimersā€“Ī±_2Ī²_2, tetramersā€“Ī±_4Ī²_4, hexamersā€“Ī±_6Ī²_6, and larger oligomers was observed during purification of the protein. Dynamic light scattering and ultracentrifugation revealed that the addition of adenosine monophosphate (AMP) or adenosine 5ā€²-phosphosulfate (APS) disrupts the oligomerization, indicating that AMP or APS binding to the APSR dissociates the inactive hexamers into functional dimers. Treatment of APSR with Ī²-mercaptoethanol decreased the enzyme size from a hexamer to a dimer, probably by disrupting the disulfide Cys156ā€”Cys162 toward the C-terminus of the Ī²-subunit. Alignment of the APSR sequences from D. gigas and A. fulgidus revealed the largest differences in this region of the Ī²-subunit, with the D. gigas APSR containing 16 additional amino acids with the Cys156ā€”Cys162 disulfide. Studies in a pH gradient showed that the diameter of the APSR decreased progressively with acidic pH. To crystallize the APSR for structure determination, we optimized conditions to generate a homogeneous and stable form of APSR by combining dynamic light scattering, ultracentrifugation, and electron paramagnetic resonance methods to analyze the various oligomeric states of the enzyme in varied environments

    Thrombolysis in Myocardial Infarction Frame Count in Single-Vessel Disease After Angioplasty

    SUMMARYBackgroundWe compared the thrombolysis in myocardial infarction (TIMI) frame count and examined the impact of angioplasty on the count between patients with normal coronary angiograms and those with single-vessel disease (SVD).MethodsIn 780 consecutive patients referred for coronary angiography, TIMI frame count was measured for 149 patients who had SVD and 32 patients with normal angiograms who underwent the procedure for electro-physiologic study or valvular heart disease survey.ResultsComparison of each of the three vessels in the normal vessel group with the corresponding non-stenotic vessels in the SVD group showed similar counts in each of the left anterior descending artery (LAD), left circumflex artery (LCX), and right coronary artery (RCA). For the stenotic vessels, after successful angioplasty, the counts were all reduced (LAD, 54.5 Ā±28.8 vs. 34.0 Ā±19.3; LCX, 67.3 Ā±31.1 vs. 34.1 Ā±19.0; RCA, 33.2 Ā±28.1 vs. 19.3 Ā±7.9; all p <0.05). In addition, the count in the RCA after angioplasty was lower, compared with the RCA of the normal group (19.3 Ā±7.9 vs. 29.1 Ā±14.6, p = 0.001). Multivariate analysis showed that the use of oral calcium channel blockers was the only independent predictor for the reduction in RCA after angioplasty.ConclusionIn patients with SVD, the data of TIMI frame count in the nonstenotic vessels were similar to those without the disease, suggesting that the count in the normal artery is not affected by the adjacent stenotic artery. For the stenotic vessels, angioplasty had differential effects on each of the three arteries, indicating the existence of distinct properties, which is affected by calcium channel blockers, for individual coronary arteries in response to atherosclerosis and/or angioplasty

    A mobile computing device (e.g., a mobile phone, camera, tablet computer, wearable and/or headset device) may include an integrated display device (e.g., a presence-sensitive screen) at which a user interface is presented to provide perception field functionality, which enables real-time depth estimation for static or moving objects that are detected by the mobile computing device based on sensory input from an onboard camera. In various examples, this functionality may be embodied in a portable and flexible library (e.g. Android library) that is installed on the mobile computing device. The purpose of perception field monitoring is to provide fast and efficient algorithms for spatial object mapping to enable real-time distance estimation of static and moving objects on a mobile computing device. The implementation of these algorithms may provide spatial location information of targeted objects, as well as distance information associated with objects that are detected by the device. In certain cases, mobile applications executing on the device may utilize such information to provide assistance to visually impaired users by creating audible alerts

    Gender Difference in Statin Intervention on Blood Lipid Control among Patients with Coronary Heart Disease

    SummaryBackgroundThe aim of this study was to clarify the current status in the effective control of dyslipidemia in Taiwanese women and men with coronary heart disease (CHD).Materials and methodsA total 1584 patients with CHD (1188 men, aged 64.8Ā Ā±Ā 11.6 years and 396 women, aged 69.0Ā Ā±Ā 9.8 years) from 3486 patients who had atherosclerotic vascular disease and complete lipids measured values [total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C)] were used for analysis.ResultsThe waist, height, weight, and creatinine levels were higher in men than in women. The systolic blood pressure, TC, HDL-C, LDL-C, fasting blood glucose, and platelet were lower in men than in women. Men were more likely to achieve the target goal than women in TCĀ <Ā 160Ā mg/dL, LDL-CĀ <Ā 100Ā mg/dL, and TGĀ <Ā 150Ā mg/dL as well as to achieve HDL-C goal.ConclusionA significant gap was found between the guidelines and clinical practice in statin intervention among these CHD patients, particularly for women.     Improving Success Rates of Percutaneous Coronary Intervention for Chronic Total Occlusion at a Rural Hospital in East Taiwan

    SummaryBackgroundWe aimed to report the results of percutaneous coronary intervention for chronic total occlusion (CTO) in a remote hospital of southeast Taiwan that does not have on-site coronary artery bypass graft support and has insufficient medical resources.MethodsFrom 2006 to 2009, we identified 96 patients who underwent percutaneous coronary intervention and whose coronary angiogram showed CTO lesions. On-site cardiovascular surgeons were unavailable from 2006 to 2009.ResultsThe success rate (test for trend, pĀ =Ā 0.02) and numbers of guidewires used (test for trend, pĀ =Ā 0.59) significantly increased from 2006 to 2009, and the procedural time reduced significantly (test for trend, pĀ =Ā 0.001). The volume of contrast media injected decreased, although this result was not statistically significant (pĀ =Ā 0.70).ConclusionOur experience in managing CTO lesions substantially improved and the procedural time reduced over 4 years, even when constrained by a relative shortage of medical resources

    Areca Users in Combination with Tobacco and Alcohol Use Are Associated with Younger Age of Diagnosed Esophageal Cancer in Taiwanese Men

    BACKGROUND: Whether the habitual use of substances (tobacco, alcohol, or areca nut (seed of the Areca palm)) can affect the age of esophageal squamous cell carcinoma (ESCC) presentation has rarely been examined. METHODS: The study subjects were those who were males and the first time to be diagnosed as ESCC (ICD-9 150) and who visited any of three medical centers in Taiwan between 2000 and 2009. A standardized questionnaire was used to collect substance uses and other variables. RESULTS: Mean age (Ā±SD) at presentation of ESCC was 59.2 (Ā±11.3) years in a total of 668 cases. After adjusting for other covariates, alcohol drinkers were 3.58 years younger to have ESCC than non-drinkers (pā€Š=ā€Š0.002). A similar result was found among areca chewers, who were 6.34 years younger to have ESCC than non-chewers (p<0.0001), but not among cigarette smokers (pā€Š=ā€Š0.10). When compared to the group using 0-1 substances, subjects using both cigarettes and alcohol were nearly 3 years younger to contract ESCC. Furthermore, those who use areca plus another substance were 7-8 years younger. Subjects using all three substances had the greatest age difference, 9.20 years younger (p<0.0001), compared to the comparison group. CONCLUSION: Our findings suggest that habitually consuming tobacco, alcohol, and areca nut can influence the age-onset of ESCC. Since the development of ESCC is insidious and life-threatening, our observation is worthy to be reconfirmed in the large-scale and long-term follow-up prospective cohort studies to recommend the screening strategy of this disease
